Book excerpt: Regulation of gene expression is important to cellular growth and differentiation of eukaryotes, as it plays key roles in determining the eventual expression levels and cellular localizations of a large number of proteins. RNA-binding proteins (RBPs) with conserved RNA-binding domains (RBDs) are key molecules in controlling gene expression. These conserved RBDs can specifically interact with target RNAs through specific sequences and structures to regulate mRNA stability, translation efficiency and subcellular localization. One of the well-studied RBPs is the Puf family RBPs. Members of the Puf family share a highly conserved Puf RBD, which typically consists of eight imperfect tandem repeats of ~36 amino acids (aa). Here I report the studies of two Puf members, PfPuf3 in Plasmodium falciparum and its ortholog PyPuf3 in Plasmodium yoelii. Both genes could not be disrupted, suggesting that they may be essential for the intraerythrocytic developmental cycle (IDC). A time-course study of both PfPuf3 mRNA and protein indicated that PfPuf3 was expressed during the entire IDC, with peak expression in early trophozoites. Cellular fractionation demonstrated that PfPuf3 preferentially partitioned to the nuclear than the cytoplasmic fractions. Interestingly, tagging of the endogenous of PfPuf3 and PyPuf3 with the green fluorescent protein showed that both proteins were limited to the nucleus, which is in contrast to the cytoplasmic localization of PfPuf1 and PfPuf2. Further, we found PfPuf3 co-localized with a well-known nucleolus maker PfNop1, demonstrating that PfPuf3 is a nucleolar protein. Using a parasite line with PfPuf3-tagged with PTP (ProtC-TEV-ProtA), we were able to affinity purify the PfPuf3 protein complex and the associated RNAs. Proteomic analysis of the PfPuf3 complex identified 32 proteins that are associated with the 60S ribosome subunit, suggesting that PfPuf3 might be involved in ribosomal biogenesis. RNA sequencing analysis of the purified PfPuf3 complex revealed significant enrichment of the 28S rRNA and ITS2 (internal transcribed spacer 2), implying that PfPuf3 may recognize and bind to these rRNA sequences. Taken together, these results demonstrated nucleolar localization of PfPuf3 and suggested an essential function of PfPuf3 in ribosomal biogenesis.

In the United States, it is estimated that approximately 30% of cats, the primary carriers, have been infected by T. gondii. Most humans contract toxoplasmosis by eating cyst-contaminated raw or undercooked meat, vegetables, or milk products or when they come into contact with the T. gondii eggs from cat feaces while cleaning a cat's litterbox, gardening, or playing in a sandbox. Approx 1 in 4 (more than 60 million) people in the USA are infected with the parasite, and in the UK between 0.5 and 1% of individuals become infected each year. By the age of 50, 40% of people test positive for the parasite. The predilection of this parasite is for the central nervous system (CNS) causing behavioral and personality alterations as well as fatal necrotizing encephalitis, and is especially dangerous for HIV infected patients. Though there have been tremendous strides in our understanding of the biology of Toxoplasma gondii in the last decade, there has been no systemic review of all of the information that has accumulated. Toxoplasma gondii provides the first comprehensive summary of literature on this organism by leading experts in the field who were responsible for organising the 7th International Congress on Toxoplasmosis in May 2003. It offeres systematic reviews of the biology of this pathogen as well as descriptions of the methods and resources used. This notion implies different evolutionary strategies of host exploitation. Typically, parasites establish long-term infections and induce relatively little mortality, as they often limit pathological changes by modulating host cells and downregulating adverse immune responses. Their pattern of distribution tends to be endemic rather than epidemic. Despite these seemingly benign traits, parasites usually cause substantial chronic morbidity, thus constituting an enormous socioeconomic burden in humans, particularly in resource poor countries, and in livestock worldwide. Parasite-induced fitness costs are an evolutionary force that can shape populations and contribute to species diversity. Therefore, a thorough understanding of parasites and parasitic diseases requires detailed knowledge of the respective biochemical, molecular and immunological aspects as well as of population genetics, epidemiology and ecology. This Research Topic (RT) bridges disciplines to connect molecular, immunological and wildlife aspects of parasitic infections. The RT puts emphases on four groups of parasites: Plasmodium, Toxoplasma, Giardia and intestinal helminths. Co-infections are also covered by the RT as they represent the most common form of parasite infections in wildlife and domestic animal populations. Within the four types of parasites the following topics are addressed: (1) Experimental models: hypothesis testing, translation and limits. (2) Critical appraisal of experimental models.
