Book excerpt: Over the past ten years, metabolomics strategies have allowed the relative or absolute quantitation of metabolite levels for the study of various biological questions in plant sciences. For fruit studies, in particular, they have participated in the identification of the genes underpinning fruit development and ripening. This book proposes examples of the current use of metabolomics studies of fruit for basic research or practical applications. It includes articles about several tropical and temperate fruit species. The studies concern fruit biochemical phenotyping, fruit metabolism during development and after harvest, including primary and specialized metabolisms, or bioactive compounds involved in fruit growth and environmental responses. The analytical strategies used are based mostly on liquid or gas chromatography coupled with mass spectrometry, but also on nuclear magnetic resonance and near-infrared spectroscopy. The effect of genotype, stages of development, or fruit tissue type on metabolomic profiles and corresponding metabolism regulations are addressed for fruit metabolism studies. The interest in combining other omics with metabolomics is also exemplified.

In addition of protecting the seeds, these specialized organs unique to plants were developed to promote seed dispersal via the contribution of frugivorous animals. Fruit development and ripening is a complex process and understanding the underlying genetic and molecular program is a very active field of research. Part of the ripening process is directed to build up quality traits such as color, texture and aroma that make the fruit attractive and palatable. As fruit consumers, humans have developed a time long interaction with fruits which contributed to make the fruit ripening attributes conform our needs and preferences. This issue of Frontiers in Plant Science is intended to cover the most recent advances in our understanding of different aspects of fleshy fruit biology, including the genetic, molecular and metabolic mechanisms associated to each of the fruit quality traits. It is also of prime importance to consider the effects of environmental cues, cultural practices and postharvest methods, and to decipher the mechanism by which they impact fruit quality traits. Most of our knowledge of fleshy fruit development, ripening and quality traits comes from work done in a reduced number of species that are not only of economic importance but can also benefit from a number of genetic and genomic tools available to their specific research communities. For instance, working with tomato and grape offers several advantages since the genome sequences of these two fleshy fruit species have been deciphered and a wide range of biological and genetic resources have been developed. Ripening mutants are available for tomato which constitutes the main model system for fruit functional genomics. In addition, tomato is used as a reference species for climacteric fruit which ripening is controlled by the phytohormone ethylene. Likewise, grape is a reference species for non-climacteric fruit even though no single master switches controlling ripening initiation have been uncovered yet. In the last period, the genome sequence of an increased number of fruit crop species became available which creates a suitable situation for research communities around crops to get organized and information to be shared through public repositories. On the other hand, the availability of genome-wide expression profiling technologies has enabled an easier study of global transcriptional changes in fruit species where the sequenced genome is not yet available. In this issue authors will present recent progress including original data as well as authoritative reviews on our understanding of fleshy fruit biology focusing on tomato and grape as model species.

Book excerpt: 'Honeycrisp' apple fruit marketability is negatively impacted by postharvest physiological disorders, including soft scald and soggy breakdown. These non-pathogenic disorders are thought to be provoked by environmental stresses, including postharvest chilling. Advanced harvest maturity also enhances disorder risk. The goal of this research was to understand metabolism associated with development of soft scald and soggy breakdown of Honeycrisp in order to develop risk management tools for their control. Initially, field weather station data and standard quality/maturity metrics were evaluated to determine their utility for disorder risk assessment. These metrics, including Brix, internal ethylene concentration, starch, firmness and titratable acidity, do not reliably predict fruit susceptibility to soft scald and soggy breakdown. Further tests characterized the metabolome associated with symptomatic fruit tissue, adjacent healthy tissue, and tissue from entirely healthy fruit. Metabolites associated with oxidative stress, membrane disruption, and fermentative metabolism were linked with soggy breakdown symptoms, providing a metabolic fingerprint for this disorder. A separate test sought to determine if internal CO2 injury is distinct from soggy breakdown visually as well as metabolically by evaluating the impact of elevated CO2 partial pressures on internal disorder development. The metabolic profile of high CO2-induced cortex browning was not different from cortex browning provoked by chilling. However, visual symptoms sometimes differed between ostensible high CO2 CA injury compared with soggy breakdown, and succinic acid was elevated in fruit stored in high CO2. Finally, to identify candidate metabolites/transcripts for possible risk management tools, metabolites and gene expression differing prior to soft scald development were determined using metabolomic and transcriptomic profiling. Several classes of metabolites and potentially associated transcripts were differently regulated according to soft scald susceptibility, including hydroxylated triterpenoids, sesquiterpenoids, phytosterols, and their conjugates. Overall, results affirm that pre- and postharvest environmental factors, CA and chemical treatments, and physiological maturity alter fruit metabolic activities related to soft scald/soggy breakdown and CA injury. Fruit at risk for these postharvest disorders may not be detectable with standard quality metrics, and results indicate that monitoring a set of metabolites and transcripts could lend accuracy to soft scald/soggy breakdown risk assessment and enable better storage management decisions.

Yet, prior to tbe symposium, only a few of tbe participants knew one anotber. We decided to expand tbe symposium into a book. The book would, we boped, accomplish for otbers wbat we bad discovered in ourselves. That is, the field of Natural Products is broad, but similar in techniques and approach, ancient but modern, and bas been and continues to be extremely valuable to humankind. We wanted the book to serve as an introductory text for courses and as a reference work for the future. We also determined to include the structure of every chemical in the chapter where it was mentioned so the reader would not have to find the structure somewhere else or to try and deduce the structure from the chemical name. Little did we know what an undertaking these goals would be or the time this would take.
