Unfortunately, first-generation ABCB1 inhibitors had minimal effects in human phase II clinical trials due to low specificity and non-specific toxicity issues. We aimed to identify treatments that can eradicate ABCB1-dependent multidrug resistance. Using multiple Taxanes resistant mCRPC cell lines, we observed that this resistance is regulated by overexpression of ABCB1, thereby recapitulating the clinical response of mCRPC patient with acquired Taxanes resistance. Using cell-based screen with FDA-approved drugs, we found that DNA damaging compounds, Camptothecin (CPT), and Cytarabine (Ara-C), are cytotoxic to Taxanes-resistant cells. Treatment with either compound eradicated the Taxanes-resistant cells at equivalent concentrations as those for the Taxanes-responsive (parental) cells. Collectively, our data suggest a conceptually new treatment modalities for patients diagnosed with terminal mCRPC patients using Mps1i, CPT, or Ara-C to overcome Taxane resistance or ABCB1 dependent MDR.
