Therefore, this book deals with algorithms that are involved in understanding chemical space and selection of diverse sets of structures. The algorithms also deal with the problem of focused diversity where chemical libraries are being created within a structured physical volume. Diversity is necessarily connected to combinational chemistry, although this book is limited to the application of diversity methods to combinational chemistry and does not deal with synthetic methods. It is this focus on algorithms and strategies for exploiting molecular diversity that makes it different from books on combinational chemistry. The intended readership of the book falls into two categories: those actively engaged in applying molecular diversity in the chemical industry and those in academia who are developing strategies to embrace, understand and accept the many problems thrown up by this new research field of molecular diversity.

Book excerpt: Barnase was successfully displayed on the surface of m13 phage as an N-terminal fusion with coat protein III. The successful display was verified by Western Blotting of purified phage. For selection of a thermostable barnase, the buried residues Ala11, Leu14, Leu20, Ala74, Ile88, Tyr90, and Ile96 were replaced randomly with Phe, Leu, Val, Ile, Ala, Pro, Gly, Ser, or Thr, giving a library of 2x10(exp 6) members. The transformation yield obtained (4x10(exp 6) allows sampling of all possible members. We are currently selecting the phage library on immobilized barstar (the femtomolar inhibitor of barnase) at elevated temperature. Barstar N7C, a mutant constructed in this lab, was linked via a disulfide to a chromatography support. This allows specific elution under mild reducing conditions. Barnase has also been successfully expressed on the surface of phage when attached to the C-terminus of an Fab light chain, with the heavy chain attached to the gene III protein. Again, western blotting analysis was used to verify the success of display. Replacement of the barnase gene with cDNA should allow the successful surface display of cDNA libraries on phage.

Do different sets of data show similar or conflicting pictures of genetic diversity? Will the large genome size reduce the role of barley as a model organism in these current sequencing days? Or, are there still major reasons to continue to work with this beautiful crop? The aim of this book is to cover the complex issue of diversification in time and space in a single crop: barley. Leading scientists from various fields describe the entire variation pattern in different sets of characters and an attempt is made for a synthesis to a holistic picture. At a stroke, it seems to be possible to synthesise more molecules in a month than have previously been made in the whole of the distinguished history of organic chemistry, Furthermore, all the molecules can be screened in the same short period. However, like any weapons of immense power, these techniques must be used with care, to achieve maximum impact. The costs of implementing and running high-throughput screening and combinatorial chemistry are high, as large dedicated facilities must be built and staffed. In addition, the sheer number of chemical leads generated may overwhelm the lead optimisation teams in a hail of friendly fire. Mother nature has not entirely surrendered, as the number of building blocks that could be used to build libraries would require more atoms than there are in the universe. In addition, the progress made by the Human Genome Project has uncovered many proteins with different functions but related binding sites, creating issues of selectivity. Advances in the new field of pharmacogenomics will produce more of these challenges. There is a real need to make hi- throughput screening and combinatorial chemistry into 'smart' weapons, so that their power is not dissipated. That is the challenge for modellers, computational chemists, cheminformaticians and IT experts. In this book, we have broken down this grand challenge into key tasks.

Members of the genus Chaetomium are capable of colonizing various substrates and are well-known for their ability to degrade cellulose and to produce a variety of bioactive metabolites. More than 200 compounds have been reported from this genus. A huge number of new and bioactive secondary metabolites associated with unique and diverse structural types, such as chaetoglobosins, epipolythiodioxopiperazines, azaphilones, depsidones, xanthones, anthraquinones, chromones, and steroids, have been isolated and identified. Many of the compounds have been reported to possess significant biological activities, such as antitumor, antimalarial, cytotoxic, enzyme inhibitory, antimicrobial, phytotoxic, antirheumatoid and other activities. Chaetomium taxa are frequently reported to be cellulase and ligninase producers with the ability to degrade cellulosic and woody materials.
