Book excerpt: The history of research on hog cholera (HC)/classical swine fever (CSF) can be roughly divided into three phases which are characterized by the methods available at the time for demonstrati ng the causati ve agent. Phase covered the period before the viral etiology of HC was discovered by de Schweinitz and Dorset (1904)*. Thereafter (Phase II) the detection of HC virus (HCV) was accomplished by laborious, time-consuming and costly pig inoculation experiments. This explains the extensive search for methods not only for detection but also for accurate infectivity titration as well as for applicable serological techniques to solve urgent problems concerning the pathogenesis, diagnosis, epidemiology and prophylaxis of HC. It was not before the late fifties that HC research entered Phase III when fluorescent antibody techniques offered not only the means for detection and titration of HCV in porcine cell cultures but also for more intensive research on hog cholera and its virus. And yet, there are a number of questions to be answered, e. g. on the genetic and antigenic relation of HCV to bovine viral diarrhea (BVD) virus. There are indications that Phase IV of HC research will bear the stamp of biotechnology. In view of this development it appears appropriate to give an up-dating and summarizing account of HC/CSF including comparative aspects of infections caused by structurally related viruses. Book excerpt: This investigation was undertaken to develop a system by which one could induce viral and/or bacterial diarrhea in pigs at will, and study the physiological and morphological changes induced by the pathogen preceeding overt signs of diarrhea. A strain of transmissible gastroenteritis virus (TGE) obtained from Purdue Univ. induces diarrhea in young (5 to 15 day old) pigs within 48 hrs. after oral administration. To study the earliest stages of infection, TGE virus was introduced directly into an intestinal loop in situ, and allowed to remain there from 2 to 24 hrs. before the animals were sacrificed. Histological studies of samples taken from a loop 2 hrs. after infection revealed a dilation of the Golgi apparatus and the endoplasmic reticulum of the apical cytoplasm. Disaccharidase activity in homogenates of intestinal mucosa from control and infected pigs was studied. All enzymes were greatly reduced at 24 to 48 hrs. after oral infection. Lactase activity was reduced to less than 25% of control levels. Jejunum was the region of the digestive tract where reduction in enzyme levels was greatest. Colonic mucosa exhibited very low enzymatic activity in control animals. Results of Lactose Tolerance Tests performed on control and TGE infected pigs suggest that lactose absorption is markedly impaired 48 hrs. after oral infection.

Book excerpt: The Geographical Distribution of Animal Viral Diseases attempts to shed some light on the global distribution of 110 different viral diseases, mainly of livestock and companion animals. The world literature was screened for 110 different viruses, and maps were prepared. These maps delineate the global distribution of pathogenic viruses based on authenticated reports from a variety of reliable sources. Four viruses were categorized as affecting more than one species to a significant degree (astrovirus, rabies, rotaviruses, and Rift Valley fever). The largest number of maps involved viruses that affect humans. Of the 28 viruses a large number were from the California encephalitis group. Ten of the 28 viruses were reported only in the Eastern Hemisphere, 14 only in the Western Hemisphere, and four were worldwide. Birds were the next most frequently affected group with the 15 viruses, followed by pigs with 14 viruses. Overall the vector-borne viruses appear to have much sharper and clear-cut geographical boundaries than the others.
