Different strains of MG have been used as vaccines in multiple-age commercial layer farms in an effort to protect the birds against more virulent field strains. The lower level of protection afforded by the low virulent MG strain vaccines provides an opportunity to the use of an overlay (revaccination) with an F-strain M. gallisepticum (FMG) later in their production cycles. In the present study, three trials were conducted to investigate the effects of prelay vaccinations of ts-11 strain MG, MG-Bacterin, or their combination, in conjunction with a FMG overlay after peak production in commercial layers. The following treatments were utilized at 10 wk of age (woa): 1) Control (no vaccinations); 2) ts-11 MG vaccine; 3) MG-Bacterin vaccine; and 4) ts-11 MG and MG-Bacterin combination. At 45 woa, all the birds in trial 1 and half of the birds in each treatment group in trials 2 and 3 were overlaid with an FMG vaccine. Various parameters including performance, internal egg and eggshell quality, blood, and visceral characteristics of the birds were evaluated.

Book excerpt: Chickens infected with Mycoplasma gallisepticum, the primary etiologic agent of chronic respiratory disease (CRD), present with significant respiratory complications. The host response is often immunopathogenic and contributes to increased tracheal thickness due to the chemotaxis of leukocytes, cellular damage at the mucosal surface, and the inability to clear the infection. The response to M. gallisepticum is complex and the differences that lead to a maladaptive versus beneficial response are poorly understood. To elucidate the extent of the host response to virulent and attenuated strains, we conducted next-generation sequencing on samples taken directly from the host’s tracheal mucosa after infection with either virulent Rlow, or laboratory attenuated vaccine strains Mg 7 or GT5. In the first study, transcriptional analysis of the host response to virulent Rlow over a 7-day infection was conducted on samples from days 1, 3, 5 and 7. Data indicated a rapid, pro-inflammatory response, identifying numerous genes involved in immune pathways that peaked 3 days post-infection, and a significant metabolic response observed on days 1 and 3. Further analysis revealed increased expression of host cell receptors that likely heightens their sensitivity to damage- and pathogen- associated molecular patterns and results in the production of cytokines and chemokines that recruit and activate immune cells. In the second study, comparative transcriptomics identified differences between the early host response to Rlow and laboratory vaccine strains, Mg 7 and GT5. Infection with these attenuated strains resulted in a dampened host response involving significantly fewer immune and metabolism-related genes, limited differential expression of host cell receptors, and decreased production of cytokines and chemokines. Future vaccines should aim to target immune genes active in Mg 7 and GT5 infection while avoiding those uniquely differential in Rlow infection to induce a protective, and durable adaptive response while avoiding immune-related exacerbation of disease.

Book excerpt: Mycoplasma gallisepticum (MG) and M. synoviae (MS), the most pathogenic mycoplasmal pathogens of poultry, are both horizontally and vertically transmitted. Targeted sequencing of the 16S-23S rRNA intergenic spacer region (IGSR), mgc2, MGA_0319 and gapA genomic regions was used to develop a database of South African MG sequences, and to elucidate the molecular epidemiology of MG in South Africa. The sequences of the twelve unique MG genotypes identified were highly similar to the available live MG vaccines, necessitating a combination of IGSR and mgc2 sequencing for differentiation. Targeted genetic sequencing was instrumental in identifying previously reported cases of apparent reversion to virulence and vertical transmission of the live ts-11 vaccine in the state of Georgia. The egg transmission and pathogenicity of ts-11 vaccine and ts-11-like isolates from ts-11 vaccinated breeders (K6222B) and their broiler progeny (K6216D) were evaluated in this research. K6216D transmitted via the egg at an average rate of 4.0% in the third and fourth weeks post infection, while egg transmission of K6222B and ts-11 vaccine was not detected. K6216D and K6222B were significantly more virulent and invasive than ts-11 vaccine. These results provide the first conclusive evidence of transovarian transmission of an isolate of the ts-11 genotype. A severe outbreak of MS in the state of Arkansas prompted an investigation of the horizontal transmission of two MS isolates genotyped as S-10 and S-17 based on targeted genetic sequencing. MS transmission from infected seeder chickens was detected by real-time PCR at 6 weeks post infection in both the S-10 and S-17 groups; however, the transmission patterns differed, with S-10 transmitting to adjacent contact chickens and S-17 to fomite contact chickens, but not to fomite contacts medicated with tylosin. A novel infection model incorporating mucin was subsequently developed to evaluate the fomite transmission of MS. Low-level MS transmission from S-17-inoculated cotton fabric, and from S-10-inoculated plastic, cotton fabric and feather fomites to sexually mature broiler breeder chickens was detected by real-time PCR, but not by culture or serology, 3 weeks after fomite placement. These studies represent the first experimental evaluations of the horizontal transmission of MS in sexually mature chickens.

In addition to the PPLO specialists, the conference invited Julius Marmur to compare PPLO DNA to DNA of other organisms; David Garfinkel, who was one of the first to develop computer models of metabolism; Cyrus Levinthal to talk about coding; and Henry Quastler to discuss information theory constraints on very small cells. The conference was an announcement of the role of PPLO in the fundamental understanding of molecular biology. Looking back 40-some years to the Connecticut meeting, it was a rather bold enterprise. The meeting was international and inter-disciplinary and began a series of important collaborations with influences resonating down to the present. If I may be allowed a personal remark, it was where I first met Shmuel Razin, who has been a leading figure in the emerging mycoplasma research and a good friend. This present volume is in some ways the fulfillment of the promise of that early meeting. It is an example of the collaborative work of scientists in building an understanding of fundamental aspects of biology.
