Book excerpt: Integration of living cells with novel microdevices requires the development of innovative technologies for manipulating cells. Chemical surface patterning has been proven as an effective method to control the attachment and growth of diverse cell populations. Patterning polyelectrolyte multilayers through the combination of layer-by-layer self-assembly technique and photolithography offers a simple, versatile and silicon compatible approach that overcomes chemical surface patterning limitations, such as short-term stability and low protein adsorption resistance. In this study, direct photolithographic patterning of poly(acrlic acid)(PAA)/poly(acryl amide (PAAm) and PAA/poly(ally amine hydrochloride) (PAH) polyelectrolyte multilayers was developed to pattern mammalian neuronal, skeletal and cardiac muscle cells. For all studied cell types, PAA/PAAm multilayers behaved as a negative surface, completely preventing cell attachment. In contrast, PAA/PAH multilayers have shown a cell-selective behavior, promoting the attachment and growth of neuronal cells (embryonic rat hippocampal and NG108-15 cells) to a greater extent, while providing a little attachment for neonatal rat cardiac and skeletal muscle cells (C2C12 cell line). PAA/PAAm multilayer cellular patterns have also shown a remarkable protein adsorption resistance. Protein adsorption protocols commonly used for surface treatment in cell culture did not compromise the cell attachment inhibiting feature of the PAA/PAAm multilayer patterns. The combination of polyelectrolyte multilayer patterns with different adsorbed proteins could expand the applicability of this technology to cell types that require specific proteins either on the surface or in the medium for attachment or differentiation, and could not be patterned using the traditional methods.

In the last 15 years the LbL technique has seen considerable developments and has now reached a point where it is beginning to find applications in bioengineering and biomedical engineering. The book gives a thorough overview of applications of the LbL technique in the context of bioengineering and biomedical engineering where the last years have witnessed tremendous progress. The first part familiarizes the reader with the specifics of cell-film interactions that need to be taken into account for successful application of the LbL method in biological environments. The second part focuses on LbL-derived small drug delivery systems and antibacterial agents, and the third part covers nano- and microcapsules as drug carriers and biosensors. The fourth and last part focuses on larger-scale biomedical applications of the LbL method such as engineered tissues and implant coatings.

Book excerpt: In recent decades, dynamic Layer-by-Layer (LbL) assembly of multilayer thin films has been widely recognized for many reasons, especially for its versatile functionality and ease of fabrication. Films can be fabricated towards huge amounts of materials, such as polyelectrolytes, organic components, polymeric microgels, etc. Through the long history of LbL assembly research, few papers proposed ideas of making porous films or foamed films directly or indirectly, as foamed material contained plenty of pores, which could be used in insulation materials and fast food-packages. In several papers, the step of acid treatment aimed for irreversible structure transformation applied into films fabricated by the LbL assembly method. This indirect method makes procedures more complex, so direct fabrication with LbL is necessary. However, time-consuming problems and challenges from external environmental factors still limits the development of this technique. In this thesis, a facile fabrication of porous films through the LbL assembly method was introduced. Strong polyelectrolyte, polycation poly(diallyldimethylammonium-chloride) (PolyDADMAC) and polyanion poly(styrene sulfonate acid) (PSSA) are utilized as the original material. And PolyDADMAC was slightly modified by sodium hydroxide (NaOH) and carbon dioxide (CO2) in order to get the bicarbonate structure at the side groups along the chain of the polyelectrolyte. In order to deeply study the films, some other particles, such as the surfactant brij-76 and RDS laponite were added into certain solutions for comparison. Films were achieved by alternately putting clean glass slides into the polycation and polyanion solutions with the rinsing step in between. 20-bilayer film can be achieved after repeating the procedures above 20 times. The principle is that the bicarbonate functional group will react with the hydrogen ions for the sake of letting the gas carbon dioxide out and then make the porous structure throughout the film. The morphology of the films measured by an atomic force microscope (AFM) and a scanning electron microscope (SEM) shows that porous structures are obvious through the whole film when no sodium bicarbonate (NaHCO3) is added into the anion solution. The results help to solve the problem of directly making the foaming films as well as simplifying the procedures.

Furthermore, the basic idea of making a synthetic material that can imitate the func tions of natural materials probably could be traced back into antiquity. From the dawn of creation, people have probably attempted to duplicate or modify the activities of the natural world. (One can even find allusions to these attempts in the Bible; e. g. , Genesis 30. ) The term "mimetic" means to imitate or mimic. The word "mimic" means to copy closely, or to imitate accurately. Biomimetic, which has not yet entered most dictionaries, means to imitate or mimic some specific bio logical function. Usually, the objective of biomimetics is to form some useful material without the need of utilizing living systems. In a simi lar manner, the term biomimetic polymers means creating synthetic poly mers which imitate the activity of natural bioactive polymers. This is a major advance in polymer chemistry because the natural bioactive polymers are the basis of life itself. Thus, biomimetic polymers imitate the life process in many ways. This present volume delineates some of the recent progress being made in this vast field of biomimetic polymers. Chemists have been making biomimetic polymers for more than fifty years, although this term wasn't used in the early investigations.
