Book excerpt: La spectromicroscopie infrarouge est une technique d'analyse extrêmement performante capable à la fois de caractériser des composés chimiques par leurs bandes d'absorption et de les localiser spatialement grâce à un microscope. L'expérience AFMIR (Atomic Force Microscope InfraRed) est une méthode récente et originale de spectromicroscopie infrarouge sub-Iongueur d'onde qui couple un laser infrarouge pulsé accordable et un microscope à force atomique (AFM). En utilisant l'effet photothermique, elle permet de faire de la spectroscopie infrarouge « ultra locale» et de réaliser des cartographies chimiques avec une très bonne résolution latérale (quelques dizaines de nanomètres). Au cours de cette thèse, un travail de modélisation a été mené afin d'estimer la résolution spatiale de la technique AFMIR et de comprendre tous les phénomènes physiques qu'elle met enjeu. Parallèlement à cette étude théorique, une étape de développement instrumental a été nécessaire pour adapter l'expérience à des études en milieu liquide et permettre l'analyse d'échantillons biologiques vivants. Enfin, trois problématiques ont été abordées : infection de la bactérie Escherichia coli par le phage T5, production du polymère PHB par la bactérie Rhodobacter capsulatus et détermination de l'organite cible d'une hormone pour une lignée cellulaire hormono-indépendante du cancer du sein. L'ensemble des résultats a été confirmé avec des mesures de microscopie électronique en transmission et de spectroscopie d'absorption infrarouge classique. La technique AFMIR est maintenant opérationnelle pour être proposée à d'autres chercheurs du domaine de la biologie.

This unique volume provides a comprehensive review and introduction of defects and structural properties of GaN and related compounds for newcomers to the field and stimulus to further advances for experienced researchers. Given the current level of interest and research activity directed towards nitride materials and devices, the publication of the volume is particularly timely. Early pioneering work by Pankove and co-workers in the 1970s yielded a metal-insulator-semiconductor GaN light-emitting diode (LED), but the difficulty of producing p-type GaN precluded much further effort. The current level of activity in nitride semiconductors was inspired largely by the results of Akasaki and co-workers and of Nakamura and co-workers in the late 1980s and early 1990s in the development of p-type doping in GaN and the demonstration of nitride-based LEDs at visible wavelengths. These advances were followed by the successful fabrication and commercialization of nitride blue laser diodes by Nakamura et al at Nichia. The chapters contained in this volume constitutes a mere sampling of the broad range of research on nitride semiconductor materials and defect issues currently being pursued in academic, government, and industrial laboratories worldwide.
