Nous avons également montré à travers une étude thermodynamique et cinétique appuyée par de nombreux exemples, que cette technique permettait de diminuer d'un facteur 5 à 10 les temps d'analyse, d'améliorer la forme des pics et de réduire la teneur en modificateur organique dans la phase mobile. Le couplage avec le détecteur à ionisation de flamme (FID) après optimisation des paramètres de fonctionnement, a été réalisé avec des phases mobiles aqueuses. L'intérêt d'un tel couplage a été démontré par l'analyse de nombreuses familles de solutés non chromophores. La HTLC a également été couplée à la spectrométrie de masse et des applications variées ont permis de mettre en évidence l'avantage d'un tel couplage

Book excerpt: Ces travaux de thèse ont porté sur la caractérisation de mélanges complexes oxygénés avec l'étude d'échantillons issus de la biomasse lignocellulosique, et de mélanges complexes hydrocarbonés via l'exemple des coupes lourdes pétrolières. La complexité chimique de ces deux types d'échantillon est propre à chacun et provient de multiples facteurs : i) les produits issus de la transformation de la biomasse lignocellulosique sont constitués de composés oxygénés étendus sur une large gamme de polarité, de masse moléculaire mais aussi de concentration, ii) les coupes lourdes pétrolières possèdent des milliers de constituants hydrocarbonés ainsi que des composés soufrés et azotés. Dans les deux cas, l'objectif de ces travaux de recherche est d'améliorer la connaissance de la composition des échantillons, ce qui est indispensable pour développer des procédés de transformation encore plus performants. Pour atteindre cet objectif, des méthodes séparatives adaptées aux propriétés des échantillons ont été mises en place et couplées à la spectrométrie de masse haute résolution (HRMS). Une méthodologie via l'utilisation de plans d'expériences a été proposée afin d'optimiser l'interface entre la dimension de séparation et la HRMS. Dans le cas des échantillons issus de la biomasse, dans un premier temps, le couplage de la chromatographie en phase liquide à polarité de phase inversée (RPLC) et de la HRMS avec une source electrospray (ESI) et une source d'ionisation chimique à pression atmosphérique (APCI) a permis une première description des échantillons. Des expériences de fragmentations successives ont mis en évidence des structures inédites. Par la suite, une méthode de chromatographie bidimensionnelle en ligne (LCxLC) dédiée à l'analyse des composés les plus polaires a été optimisée et couplée à la HRMS. La mobilité ionique (IMS) a également été étudiée afin d'évaluer son intérêt au sein des couplages réalisés. Dans le cas des coupes lourdes pétrolières, une méthode associant la chromatographie de partage centrifuge (CPC) en première dimension, la chromatographie en phase supercritique (SFC) en deuxième dimension couplée à la HRMS avec une source de photo-ionisation à pression atmosphérique (APPI) en tant que troisième dimension s'est révélée être une approche pertinente pour quantifier les hydrocarbures aromatiques polycycliques présents dans les distillats sous vide. Ce couplage hors-ligne de la CPC à la SFC a permis de construire des cartographies 2D organisées selon le degré d'insaturation et le nombre de cycles aromatiques des composés détectés, reflétant la composition détaillée des échantillons investigués. Pour terminer, les avantages et inconvénients de l'ensemble des méthodes développées au cours de cette thèse ont été discutés. Les spécificités des deux spectromètres de masse utilisés (FT-ICR et timsTOF) sont comparées en termes de résolution, vitesse d'acquisition et mécanismes de fragmentation. Une capacité de pics a été évaluée pour chaque couplage, ce qui a permis de les positionner les uns par rapport aux autres en perspective de la complexité des échantillons, du traitement des données et du besoin analytique.

L’utilisation d’un support de séparation en carbone graphite poreux et l’optimisation du couplage a rendu possible l’analyse simultanée des explosifs organiques les plus couramment rencontrés. Les effets de matrice sur les performances de la méthode ont ensuite été évalués puis éliminés en partie avec la mise en place d’une procédure de purification de l’échantillon par extraction sur phase solide. Enfin, un protocole général de traitement de l’échantillon incluant les étapes d’extraction, de purification et de concentration a été proposé puis validé avec l’analyse d’échantillons réels obtenus lors de simulations d’attentats avec engins explosifs improvisés
