Book excerpt: The incidence of neurodegenerative diseases including Parkinson's, Alzheimer's, and Huntington's disease is ever-increasing. Therefore, it is essential that we understand the mechanisms behind the pathology of these disorders. Extensive study has been conducted on the factors that play a role in these disorders. Through these studies, several pathways have been elucidated that further enlighten our understanding of these pathologies. However, many studies have been conducted on model systems that do not fully recapitulate all of the pathological hallmarks of these diseases. Additionally, a greater understanding into the interplay between multiple pathways has been lacking. Previously, we established a Drosophila model for Parkinson's disease that demonstrates that both genetic and environmental factors play a role in the susceptibility and resistance of the organism to neurodegenerative disease. The herbicide paraquat leads to degeneration of dopamine neurons and reduced mobility as is seen in Parkinson's patients. Using this model, we determined that an integration of the inflammatory and hypoxia pathways ensues whereby ingestion of paraquat induces the production of nitric oxide synthase (NOS) that is associated with hyper-branching of the trachea; this also occurs in adult Drosophila brains. Additionally, we have investigated the role of von Hippel Lindau (VHL), an E3 ubiquitin ligase that regulates the hypoxia response, in sensitivity to paraquat, induction of the inflammatory pathway, and neurodegeneration. We have found that loss of VHL along with exposure to paraquat leads to exacerbated survival truncation and movement abnormalities. Furthermore, Catsup protein levels (a negative regulator of dopamine biosynthesis) are altered in these mutants demonstrating a putative role for VHL in Catsup regulation. Here, we also show that a metabolite from the common soil bacteria Streptomyces venezuelae leads to decreased survival and loss of dopamine neurons and induction of NOS. Additionally, Îł-synuclein, a gene implicated in familial Parkinson's disease was studied to determine susceptibility to this bacterial metabolite. We found that expression of Îł-synuclein in flies leads to increased sensitivity and loss of dopamine neurons due to the toxic bacterial metabolite. Taken together, this work provides a mechanism behind the pathology of neurodegenerative diseases and signifies that both genetic and environmental factors play a role in the development and progression of these diseases. It also demonstrates that a dynamic interplay of pathways exists and provides avenues by which future therapeutics may be developed.

Book excerpt: Humans are potentially exposed to more than 80,000 toxic chemicals in the environment, yet their impacts on brain health and disease are not well understood. The sheer number of these chemicals has overwhelmed the ability to determine their individual toxicity, much less potential interactive effects. Early life exposures to chemicals can have permanent consequences for neurodevelopment and for neurodegeneration in later life. Toxic effects resulting from chemical exposure can interact with other risk factors such as prenatal stress, and persistence of some chemicals in the brain over time may result in cumulative toxicity. Because neurodevelopmental and neurodegenerative disorders - such as attention-deficit hyperactivity disorder and Parkinson's disease - cannot be fully explained by genetic risk factors alone, understanding the role of individual environmental chemical exposures is critical. On June 25, 2020, the National Academies of Sciences, Engineering, and Medicine's Forum on Neuroscience and Nervous System Disorders hosted a workshop to lay the foundation for future advances in environmental neuroscience. The workshop was designed to explore new opportunities to bridge the gap between what is known about the genetic contribution to brain disorders and what is known, and not known, about the contribution of environmental influences, as well as to discuss what is known about how genetic and environmental factors interact. This publication summarizes the presentation and discussion of the workshop.

They are characterized by the degeneration of specific cell populations that include several pathologies which contribute significantly to morbidity and mortality in the elderly population. Therefore, in recent years, the study of neuroscience has gained significant importance. Most of these neurodegenerative disorders are the result of a complex interaction between genetic and environmental factors that generate progression and can even determine its severity. The presence of mutations in genes as LRRK2, SNCA, PARK7, PARK2 or PINK1 is associated with Parkinson's disease. Mutations in genes such as APP, PS1 and PS2 are associated with familial Alzheimer's disease; while HTT gene mutations are the cause of Huntington's disease. In most cases, this condition is inherited in an autosomal dominant pattern, which means one copy of the altered gene in each cell is sufficient to cause the disorder. It is known that these mutations can also alter the proteins function; however, it has not yet been possible to fully understand how some genetic changes cause the disease or influence the risk of developing these disorders. Most symptoms seen in these conditions occurs when specific nerve cells are damaged or die generating a loss in brain communication. Also many of these mutations generate aggregation of intracellular or extracellular proteins affecting cell function and eventually causing neuronal death. It is unclear whether the presence of these aggregates play an important role in nerve cell death during the development of neurodegenerative diseases, or if they are simply part of the response of cells to the disease. Other mutations affect the mitochondrial function generating alterations in energy production and promoting the formation of unstable molecules such as free radicals. Under normal conditions, the harmful effects caused by free radicals, are offset within the cell. However, in pathological conditions, the presence of mutations can alter this process by allowing the accumulation of radicals and damaging or killing cells. On the other hand, we also know that these diseases may not have a direct genetic component, thus, the study of sporadic type neurodegenerative diseases is much more complex. Histopathological lesions as well as the cellular and molecular alterations are generally indistinguishable from familial cases. For this reason, it is important to understand the genetic and molecular mechanisms associated with this type of pathologies. In this sense, this issue aims to understand the molecular processes that occur in the brain, and how these are influenced by the environment, genetics and behavior.
