Book Electron Microscopic Immunocytochemistry

Download or read book Electron Microscopic Immunocytochemistry written by Julia M. Polak and published by . This book was released on 1992 with total page 292 pages. Available in PDF, EPUB and Kindle. Book excerpt: Electron microscopic immunocytochemistry is a powerful technique combining the resolution of the electron microscope with the specificity and sensitivity of immunocytochemistry. This book discusses methods designed to make this intricate procedure more accessible, and covers recent developments that have simplified the technique and broadened the range of applications. It presents the principles of EM immunocytochemistry, detailed protocols, and examples of ways in which the technique can be applied to a variety of fields, including biomedical research and diagnostic pathology. The book begins with an overview of EM immunocytochemical procedures and of applications in histopathology. The three major chapters cover post-embedding, pre-embedding, and ultra-thin cryosection techniques. The final section illustrates applications of EM immunocytochemistry to the study of endocrine tumors, renal disease, neuropathology, dermatology, and microbiology. The emphasis throughout this detailed volume is on providing practical guidance to scientists and clinicians who may be using the technique for the first time.

Book Fine Structure Immunocytochemistry

Download or read book Fine Structure Immunocytochemistry written by Gareth Griffiths and published by Springer Science & Business Media. This book was released on 2012-12-06 with total page 477 pages. Available in PDF, EPUB and Kindle. Book excerpt: Electron microscopy in the biological sciences can be divided into two disciplines. The first, concerned with high resolution detail of particles or periodic structures, is mostly based on sound theoretical principles of physics. The second, by far the larger discipline, is interested in the information obtainable from thin sections. The theoretical back ground to those groups of techniques for preparing and looking at thin sections is often inexact and "loose", for want of a better word. What should be chemistry is often closer to alchemy. This kind of electron microscopy is often enshrined with mystical recipes, handed down from generation to generation. Admittedly, many of the processes involved, such as those required to embed tissue in epoxy resins, involve multiple interconnected steps, which make it difficult to follow the details of anyone of these steps. If all these steps are shrouded in some mystery, however, can one really trust the final image that emerges on the EM screen? When we present the data in some semi quantitative form is there really no better way to do it than to categorize the parameters with ++, +/-, etc? What happens when one labels the sections with antibodies? Does the whole business necess arily need to be more of an "art" than a "science"? Upon reflecting on these problems in 1981, I had the impression that many of the multi-authored textbooks that existed then (and that have appeared since) tended to exacerbate or at least perpetuate this

Book Autoradiography and Immunocytochemistry

Download or read book Autoradiography and Immunocytochemistry written by M.A. Williams and published by Elsevier. This book was released on 2013-10-22 with total page 238 pages. Available in PDF, EPUB and Kindle. Book excerpt: Autoradiography and Immunocytochemistry sets out in detail the preparation of autoradiographs and methods of preparing and use of immunological staining reagents. This book begins with an introduction to the demand for autoradiography and immunocytochemistry and their most advantageous use, followed by a discussion on the immunocytochemistry at the electron microscopical (EM) level in relation to the preparation of labeled antibodies. Other topics include the general remarks on “staining with antibodies; applying Fer-Ab conjugates to ultrathin sections; and controls in immunoferritin staining. The multistep antibody staining methods with unmodified proteins; control observations in staining experiments using lectins; and pattern analysis in EM immunocytochemistry are also covered. This text concludes with a description of the preparation of electron microscope autoradiographs, including the radioisotopes in EM autoradiography, preparation of radioactively-labeled tissues and cells, and methods of applying nuclear emulsions. This publication serves as a detailed laboratory guide for researchers and workers to successfully conduct electron microscope techniques.

Book Immunohistochemistry and Immunocytochemistry

Download or read book Immunohistochemistry and Immunocytochemistry written by Simon Renshaw and published by John Wiley & Sons. This book was released on 2017-02-15 with total page 251 pages. Available in PDF, EPUB and Kindle. Book excerpt: Immunohistochemistry and immunocytochemistry are invaluable tools for the visualization of tissue and cellular antigens in diagnostic and biological research environments. The need to obtain accurate, reliable and reproducible results is paramount. It is with this fundamental aim in mind that we have compiled Immunohistochemistry: Essential Methods. We have achieved this by examining each aspect of immunochemistry in turn, with each chapter including detailed information regarding the subject matter in question. Each chapter is written by an expert in their field and includes protocols that are typically used in their own research. Subjects covered are, amongst others, antibodies and their production; selection of reporter labels; immunochemical staining methods and experimental design (both using single and multiple reporter labels); quality assurance; automated immunochemistry; confocal microscopy and electron microscopy. In addition, benefits and limitations of each approach are discussed within the chapters.

Book Microinjection and Transgenesis

Download or read book Microinjection and Transgenesis written by Angel Cid-Arregui and published by Springer Science & Business Media. This book was released on 2012-12-06 with total page 619 pages. Available in PDF, EPUB and Kindle. Book excerpt: The establishment of microinjection protocols about 20 years ago for cultured cells and shortly thereafter for the generation of transgenic mice by microinjection of DNA into fertilized mouse eggs greatly influ enced many fields of biology. Not only have the data generated using these approaches contributed to a large extent to our present under standing of gene regulation and cellular function of higher eukaryotic cells, but current knowledge and future developments in this area will certainly have a great impact on basic and applied research for many years to come. This laboratory manual describes the current state of the art in this research area and focuses primarily on both the experimental strategies with an extensive bibliography and the detailed procedures. A large number of studies are presently being performed and a great variety of new experimental designs are rapidly being developed. The book con tains protocols on injection of somatic cells as well as on injection of embryos, the use of similar equipment being a common feature. In the articles dedicated to somatic cells, full descriptions of the manual and automatic injection systems are given as well as the methods for the analysis of injected cells by video-microscopy, electron microscopy or in situ hybridizations. In addition, comprehensive protocols are given for injection experiments with very different purposes, such as to study sig nal transduction or microtubule dynamics.
