Book excerpt: The focus of this thesis is the scanning electrochemical microscopy (SECM) investigation of the electron transport in lipid monolayers and in films of gold nanoparticles. The quality and stability of lipid monolayers were characterized by various analytical techniques such as fluorescence microscopy and spectroscopy, contact angle, and electrochemistry using a monolayer modified electrode. In the SECM measurement, the monolayer electron transport was not detected. The microscopic monolayer quality turned out to be not suitable for studying the electron transport. The conductivity of monolayer protected gold clusters (MPCs) with different core sizes (1-3 nm diameter) were investigated using SECM. These AuMPCs were characterized by UV-VIS, voltammetry, and transmission electron microscopy (TEM). In SECM measurements, the film conductivity has been extracted from the feedback currents. The conductivity appears to be dependent on the MPC core size, but the dependence becomes weaker with core size dispersity. The disordering effect on the film conductivity was investigated along with the core size effect.

Book excerpt: "Electrochemical scanning probe microscopy has exploded in popularity since the late 1980's and widespread adoption for routine materials analysis is swiftly become a reality. In practice, for any technique, widening the material scope, particularly for industrially-relevant materials that bring much greater levels of heterogeneity and complexity in structure, is accompanied by new instrument and theory development.This thesis demonstrates that electrochemical microscopy is capable of quantitative analysis of substrate permeability - a very common but challenging to acquire property in industrially-relevant composite electrodes and corroding materials. Measurements are performed on permeable materials in situ and can thus account for material swelling and active growth of surface films as well as identify electroactive nanoscale defects. Specifically we explore each of the major electrochemical microscopy methods, scanning electrochemical microscopy (SECM), scanning ion conductance microscopy (SICM) and scanning electrochemical cell microscopy (SECCM) and through a combination of experiment and simulation, we demonstrate that mass transport within permeable surface layers can be predicted and therefore isolated from the electrochemical reactivity of the substrate.With SECM, we derived and validated analytical equations that allow rapid determination of film porosity through non-linear curve fitting of routine approach curve measurements. Furthermore, we validated the possible implementations of the equations based on the morphology of the surface film and identified any limitations for routine use. For SICM, we demonstrated that the critical parameter, ionic conductivity within a permeable layer has a significant impact on routine imaging but can also be quantified with approach curves. We present for the first time, direct measurement of ionic conductivity within a permeable film with SICM using a lithium-ion battery cathode film. Finally, with SECCM we took advantage of its well-defined spatial resolution to resolve and assess the size of nanoscale defects in a polymeric thin film. We also introduced the density plot as a new analysis method for SECCM that consolidated the large data sets obtained by experiment and facilitates correlation with simulation"--

Book excerpt: "This thesis presents an electrochemical approach for the non-invasive study of biological systems, and demonstrates the usefulness of scanning electrochemical microscopy (SECM) for quantification of chemical flux in live cells. We first explore the presence of D-serine within the brain. This neurotransmitter plays an important role in proper brain development, and has also been linked to several pathological diseases such as schizophrenia and Alzheimer's disease. Consequently, quantification of local changes in D-serine concentration is essential to further understanding of its role in these processes. A biosensor was thus developed for the detection of D-serine with high sensitivity. Using this microscale biosensor, the release of endogenous D-serine was measured in vivo inside the brain of Xenopus laevis tadpoles, revealing an overall concentration of 2.5 μM. However, the primary site and mechanism of release of D-serine remained unclear, requiring a more localized measurement. After optimizing for improved response time and selectivity, the biosensor was used in a SECM configuration to measure localized release of D-serine from a model system. This SECM methodology provided quantitative imaging of D-serine with high spatial resolution, and could be useful to investigate chemical flux in other biological samples. We then shifted our focus to quantitative imaging of another complex live cell process, specifically multidrug resistance. This defensive mechanism allows cancer cells to pump out chemotherapeutic drugs before they can kill the cell, severely impeding treatment success. Most investigations of transmembrane efflux pumps, including multidrug resistance-associated protein 1 (MRP1), focus on expression to determine their effect in specific cancer types. Researchers have been unable to establish a clear correlation between expression and patient survival. However, the relationship between transporter expression and functional activity is often overlooked. Using SECM with complementary redox mediators and novel cell patterning substrates, we measured MRP1 functional activity in single cancer cells. By challenging parental cell lines with doxorubicin, a drug used in many chemotherapy regimens, we produced six cell populations with increasing MRP1 expression, and measured their concurrent functional activity. This comparative study revealed that in fact MRP1 expression and functional activity do not correlate, and suggest that the measurement of functional activity should be prioritized to identify the effect of MRP1 on chemotherapy treatment success." --
