Book excerpt: The mammary glands (MG) of pigs represent an excellent model for the human breast given their many shared histomorphological features. However, the MG of pigs have not yet been studied for histomorphological changes across the estrous cycle or in response to excess dietary energy. The MG of humans and other species such as the mouse, rat, and cow undergo phases of histomorphological development and regression during the estrous cycle as a result of fluctuating levels of hormones including estrogen, progesterone, and prolactin. In one experiment, we sampled the MG of five nulliparous pigs at several points throughout the estrous cycle to determine histomorphological development and the rates of epithelial division. Epithelial proliferation in the porcine MG peaked around estrus, leading to a more advanced morphological development during diestrus when more branched terminal ductal lobular units were present. In another two experiments we evaluated the effects of varying dietary energy and lysine levels on MG development in pigs as a model for the effect of obesity on human breast development. In the first experiment, Landrace x Large White gilts were assigned to six dietary treatment groups containing three different levels of ME (85%, 100% or 115% of their daily requirement) and two levels of lysine (85% or 100% of their daily requirement). For the second experiment, gilts from six littermate pairs were pair-fed either a standard commercial diet or a diet containing 120% ME (HED diet) supplemented with tallow and sucrose. In the second experiment, MG biopsies and blood samples were collected at the beginning of the treatment period as well as at several calendar and biological ages. In both experiments, MG samples were collected at necropsy for whole mount (WM) and histological analysis, and immunohistochemistry for the proliferation marker Ki67. In Exp. 1, there was no difference in mammary development or epithelial cell proliferation rate due to either dietary energy level or lysine level. Histomorphological analysis of biopsy and necropsy samples from the second experiment revealed more developed TDLU in HED gilts, coincident with lower rates of epithelial proliferation. In addition to decreased MG development at the cellular level, HED gilts developed a larger fat pad that was not as fully penetrated by the ductal tree. Elucidating the effects of the estrous cycle and excess dietary fat on MG development before, during, and after puberty stands to provide a better understanding of normal fluctuations in MG development and how obesity can affect both normal MG physiological function and breast cancer risk.

Studies are also done in her lab on partitioning of heat loss from skin and pulmonary system of cattle and buffaloes as a result of exercise or exposure to heat stress. Dr. R.C. Upadhyay is working as Head, Dairy Cattle Physiology Division at National Dairy Research Institute, Karnal (India). He graduated in Veterinary Sciences and obtained his PhD degree in Animal Physiology. His area of recent research is climate change, stress, and environmental physiology. His major research accomplishment is on climate change impact assessment of milk production and growth in livestock. His work also involves studying methane conversion and emission factors for Indian livestock and use of IPCC methodology of methane inventory of Indian livestock. Heat shock protein-70 expression studies in cattle and buffaloes are also done in his lab. Draught animal power evaluation, fatigue assessment, work-rest cycle and work limiting factors form the highlights of his work. Studies on partitioning of heat loss from skin and pulmonary system of cattle and buffaloes and electrocardiographic studies in cattle, buffalo, sheep and goat are also undertaken in his lab. He has more than 75 research papers, four books and several book chapters to his credit. Technologies developed and research done by him include methodology of methane measurement: open and closed circuit for cattle and buffaloes; inventory of methane emission from livestock using IPCC methodology; livestock stress index: thermal stress measurement based on physiological functions; and draught power evaluation system and large animal treadmill system. He received training in Radio-nuclides in medicine at Australian School of Nuclear Technology, Lucas heights, NSW, Australia in 1985 and Use of radioisotopes in cardiovascular investigations at CSIRO, Prospect, NSW, Australia, during 1985-86. He has guided several post-graduate and PhD students. He is recipient of Hari Om Ashram Award-1990 (ICAR) for outstanding research in animal sciences.
