Book excerpt: This project was conducted to fill the gaps in understanding of the influence of salinity and dissolved organic carbon (DOC) on the acute toxicity of individual metals (silver, copper, zinc) and their mixtures to the saltwater rotifer Brachionus plicatilis. Forty-eight-hour static-acute toxicity tests were conducted at three salinity levels (4, 14, 26 g/L salinity) and a constant DOC level. Fortyeight-hour static-acute toxicity tests were also conducted testing individual metals at constant salinity (14 g/L) and varying DOC levels (1.4, 5.0, 10.0 mg C/L). Metal toxicity was also evaluated using a toxicity unit approach to determine if additive, synergistic, or antagonistic mechanisms occurred when exposed to bimetal mixtures. The effects salinity has on metal toxicity were mixed and depended on the exposure time and metal. Increases in DOC decreased metal toxicity in the majority of treatments. Finally, over 90% of the metal treatments indicated antagonistic or additive effects when in mixture.

Dissolved organic matter (DOM) and reduced sulfur species can complex copper, influencing speciation and decreasing bioavailability. DOM composition in estuaries can vary, depending on allochthonous, autochthonous, or wastewater source. At a molecular level, variability in DOM quality potentially results in different copper binding affinities. The aim of this study was to characterize and quantify DOM and reduced sulfur in estuaries and investigate possible correlations between these parameters and the capacity to complex copper, reducing its toxicity. This study will have implications on the development of marine-specific toxicity prediction models. DOM was characterized in seventy-one estuarine samples through DOC concentration and fluorescence measurements, combined with spectral resolution techniques, to quantify humic-, fulvic-, tryptophan-, and tyrosine-like fractions. Reduced sulfur was measured by the chromium-reducible sulfide (CRS) technique. Acute copper toxicity tests were done on a subset of samples expressing extreme DOC, fluorescent allochthonous, autochthonous, and CRS concentrations. The results showed significant differences in DOM quality, independent of DOC concentration. In terms of total fluorescent material, humic-like material ranged from 9.48% to 66.1%, followed by fulvic-like with a range of 14.5% to 63.2%, and 0.00% to 36.5% for tryptophan-like and 0.64% to 25.2% for tyrosine-like material. CRS was widely variable among the samples; concentrations ranging from 0.5 nM to 7800 nM. The toxicity results suggested DOC was a very good predictive measure of copper EC50 in estuaries (r2 = 0.84) independent of DOM quality. Furthermore, CRS was saturated at low copper concentrations indicating strong binding sites for copper, suggesting that while CRS is protective, it does not bind copper at toxicologically relevant concentrations and therefore is not a good predictive measure of copper toxicity.

Book excerpt: In San Diego Bay, marine organisms are exposed to non-point source stormwater effluent containing the contaminants of concern copper and zinc. United States Environmental Protection Agency (U. S. EPA) Whole Effluent Toxicity (WET) test methods were developed to evaluate the toxicity of continuous point source discharges. These tests are now applied to episodic stormwater discharges. There is concern that static or static-renewal WET methods are not representative the episodic, short-term nature of stormwater discharges. This study modified WET test methods to assess the relative toxicity of copper and zinc under three discharge scenarios, corresponding to the 50th , 75th, and 95th percentile of historical rainfall durations in San Diego. Pulsed contaminant studies were initiated with two common WET test species, the purple sea urchin (Strongylocentrotus purpuratus), and mysid shrimp (Americamysis bahia), representing chronic and acute toxicity test endpoints. To mimic San Diego rain events, laboratory assays were performed for copper and zinc individually and as mixtures at time durations of 3, 6, and 12-hours, followed by transfer to uncontaminated filtered seawater from San Diego Bay for the remainder of the traditional 96-hour WET testing period. LC50 and EC50 values were calculated for copper and zinc exposures at each time-point. Traditional 96-hour static reference toxicant tests were performed concurrently as a means to compare static exposure LC/EC50 values to those found from pulsed toxicity tests. For both sea urchin and mysid shrimp, an increase in contaminant exposure time corresponded to increased toxicity, resulting in LC/EC50 values up to two orders of magnitude greater than standard 96-hour tests. Copper and zinc mixtures elicited less than additive toxicity for all exposure regimes, suggesting that mixtures were less toxic than when exposed as single metal exposures for these contaminants. Exposure times used in standard WET testing likely overestimate the toxic effects of short-term episodic stormwater discharges influencing San Diego Bay. The results suggest that current WET testing protocols may be unnecessarily conservative for estimating episodic discharges toxicity.

Such an approach is unfortunately hampered by the following constraints : the costs as weil as the technical difficulties of analyzing every individual chemical which may be present in the sampies, and the difficulty of assessing the hazards and risks of environmental contaminations from a set of chemical data. During the last decades the scientific and regulatory community has gradually realized that biological methodologies have to be taken into consideration for an ecologically meaningful assessment of the toxicological hazards of contaminants. Effect evaluations obtained with biological techniques indeed integrate the impact of all the contaminants to which living biota are exposed. Bioassays with selected test species representative for the biological commumtles of the environments under consideration, are now applied more or less regularly to determine toxic and genotoxic effects. Taking into account the species specific and chemical specific character of toxicity to biota, the necessity of a «battery of tests» approach with species of different trophic levels is currently also generally accepted and implemented. It is dear that a balanced partnership between chemical, biological, toxicological and microbiological analyses is always the best strategy for generating the broadest information base on environmental hazards.
