Book excerpt: Abstract: Nine papers developed from a 1981 international workshop summarize current knowledge of the role of proteins (and amino acids) in lipid metabolism and atherosclerosis for nutritionists, clinicians, and other health professionals. The topics range from studies on lipid metabolism and experimental atherosclerosis in animals (rats, rabbits, swine), to effects of proteins on human serum lipids and lipoproteins. Other papers discuss the effects of soy protein and casein, influences of dietary protein on serum cholesterol, and the immunological aspects of atherosclerosis. The results of these studies indicate that dietary protein can make a significant contribution to atherosclerosis, that soy protein produces lower serum cholesterol levels than casein (the dynamics of which are described), and that diets containing animal protein are generally more cholesterolemic and atherogenic (in rabbits, specifically) than diets containing vegetable protein. The protein effect can be modified by other dietary components (e.g., fiber, fat). Other findings (e.g., the role of dietary protein in the immunological aspects of atherosclerosis) also are reported. (wz).

Since the first of these symposia in 1960 these triennial meetings have been devoted to the exploration of new ideas, new data and new concepts related to lipid metabolism and atherosclerosis. The sixth Meeting was particularly stimulating in this regard. The concept of the "protective" action of HDL was thoroughly explored within the framework of its molecular biology with data on its epidemiological as well as its in vitro mechan ism(s) of action being discussed. The action of drugs on arterial and HDL metabolism was also discussed as were newer aspects of platelet aggregation, especially as related to prostaglandins. New ground was also broken in discussions of lipid mobilization and mechan isms of hypocholesteremia. Although mice including genetically modified mice offer many possibilities, other non-murine species are absolutely required in some circumstances. Rabbit is one of these species, which has been widely used in biomedical studies. This animal is genetically and physiologically closer to humans including cardiov- cular system and metabolism characteristics. Rabbit is thus more appropriate than mice to study some diseases such as atherosclerosis and lipid metabolism. Because of its larger size, surgery manipulation, bleeding, and turn-over studies are much easier performed in rabbits than in mice. Furthermore, transgenic rabbits can be produced using microinjection and other methods such as lentiviral v- tors. Cloning in rabbits has been proved possible, even though still laborious and time-consuming. Hopefully, functional rabbit ES cell lines will be available in the coming years. Gene deletion or knock-out in rabbits will then become possible.

In this volume, which contains 59 chapters, an internationally recognized group of authors con tribute up-to-date accounts of molecular and cellular processes occurring in the vessel wall in atherogenesis and describe approaches to the prevention and treatment of atherosclerosis. The volume is divided into six major sections. Two sections deal with current aspects of lipoprotein metabolism. In Part I, we are alerted to the impact on li poprotein metabolism of structural heterogeneity within the four broad lipoprotein classes. Attention then turns to the components that orchestrate lipoprotein metab olism. Apolipoprotein identities, processing, and functions are described, as are the roles of lipid transfer proteins in plasma lipoprotein remodeling. Hepatic lipase synthesis and secretion are described. In Part II, Nobel Laureates Michael S. Brown and Joseph L. Goldstein describe mutations in the LDL receptor that reveal the functions of its various domains and point out how understanding the LDL receptor has provided a rational basis for its regulation. A discussion of the role of receptors in regulating cholesterol uptake by tissues follows, with emphasis on receptor-ligand interactions. Additional pathways for cholesterol delivery to cells are explored, as are pathways for cholesterol egress.
