Book excerpt: Intake of polyunsaturated fatty acids (PUFA) has changed drastically in the past century in the American diet has received attention due to potential effects on chronic inflammation-related metabolic diseases. In this project, the effects of dietary PUFA content and the n-6 to n-3 ratio on inflammatory responses in the acute and chronic inflammation models were evaluated. The PUFA content was modified on a Western diet background to deliver both n-6 and n-3 intakes at the 10th and 90th percentile of the population in the United States, and models of acute and chronic inflammation were tested in mice model. The experimental PUFA diets had a modest effect on the response to lipopolysaccharide (LPS) in the acute inflammation model. A high n-6 to n-3 ratio promoted lipogenesis. In addition, high n-6 PUFA enhanced the inflammatory responses via the NF-ÎðB pathway. In chronic inflammation model, low-grade inflammatory stress was induced by osmotic mini-pump delivery of LPS. A high n-6 intake increased glucose intolerance. On the other hand, high n-6 intake promotes cholesterol ester accumulation in both acute and chronic inflammation models.

They have evolved from relatively simple hydrocarbons that serve as depot storages of metabolites and barriers to the permeation of solutes into complex compounds that perform a variety of signalling functions in higher organisms. This volume is devoted to the polar lipids and their constituents. We have omitted the neutral lipids like fats and oils because their function is generally to act as deposits of metabolizable substrates. The sterols are also outside the scope of the present volume and the reader is referred to volume 28 of this series which is the subject of cholesterol. The polar lipids are comprised of fatty acids attached to either glycerol or sphingosine. The fatty acids themselves constitute an important reservoir of substrates for conversion into families of signalling and modulating molecules including the eicosanoids amongst which are the prostaglandins, thromboxanes and leucotrienes. The way fatty acid metabolism is regulated in the liver and how fatty acids are desaturated are subjects considered in the first part of this volume. This section also deals with the modulation of protein function and inflammation by unsaturated fatty acids and their derivatives. New insights into the role of fatty acid synthesis and eicosenoid function in tumour progression and metastasis are presented.

It is fascinating that dietary alterations of fatty acid intake can result in a range of salutory changes in a great variety of medical conditions. Most of the good scien tific work which has led to these observations has been performed over just the last two decades. This is of course not a very long time in the context of the history of the human species. Recently performed analysis of fat intake from paleolithic times has indicated that our hunter-gatherer ancestors consumed as much cholesterol as modern Western man, but strikingly less saturated fatty acid and more polyunsatu rates, including n-3 fatty acids. Wild game has the terrestrial source of n-3 incorpo rated in its fat since browsing animals derive 18:3n-3 (alpha-linolenic acid) natural ly from leafy plants. There is, however, little opportunity for modern Western man to get n-3 fatty acids from the diet if one does not consume fish. Modern agribusiness provides ani mal feeds high in n-6 fatty acids, mostly derived from linoleic acid (18:2n-6) in corn feed. Therefore, grazing animals have no access to alternative fatty acids in either feed or grasses, the latter containing little or none of these potentially beneficial highly polyunsaturated fatty acids.

Among the polyunsaturated fatty acids, both n-3 and n-6 fatty acids exhibit differential effects, and their dietary ratio plays a significant role in the overall impact in an individual. This is most evident with the fact that the decrease in the n-3 intake in recent times has significantly contributed to the severity of chronic inflammation. The mechanism by which these fatty acids and bioactive unsaponifiable embedded in the dietary oils modulate the critical genes, thereby alter the pathology of inflammatory complications is under study for many decades. In this chapter, the role of polyunsaturated fatty acids and their modulatory effects on some of the significant inflammatory complications are discussed.

The conference had two objectives: (1) to review the research data on the health effects of polyunsaturated fatty acids in seafoods in terms of the impact of omega-3 fatty acids on eicosanoid formation, thrombosis and inflammation, and the role of docosahexaenoic acid in membrane function and metabolism, and (2) to develop a research agenda to determine the spectrum of the health effects of polyunsaturated fatty acids of seafood origin in the American diet. The 1985 conference established the fact that omega-3 fatty acids of marine origin - eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) - play important roles in prostaglandin metabolism, thrombosis and atherosc1erosis, immunology and inflammation, and membrane function. In response to the conference recommendations, the Congress of the United States provided special funding for the establishment of a "test materials 1aboratory" within the US Department of Commerce to produce under documented quality contr01 the types and quanti ties of omega-3 te~3t materials required by biomedica1 researchers. The forms of test materials to be produced include refined fish oil, polyunsaturated fatty acid enhanced triglycerides, concentrates of esters of fatty acids, purified omega-3 fatty acids, and omega-3 mono-, di- and tri~lyceride mixtures.

Book excerpt: N-6 polyunsaturated fatty acids (PUFA) such as arachidonic acid (ARA, 20:4) and n-3 PUFA as docosahexaenoic acid (DHA, 22:6) are precursors to bioactive lipid mediators, which have important roles in many brain functions including inflammation. Previous studies have shown that dietary n-6 PUFA deprivation in pups can change brain ARA and DHA levels. However, the effects of n-6 PUFA deprivation on brain lipid metabolism during maternal stage has yet to be studied. Moreover, n-6 PUFA have been linked to the pathogenesis of many neurological disorders through many mechanisms, including conversion to ARA-derived lipid mediators (pro-inflammatory mediators) in the brain. Though diets low in n-6 PUFA are known to reduce levels of ARA and ARA-derived lipid mediators, no one has yet investigated the effects of lowering dietary n-6 PUFA on modulating brain inflammation and resolution in neuroinflammation. The first objective of this thesis was to examine how lowering dietary n-6 PUFA, from gestation, would change levels of hippocampal n-6 and n-3 PUFA and their lipid mediators in comparison to the offspring period. We found that maternal exposure to an n-6 PUFA deprived diet does not exacerbate the reduction in ARA and ARA-derived lipid mediators within hippocampal phospholipids (PL) relative to offspring exposure, which suggests that offspring exposure is a main regulating factor of these PUFA and their derivatives. Therefore, we used the offspring stage as the feeding period for the subsequent neuroinflammation study. Neuroinflammation was induced using intracerebroventricular injection of lipopolysaccharide (LPS) and the hippocampus was examined at several time points. Following LPS, different pro-inflammatory gene expression was associated with enrichment in gene expression categories associated with inflammation. However, these pro-inflammatory markers were not reduced by lowering dietary n-6 PUFA and importantly did not impair the resolution of neuroinflammation. In conclusion, we found that exposure of the offspring to an n-6 PUFA deprived diet was sufficient to lower the concentrations of ARA and other n-6 PUFA in hippocampal total PL. In a neuroinflammation model, the expression in gene ontology categories associated with inflammation was significantly increased and then resolved in response to LPS administration, however, irrespective of an n-6 PUFA diet.
