Available in PDF, EPUB and Kindle. Book excerpt: The biological membranes of cellular organization enfold an important group of membrane proteins called the ATPases, which are not only versatile in maintaining chemical gradient and electrical potential across the membrane but also bring metabolites necessary for cell metabolism and drive out toxins, waste products and solutes that otherwise can curb cell functions. ATPases are distributed virtually in all live forms starting from unicellular to multicellular and also in viruses. There are different types of ATPases, which differ in function and structure and in the type of ions they transport. The three main types of the ion pump ATPase family are: (i) P-type ATPases that transport different ions across membranes and Ca2+ATPases belongs to this catagory (ii) F-type ATPase in mitochondria, chloroplasts and bacterial plasma membranes produce ATP using the proton gradient; and (iii) V-type ATPase catalyzes ATP hydrolysis to transport solutes and maintains acidic pH in organelles like lysosomes. Genetic defects in either of the ATPases cause several diseases and a number of researches have demonstrated the involvement of the members of ATPases in the cell pathology and diseases, thereby penetrating exciting new areas of our understanding. In this book, the authors summarize recent knowledge about the molecular mechanisms associated with Ca2+-ATPase, V-ATPase and F-ATPase in intracellular and extracellular Ca2+ transport, mitochondrial ATP synthase, vesicular H+ transport, and lysosomal pH regulation. This book thereby bridges the gap between fundamental research and biomedical and pharmaceutical applications. The book provides an informative resource to improve ATPase research and modern therapeutic approaches toward different life threatening diseases that are associated with dysregulation of the ATPases.

Book Cellular Biology of Ectoenzymes

Download or read book Cellular Biology of Ectoenzymes written by Georg W. Kreutzberg and published by Springer Science & Business Media. This book was released on 2012-12-06 with total page 313 pages. Available in PDF, EPUB and Kindle. Book excerpt: Cells do not normally live as single entities but are grouped together in specific functional and structural configurations in various tissues. Intra cellular mechanisms maintain cellular viability and provide the means necessary for their specific cellular functions. The interaction between cells is maintained by mechanisms involving extracellular signalling. Such extracellular mechanisms may include special properties of the cell surface which involve immediate cell contact, but may also represent mechanisms which act at a distance and are mediated via special secretions and/or re ceptors. Recent studies on cell-cell contact have tended to stress cell sur face components directly mediating cellular interactions; the extracellular medium as a metabolically active compartment has been rather neglected. However, it represents a vital medium through which cells communicate, being important in, for example, chemotaxis in primitive organisms, and in devel~ment and in the coordination of multiple functions in multi cellular 0 ganisms. It is not surprising, therefore, that a number of mole cular me anisms have developed together with increasing biological complexi during evolution. Two aspects of the extracellular space have received increasing attention in the last few years. First, several macro molecules such as collagen, laminin and fibronectin have been identified as components of an extracellular matrix giving a structural dimension to the extracellular compartment.

Book Properties of Avian Ecto ATPases

Download or read book Properties of Avian Ecto ATPases written by and published by . This book was released on 1998 with total page 234 pages. Available in PDF, EPUB and Kindle. Book excerpt: Avian plasma membranes contain plasma membrane-bound ecto-ATPases which hydrolyze ATP in an extracellular cascade eventually producing adenosine and inorganic phosphate. Ecto-ATPases are distinct from other ATPases in that they are known to be insensitive to vanadate, oligomycin, and N-ethyl maleimide, known inhibitors of the P-, F-, and V-type ATPases, respectively. Studies conducted on chicken ecto-ATPases from cardiac and smooth muscle and liver show a variety of differing kinetic properties. The results of which suggest that although each enzyme hydrolyses extracellular ATP, they should each be viewed as distinct subtypes of the whole ecto-ATPase family. Kinetic and immunochemical studies focusing on ATPase and ADPase activities of both liver and smooth muscle tissues showed a number of tissue-dependent responses. These data suggested that in smooth muscle membranes an ecto-ATPase is responsible for the majority of ATPase activity, while a putative non-specific ADP hydrolase is responsible for the majority of ADPase activity. The data also indicate there exists an ecto-ATPDase which is responsible for a small fraction of both activities. In liver membranes, the results indicate that there exists an ecto-ATPDase which catalyzes the majority of ATPase and ADPase activities and the possible existence of an ecto-ATPase and the putative non-specific ADP hydrolase, which account for the very small fraction of both activities. A model incorporating the nucleotidase activities on both membranes will be presented and discussed. Extensive kinetic characterization of the smooth muscle ecto-ATPase provided evidence that this enzyme in chicken smooth muscle membranes is influenced by oligomerization. Molecules which shift the monomer $\leftrightarrow$ oligomer equilibrium to the left are inhibitors, while molecules which shift the monomer $\leftrightarrow$ oligomer equilibrium to the right are activators. The possible physiological implication of this model with cellular adhesion will be discussed.

Book Proteins and Proteomics of Leishmania and Trypanosoma

Download or read book Proteins and Proteomics of Leishmania and Trypanosoma written by André L.S. Santos and published by Springer Science & Business Media. This book was released on 2013-11-22 with total page 387 pages. Available in PDF, EPUB and Kindle. Book excerpt: This book contains a collection of critical reviews on the expression of biologically functional proteins in Leishmania and Trypanosoma, which was written by renowned researchers on this field. Species belonging to these trypanosomatids’ genera are etiological agents of leishmaniasis, Chagas’ disease and sleeping sickness that are extremely debilitating human infection diseases, which remain a major health problem especially in countries from Latin America, Africa and Middle East. Substantiating the problem, the currently accepted drugs for these diseases are quiet unsatisfying due to their low efficacy and high toxicity. In order to solve these real problems, several research groups around the world have become involved in the study and identification of novel potential targets in the trypanosomatid cell. Since proteins are key macromolecules involved in crucial metabolic processes of all living cells, studies have focused on the expression of specific proteins produced by Leishmania and Trypanosoma by means of different biochemical, molecular and proteomic approaches in order to explore them as targets for understanding the parasite life cycle and developing new strategies against trypanosomiasis. With these proposals in mind, the book “Proteins and Proteomics of Leishmania and Trypanosoma” encompasses (i) an integrated view about the biochemistry of parasites belonging to the Leishmania and Trypanosoma genera; (ii) an updated review on the expression of biologically relevant proteins by human pathogenic trypanosomatids and their possible role in the interaction with host cells/molecules as well as a target for development of both alternative chemotherapies and vaccine; and (iii) several pictures, diagrams and tables that can be used to illustrate both undergraduate and postgraduate teaching as well as scientific lectures, being a useful resource for students and researchers.

Book Current Directions in Insulin Like Growth Factor Research

Download or read book Current Directions in Insulin Like Growth Factor Research written by Derek LeRoith and published by Springer Science & Business Media. This book was released on 2012-12-06 with total page 413 pages. Available in PDF, EPUB and Kindle. Book excerpt: The study of the insulin-like growth factor (IGF) family has become an exciting area of investigation. Initially, this family consisted of ligands (insulin, IGF-I and IGF-m and receptors (the insulin receptor, the type I or IGF-I receptor and the type II or IGF-IYM-6-P receptor). Subsequently, it was discovered that six specific binding proteips (lGFBPs 1-6) playa major role in the actions of this growth factor family. In addition, there are now more potential receptors when one considers the possible roles of the insulin-receptor related receptor (IRR) and hybrid receptor dimers composed of insulin and IGF-I receptor (half-receptors). Another important aspect of this area of research is the realization that the IGFs are not only essential for normal growth and development but, in addition play an important role in the normal specialized function(s) of all tissues of the body, including the nervous system, skeleton, reproductive system, kidney, and the immune system, to name but a few. The development of recombi!tant human IGF-I for clinical testing has been a major breakthrough for investigators. Potential uses include wound healing, reversal of catabolic states, diabetes, bone remodeling, recovery from acute renal failure and many others. will determine both its use and its potential hazards.

Available in PDF, EPUB and Kindle. Book excerpt: A widespread appreciation for the role that calcium plays in cell physiology and patho physiology has now been achieved due to the pioneering studies of many of the scientists who attended the VIIth International Spring Symposium on Health Sciences at George Washington University in Washington, D. C. The participants in this unique meeting rep resented diverse fields of basic and clinical research, such as molecular physiology, oncology, molecular genetics, cardiology, bioenergetics, pathology, and endocrinology. The content of the proceedings of the symposium represents work in these and other areas of biomedical research. Organization of the book is aimed at striking a balance between the biochemistry 2 and physiology of normal cell Ca + metabolism and the pathological consequences of al 2 terations in cell Ca + homeostasis. The first section of the book is devoted to the transport mechanisms responsible for 2 2 regulating intracellular Ca + and the pharmacological modalities for controlling cell Ca + . Particular attention is given to the molecular basis for plasma membrane transport activities, 2 2 including the ATP-driven Ca + pump, the Na + -Ca + exchange system, and voltage sensitive 2 Ca + channels. The second section covers the exciting relationships between phosphoinositide metab 2 olism, signal transduction, and cell Ca + metabolism. This section begins with an eloquent overview by Professor Michael Berridge, who was the keynote speaker at the symposium and the recipient of the scientific merit award.

Book Molecular Parasitology

    Book Details:
  • Author : Julia Walochnik
  • Publisher : Springer
  • Release : 2016-10-20
  • ISBN : 3709114160
  • Pages : 546 pages

Download or read book Molecular Parasitology written by Julia Walochnik and published by Springer. This book was released on 2016-10-20 with total page 546 pages. Available in PDF, EPUB and Kindle. Book excerpt: In the past years, genome projects for numerous human parasites have been completed and now allow first in depth comparisons and evolutionary conclusions. The genomes of parasites reflect the coevolution with their host, metabolic capacities depending on their respective habitat in the host. Gut parasites usually have an anaerobic metabolism, while blood parasites have an aerobic metabolism, intracellular parasites escape the immune system, while extracellular parasites evade the immune system, usually by antigenic variation. Comprehensive genome data now being available allow us to address profound scientific questions, such as which traits enable the parasite to survive in the human host, which to cause disease and which can be used as drug targets. This book intends to give an overview of the state of knowledge on “the molecules” of protozoan parasites – on their genomes, proteomes, glycomes and lipidomes.
