As in many species, equine articular cartilage does not readily regenerate, and cartilage injury often results in the formation of mechanically inferior scar tissue characterized as fibrocartilage. Because this fibrocartilage is lacking in mechanical integrity, it is unable to withstand long-term physiologic loading and eventual undergoes degradation over time. This process of cartilage deterioration is known as osteoarthritis, a common condition affecting both human and veterinary species. Cartilage engineering seeks to recapitulate the organized extracellular matrix (ECM) of native cartilage. The self- assembling process (SAP) is a scaffold free approach in which chondrogenic cells are densely seeded in non-adherent wells and stimulated to generate cartilage ECM in defined medium conditions. After several weeks in culture, self-assembled chondrogenic cells yield neocartilage with morphological, biochemical, and mechanical properties similar to native cartilage. Properties of equine articular cartilage, in particular mechanical properties of the stifle and cervical facet joint, are largely understudied. This body of work aims to characterize the properties of native equine articular cartilage from these understudied joints as well as demonstrate the potential of the SAP to generate equine cartilage grafts from an equine mesenchymal stem cell (MSC) source. Additionally, this work develops an in vitro model for post-traumatic osteoarthritis (PTOA) in the equine. Taken together, these data establish design criteria for cartilage repair efforts, demonstrate the potential of equine MSCs as a cell source of cartilage repair, and propose a model for equine PTOA that can be used to develop future therapies to treat cartilage injuries in the equine athlete.
