Book excerpt: The National Institute of Standards and Technology (NIST) established a Vitamin D Metabolites Quality Assurance Program (VitDQAP) in collaboration with the National Institutes of Health Office of Dietary Supplements (NIH-ODS) in 2009. The VitDQAP at NIST administered twelve interlaboratory comparison exercises through 2016 for the measurement of vitamin D metabolites in human serum and plasma. The comparability of participant measurements for target analytes improved substantially over time through the development and promulgation of robust measurement technologies, identification and production of suitable reference materials, isolation and identification of measurement system biases, and support and encouragement of within-laboratory measurement quality control efforts. This VitDQAP Final Report summarizes the program and provides highlights of the twelve exercises conducted during the program's eight-year lifetime.

Book excerpt: The National Institute of Standards and Technology (NIST) recently established a Vitamin D Metabolites Quality Assurance Program (VitDQAP) in collaboration with the National Institutes of Health (NIH) Office of Dietary Supplements. Participants in the fourth exercise of this program, the Summer 2011 Comparability Study, were asked to use the methodology of their choice to measure concentrations of 25-hydroxyvitamin D in control and study materials distributed by NIST. The study materials consisted of SRM 968e Fat-Soluble Vitamins, Carotenoids, and Cholesterol in Human Serum (Level 1, Level 2, and Level 3) and SRM 968d Fat-Soluble Vitamins, Carotenoids and Cholesterol in Human Serum (Level 1). SRM 2972, which is comprised of separate ethanolic calibration solutions with known concentrations of 25(OH)D2 and 25(OH)D3, was provided as a control material. Participants provided their data to NIST, where it was compiled and evaluated for trueness relative to the NIST value and concordance within the participant community. A report of results was provided to all participants of the exercise, and laboratories were identified by code numbers known only to them. The results from this fourth exercise are reported along with a summary of the analytical methods used.

HAMQAP was established to enable laboratories to improve the accuracy of measurements in samples that represent human intake (e.g., foods, dietary supplements, tobacco) and samples that represent human metabolism (e.g., blood, serum, plasma, urine) for demonstration of proficiency and/or compliance with various regulations. Analytes are paired where possible to represent the full spectrum of health assessment. Exercise 6 of this program offered the opportunity for laboratories to assess their in house measurements of nutritional elements (chlorine, iodine, chromium, molybdenum, and selenium), toxic elements (arsenic, cadmium, lead, and mercury), contaminants (chlorate and perchlorate, glyphosate and aminomethylphosphonic acid (AMPA)), water-soluble vitamins (biotin and vitamin C), fat-soluble vitamins (vitamins A and E), fatty acids (select omega-3 and omega-6 fatty acids), botanicals (anthocyanidins), natural products (caffeine, theobromine, and theophylline), and proximates (fat, protein, carbohydrates, solids, ash, and calories) in foods and dietary supplements, and corresponding biomarkers/metabolites in clinical specimens (human red blood cells).
