Book excerpt: Earlier titles in this series have directed attention to selected questions of current theoretical or practical interest in developmental biology, without special emphasis upon individual biological systems. Tho choose giant polytene chromosomes as the common denominator of a new volume may at first sight seem to set rather arbitrary restrictions upon a coherent discussion of the basic problem of cellular differentiation. However, the reader will find out for hirnself that, on the contrary, such a limitation, though dicta ted by the material, focuses the analysis upon one essential point: the differential release of genetic information from operational subunits of the eukaryotic chromosome. In the organization of this volume we have not striven for completeness - some, indeed, may fee 1 that the work on Sciarids is underrepresented - but the various contributors have made every effort to render a thorough and up-to-date picture of the situation in their particular field, and on this basis to discuss all possible angles of general biological interest. Authors and editors have agreed to dedicate this volume to the memory of JACK SCHULTZ.

Contributors explore the role of chromosomes in development, highlighting important processes such as the diversification and specialization of cells; the regulation of gene function by the physical state of the chromosome; the functional organization of chromosomes; DNA replication and synthesis; and macronuclear differentiation and subnuclear assortment. This volume is organized into 11 chapters and begins with an overview of chromosome structure and function and the mechanisms underlying the role of the chromosome in embryonic development. This book then discusses the chromosomal DNA replication, the mechanisms of gene modulation, the binding of actinomycin to DNA, and the cytological aspects of DNA replication in higher animals. The next chapters explain localized DNA synthesis in polytene chromosomes; chromosomal RNA and other nuclear RNA fractions; and experimental work probing both genetic and functional mosaicism in the mouse, as well as genetic repression of the R locus in maize. The book also considers how developmental pathways are controlled by genetic mechanisms, and then concludes with a chapter on nuclear differentiation in ciliates and on the phenomenon of subnuclear assortment. This book will be of interest to developmental biologists and embryologists.

Book excerpt: In 1906 Castle, Carpenter, Clarke, Mast, and Barrows published a paper entitled "The effects of inbreeding, cross-breeding, and selection upon the fertility and variability of Drosophila." This article, 55 pages long and published in the Proceedings of the Amer ican Academy, described experiments performed with Drosophila ampe lophila Lov, "a small dipterous insect known under various popular names such as the little fruit fly, pomace fly, vinegar fly, wine fly, and pickled fruit fly." This study, which was begun in 1901 and published in 1906, was the first published experimental study using Drosophila, subsequently known as Drosophila melanogaster Meigen. Of course, Drosophila was known before the experiments of Cas tles's group. The small flies swarming around grapes and wine pots have surely been known as long as wine has been produced. The honor of what was the first known misclassification of the fruit flies goes to Fabricius who named them Musca funebris in 1787. It was the Swedish dipterist, C.F. Fallen, who in 1823 changed the name of ~ funebris to Drosophila funebris which was heralding the beginning of the genus Drosophila. Present-day Drosophila research was started just 80 years ago and first published only 75 years ago. Even though the history of Drosophila research is short, the impact and volume of study on Drosophila has been tremendous during the last decades.

It describes a novel approach for how these advances might be used in combination with existing methodologies to further the understanding of mechanisms of developmental toxicity, to improve the assessment of chemicals for their ability to cause developmental toxicity, and to improve risk assessment for developmental defects. For example, based on the recent advances, even the smallest, simplest laboratory animals such as the fruit fly, roundworm, and zebrafish might be able to serve as developmental toxicological models for human biological systems. Use of such organisms might allow for rapid and inexpensive testing of large numbers of chemicals for their potential to cause developmental toxicity; presently, there are little or no developmental toxicity data available for the majority of natural and manufactured chemicals in use. This new approach to developmental toxicology and risk assessment will require simultaneous research on several fronts by experts from multiple scientific disciplines, including developmental toxicologists, developmental biologists, geneticists, epidemiologists, and biostatisticians.
