Book excerpt: "This thesis presents an electrochemical approach for the non-invasive study of biological systems, and demonstrates the usefulness of scanning electrochemical microscopy (SECM) for quantification of chemical flux in live cells. We first explore the presence of D-serine within the brain. This neurotransmitter plays an important role in proper brain development, and has also been linked to several pathological diseases such as schizophrenia and Alzheimer's disease. Consequently, quantification of local changes in D-serine concentration is essential to further understanding of its role in these processes. A biosensor was thus developed for the detection of D-serine with high sensitivity. Using this microscale biosensor, the release of endogenous D-serine was measured in vivo inside the brain of Xenopus laevis tadpoles, revealing an overall concentration of 2.5 μM. However, the primary site and mechanism of release of D-serine remained unclear, requiring a more localized measurement. After optimizing for improved response time and selectivity, the biosensor was used in a SECM configuration to measure localized release of D-serine from a model system. This SECM methodology provided quantitative imaging of D-serine with high spatial resolution, and could be useful to investigate chemical flux in other biological samples. We then shifted our focus to quantitative imaging of another complex live cell process, specifically multidrug resistance. This defensive mechanism allows cancer cells to pump out chemotherapeutic drugs before they can kill the cell, severely impeding treatment success. Most investigations of transmembrane efflux pumps, including multidrug resistance-associated protein 1 (MRP1), focus on expression to determine their effect in specific cancer types. Researchers have been unable to establish a clear correlation between expression and patient survival. However, the relationship between transporter expression and functional activity is often overlooked. Using SECM with complementary redox mediators and novel cell patterning substrates, we measured MRP1 functional activity in single cancer cells. By challenging parental cell lines with doxorubicin, a drug used in many chemotherapy regimens, we produced six cell populations with increasing MRP1 expression, and measured their concurrent functional activity. This comparative study revealed that in fact MRP1 expression and functional activity do not correlate, and suggest that the measurement of functional activity should be prioritized to identify the effect of MRP1 on chemotherapy treatment success." --

In recent years, SICM has been applied in an ever-increasing number of areas in the bioanalytical sciences. SICM is based on an electrolyte-filled nanopipette with a nanometer-scale opening, over which an electric potential is applied. The induced ion current is measured, which allows to directly or indirectly quantify various physical quantities such as pipette-sample distance, ion concentration, sample elastic modulus among many others. This makes SICM well suited for applications in electrolytes - most prominently for the study of live cells. This book starts with a historic overview starting from the days of the invention of SICM by Paul Hansma at the University of California at Santa Barbara in 1989. SICM is a member of the family of scanning probe microscopies. It is related to another prominent member of the family, atomic force microscopy (AFM), which has found application in almost any field of nanoscale science. The advantages and disadvantages of SICM over AFM are also outlined. One of the most effective and break-through applications of SICM nanopipettes is in electrochemistry. The different routes and applications for doing electrochemistry using nanopipettes are also discussed. In addition the book highlights the ability of SICM for surface positioning with nanometer precision to open up new vistas in patch clamp measurements subcellular structures. Finally the book presents one research area where SICM has been making a lot of contributions, cardiac research and the endeavors to combine SICM with super-resolution optical microscopy for highest-resolution joint topography and functional imaging.

Book excerpt: The main theme in this dissertation is to develop Scanning Electrochemical Microscopy (SECM) based electroanalytical techniques to study quantitative biology in real time. The multidrug resistance (MDR) phenomenon in live cancer cells was studied using mimic drug molecules such as menadione with the aid of SECM. Real time quantitative detection of thiodione (menadione-conjugate) pumped out of the cells was determined to be 140 [mu]M due to exposure of 500 [mu]M menadione to the cells. Selective blocking of these MDR pumps in live intact cells was also achieved by small molecules such as MK571 as well as by the MDR specific antibody. An approximately 50% drop in thiodione flux was observed in both cases of MDR pumps inhibition. This SECM technique was also extended to measure the permeability of a highly charged hydrophilic molecule passing through the membrane of a single living cell. The permeability was measured to be 6.5 ± 2.0 × 10-6 m/s. Real time monitoring of morphological changes in a live HeLa cell due to addition of varying concentration of surfactant such as Triton X-100 was also demonstrated by SECM. This electroanalytical technique was also expanded to study quantitative microbiology. Real time quantitative detection of pyocyanin produced by Pseudomonas aeruginosa (PA14 strain) biofilm locally was determined to be 2.5 [mu]M after 6 h. Pyocyanin (PYO) was also observed to be reduced by PA14 biofilm, thus maintaining a reduced atmosphere above the biofilm even in presence of oxygen. Spatial mapping of this reduced PYO showed that this reduced zone was only formed up to 500 [mu]m above the biofilm. The cells are also able to modulate the height of the reduced PYO zone in accordance to the availability of Fe(III/II) in the solution to scavenge iron from the surrounding environment. Real time spatial mapping hydrogen peroxide across polymicrobial biofilm (Sg and Aa) was also achieved with the aid of SECM. The local peroxide concentration produced by Sg was measured to be 1 mM, which is significantly higher than the bulk peroxide concentration. This study also showed that the local concentration across the microbial film is more important than the bulk concentration since bacteria communicate locally in real world.
