Book excerpt: The objective of this study was to develop direct and indirect capture assays for use with Surface Enhanced Raman Scattering (SERS) spectroscopy. Spectroscopically bright gold nanoparticle reporters may be used as detectors for any number of SERS assays. Direct capture assays directly couple the compound of interest to gold coating a sample concentrator. For our proof of concept direct capture assay, we detected cyanide in an aqueous solution. Indirect capture assays couple a Raman reporter (gold nanoparticle tagged with a Raman active compound) to the sample concentrator via antibody antigen interactions. The Raman reporter presents a unique spectrum when excited by an 808 nm laser, and the complex is pulled out of solution using a three separate approaches, paramagnetic, buoyant and neutrally buoyant--the latter two are novel sample concentrating approaches and will be discussed in further detail within this report. The paramagnetic particles are strongly attracted to a magnet and thus the complex is pulled out of solution for interrogation with the laser. The buoyant design physically separates the positively coupled reaction from non-coupled nanoparticles. In the indirect coupled assays, the Raman reporter provides the unique spectrum for identification, and in future work, the assays can be multiplexed. Each compound of interest will be coupled to a unique Raman reporter that will allow multiplexing of this assay development. The method of detection will be SERS. SERS was chosen as the method of choice due to its high sensitivity and selectivity. The ultimate application of this technology will be for use in point of care diagnostics in the emergency department at hospitals, in ambulances, or in developing countries for diagnostic assessment of pathogenic human diseases in small, clinical settings. In this manner, the complex SERS spectra from a multiplexed assay will be reduced to an 'answer box' indicating presence or absence of a marker of interest.

Raman scattering (RS) is a widely used vibrational technique providing highly specific molecular spectral patterns. A severe limitation for the application of this spectroscopic technique lies in the low cross section of RS. Surface-enhanced Raman scattering (SERS) spectroscopy overcomes this problem by 6-11 orders of magnitude enhancement compared with the standard RS for molecules in the close vicinity of certain rough metal surfaces. Thus, SERS combines molecular fingerprint specificity with potential single-molecule sensitivity. Due to the recent development of new SERS-active substrates, labeling and derivatization chemistry as well as new instrumentations, SERS became a very promising tool for many varied applications, including bioanalytical studies and sensing. Both intrinsic and extrinsic SERS biosensing schemes have been employed to detect and identify small molecules, nucleic acids and proteins, and also for cellular and in vivo sensing.

Book excerpt: Surface enhanced Raman spectroscopy (SERS) is a highly sensitive technique for quantifying trace amounts of analyte adsorbed at a roughened metal surface. We have shown that novel gold nanostructured films, simply fabricated using gold nanoparticles and latex microspheres, can be used as highly sensitive SERS substrates. The films are templated by layers of colloidal crystals and display long ranged ordered regions, and porosity on two length scales that results in high surface area. By integrating these substrates into a flow chamber we have obtained consistent reproducible data and demonstrated the quantitative detection of sodium cyanide in the concentration range from 5 to 500 ppb, with detection significantly enhanced by lowering the pH after cyanide adsorption. We have found that the optimum size of the latex microspheres is approximately equal to the wavelength of the laser. Finally, the SERS activity of the gold substrates coated with silver nanoparticles was investigated using methyl phosphonic acid (MPA). Detection of MPA was dependent on the time between substrate coating and the SERS measurement. These studies show that novel materials formed by simple wet chemistry techniques can be used efficiently in practical devices for the detection of chemical agents.

Book excerpt: In the course of the development of surface science, advances have been identified with the introduction of new diagnostic probes for analytical characterization of the adsorbates and microscopic structure of surfaces and interfaces. Among the most recently de veloped techniques, and one around which a storm of controversy has developed, is what has now been earmarked as surface enhanced Raman scattering (SERS). Within this phenomenon, molecules adsorbed onto metal surfaces under certain conditions exhibit an anomalously large interaction cross section for the Raman effect. This makes it possible to observe the detailed vibrational signature of the adsorbate in the ambient phase with an energy resolution much higher than that which is presently available in electron energy loss spectroscopy and when the surface is in contact with a much larger amount of material than that which can be tolerated in infrared absorption experiments. The ability to perform vibrational spectroscopy under these conditions would lead to a new understanding about the chemical identity, geome try, and bonding of adsorbed material at a level previously unacces sible. It is for these reasons that the last few years have brought an explosion of activity surrounding the exploitation of SERS. The search for the origines) of the anomalous enhancement has given rise to a research sub-activity of its own. Efforts to explain the en hancement have led to an increased understanding of the whole range of phenomena associated with the interaction of photons with adsor bates and metal surfaces.

Many formats and detection strategies have been explored, each having unique advantages and disadvantages. More recently, surface-enhanced Raman scattering (SERS) has been introduced as a readout method for immunoassays, and has shown great potential to meet many key analytical figures of merit. This technology is in its infancy and this dissertation explores the diversity of this method as well as the mechanism responsible for surface enhancement. Approaches to reduce assay times are also investigated. Implementing the knowledge gained from these studies will lead to a more sensitive immunoassay requiring less time than its predecessors. This dissertation is organized into six sections. The first section includes a literature review of the previous work that led to this dissertation. A general overview of the different approaches to immunoassays is given, outlining the strengths and weaknesses of each. Included is a detailed review of binding kinetics, which is central for decreasing assay times. Next, the theoretical underpinnings of SERS is reviewed at its current level of understanding. Past work has argued that surface plasmon resonance (SPR) of the enhancing substrate influences the SERS signal; therefore, the SPR of the extrinsic Raman labels (ERLs) utilized in our SERS-based immunoassay is discussed. Four original research chapters follow the Introduction, each presented as separate manuscripts. Chapter 2 modifies a SERS-based immunoassay previously developed in our group, extending it to the low-level detection of viral pathogens and demonstrating its versatility in terms of analyte type, Chapter 3 investigates the influence of ERL size, material composition, and separation distance between the ERLs and capture substrate on the SERS signal. This chapter links SPR with SERS enhancement factors and is consistent with many of the results from theoretical treatments of SPR and SERS. Chapter 4 introduces a novel method of reducing sample incubation time via capture substrate rotation. Moreover, this work led to a method of virus quantification without the use of standards. Chapter 5 extends the methodology developed in Chapter 4 to both the antigen and ERL labeling step to perform assays with improved analytical performance in less time than can be accomplished in diffusion controlled assays. This dissertation concludes with a general summary and speculates on the future of this exciting approach to carrying out immunoassays.

Book excerpt: In the last decade, we have seen spurring growth in academic and industrial interest in Raman spectroscopy and its modifications, in particular, Surface-Enhanced Raman Spectroscopy (SERS). This is evident by the number of SERS publications in the last decade, from 580 in 2005 to almost 2000 in 2014. There are now companies worldwide that are dedicated to manufacturing SERS substrate, sensors, and other SERS-based products. The miniaturization of Raman instrumentation from bulky and expensive bench-top designs to inexpensive handheld spectrometers has allowed SERS to be employed as an in situ detection technique. Some examples of the industrial success of Raman and SERS are rapid raw material identification (RMID), Process Analytical Technology (PAT) and drug screening in pharma and biotech industries, continuous surveillance of food, water, and environmental safety, and the diagnosis and continuous monitoring of diseases like cancer, diabetes, etc. This progress in translating Raman and SERS techniques into commercial use and commercial products is possible because of the close collaboration between scientists and engineers. However, the weak signals of Raman spectroscopy is an inherent limitation of the spontaneous Raman spectroscopy technique. Therefore, several modifications have been made, including but not limited to, surface-enhanced Raman spectroscopy (SERS), spatially offset Raman spectroscopy (SORS), transmission Raman spectroscopy (TRS), coherent anti-Stoke's Raman spectroscopy (CARS), and tip-enhanced Raman spectroscopy (TERS). Spontaneous Raman spectroscopy has become a one-of-a kind, portable, and affordable technology for in-situ detection and continuous monitoring in resource-limited-settings. The primary focus of this textbook is to try to cover the fundamentals in SERS methods of preparing substrates and sensors, and efficient spectral analysis approaches like chemometrics for diverse applications, listed in the paragraph above. However, with a goal to give a true picture to the audience when assessing the scope of spontaneous Raman and Surface-enhanced Raman for specific applications, we have briefly introduced industrially successful examples of other types of Raman spectroscopy techniques, such as SORS, TRS, and CARS.

This dissertation describes the optimization and characterization of self-assembled monolayer (SAM) multilayer SERS substrates for improving performance of different types of substrates. SERS substrates derived from metal film on nanostructures were modified with multiple metal films interspaced with SAM dielectric spacers to achieve multilayered SERS substrates. The fundamental concept of this substrate geometry exploited the cumulative effect of the multiple electromagnetic fields and various properties of SAMs to optimize SERS enhancement of multilayer SERS substrates to about 20-fold compared to conventional substrates.
