Book Characterization and Control of Leukemia Stem Cells in Acute Myeloid Leukemia

Download or read book Characterization and Control of Leukemia Stem Cells in Acute Myeloid Leukemia written by Stanley Wai Kwong Ng and published by . This book was released on 2018 with total page 0 pages. Available in PDF, EPUB and Kindle. Book excerpt: The overall goal of this thesis is to explore the potential application of leukemia stem cell (LSC) properties for improving the treatment and management of adults with acute myeloid leukemia (AML), solidifying the clinical relevance of functional stem cell frequency estimates from xenotransplantation assays to human disease, and gaining insight into the gene regulatory logic underlying blood stem cells. Conventional risk stratification schemes in AML do not account for the leukemia initiating capacity of LSC within patient samples even though mounting evidence indicates its importance in the etiology and pathogenesis of AML. This may be attributed, at least in part, to insufficient data in support of the definitive relevance of xenografts (the gold standard assay for detecting LSC) to human disease. Instead, clinical schemes have focused on an ever-increasing number of genomic lesions for sorting patients into incrementally refined risk categories as newer technologies and larger study cohorts enable higher resolution and more complete capture of the continuum of genomic variation. Here, the derivation of the LSC17 gene expression score resulted in the first LSC-based diagnostic tool that complements and refines current risk assessment schemes, setting the stage for enhancing the overall accuracy of prognostic and predictive models in the clinic. The backbone of AML treatment has largely remained unchanged since the 1970s, and is curative for.

Book Identification and Characterization of a Y Chromosome encoded Gene Overexpressed in Acute Myeloid Leukaemia Cells

Download or read book Identification and Characterization of a Y Chromosome encoded Gene Overexpressed in Acute Myeloid Leukaemia Cells written by and published by . This book was released on 2009 with total page pages. Available in PDF, EPUB and Kindle. Book excerpt: Male patients with acute myeloid leukaemia (AML) who receive female haematopoietic stem cells show the best therapy outcome. Y chromosome gene expression has been evaluated in male AML M4-M5 cells and in male healthy CD14+ cells. The genes BCORL2, PCDH11, TGIF2L, and VCY were found to be up-regulated in leukaemic cells. After immunological studies, one peptide derived from the gene VCY and able to weakly stimulate female cytotoxic T-cells has been identified. Several aspects of the putative transcription factor TGIF2LY have been characterized, including its subcellular localization and identification of its DNA-binding sequence.

Available in PDF, EPUB and Kindle. Book excerpt: B cells used to be considered as a homogeneous population of cells destined to produce antibodies of increasing affinity and to maintain an immunological memory. In recent years, it has been determined that B cells can be subdivided into different subsets characterized by distinct morphologic, phenotypic, and functional features. Presenting results of research work on the definition of B cell subset populations, this book explains the basic mechanisms that control B cell activation, stimulation and regulation. Articles include studies on both normal and malignant B cells and describe the mechanisms underlying T-B cell interactions during the immune response. The most important advances in the field of immunodeficiency are also reported. This volume will be essential not only for basic and clinical immunologists, but also for hematologists, pathologists and rheumatologists with a special interest in the pathogenesis of lymphoproliferative or autoimmune disorders.

Book Genomic Applications in Pathology

Download or read book Genomic Applications in Pathology written by George Jabboure Netto and published by Springer. This book was released on 2018-12-10 with total page 638 pages. Available in PDF, EPUB and Kindle. Book excerpt: The recent advances in genomics are continuing to reshape our approach to diagnostics, prognostics and therapeutics in oncologic and other disorders. A paradigm shift in pharmacogenomics and in the diagnosis of genetic inherited diseases and infectious diseases is unfolding as the result of implementation of next generation genomic technologies. With rapidly growing knowledge and applications driving this revolution, along with significant technologic and cost changes, genomic approaches are becoming the primary methods in many laboratories and for many diseases. As a result, a plethora of clinical genomic applications have been implemented in diagnostic pathology laboratories, and the applications and demands continue to evolve rapidly. This has created a tremendous need for a comprehensive resource on genomic applications in clinical and anatomic pathology. We believe that our current textbook provides such a resource to practicing molecular pathologists, hematopathologists and other subspecialized pathologists, general pathologists, pathology and other trainees, oncologists, geneticists and a growing spectrum of other clinicians. With periodic updates and a sufficiently rapid time from submission to publication, this textbook will be the resource of choice for many professionals and teaching programs. Its focus on genomics parallels the evolution of these technologies as primary methods in the clinical lab. The rapid evolution of genomics and its applications in medicine necessitates the (frequent) updating of this publication. This text will provide a state-of-the art review of the scientific principles underlying next generation genomic technologies and the required bioinformatics approaches to analyses of the daunting amount of data generated by current and emerging genomic technologies. Implementation roadmaps for various clinical assays such as single gene, gene panels, whole exome and whole genome assays will be discussed together with issues related to reporting and the pathologist’s role in interpretation and clinical integration of genomic tests results. Genomic applications for site-specific solid tumors and hematologic neoplasms will be detailed. Genomic applications in pharmacogenomics, inherited genetic diseases and infectious diseases will also be discussed. The latest iteration of practice recommendations or guidelines in genomic testing put forth by stakeholder professional organizations such as the College of American Pathology and the Association for Molecular Pathology, will be discussed as well as regulatory issues and laboratory accreditation related to genomic testing. All chapters will be written by experts in their fields and will include the most up to date scientific and clinical information.
