Book excerpt: Annual fish species found in the family Cyprinodontidae (killifishes, pupfishes, and tooth carps) inhabit isolated bodies of freshwater throughout North and South America and Africa that can experience seasonal drought and desiccation. Many of these species survive extinction by laying drought-resistant eggs which undergo several phases of reversible developmental arrests or diapause during their ontogeny. Desert pupfish (Cyprinodon macularius) are a member of the Killifish family and are federally and California State listed as endangered. Beginning in the 19th century, desert pupfish habitat has been seriously degraded by stream bank erosion, the construction of water diversions that have decimated downstream habitat, excessive groundwater pumping, and the introduction of aquatic invasive species (AIS). AIS function as both predators and potential competitors in habitat where they coexist with pupfish. Desert pupfish live in desert creeks and pools that may be prone to periodic desiccation; if they possess the ability to lay desiccation-resistant eggs embryos of desert pupfish, like some other members of the Killifish family, may be capable of arresting development, spending considerable amounts of time in an embryonic state waiting for the return of water. This trait could be used to control AIS without the loss of pupfish populations; however this species has not yet been evaluated for its ability to lay drought-resistant eggs. I tested the hypothesis that desert pupfish do possess this reproductive adaptive strategy, and that this ability could be used to better manage pupfish and their habitat. Using pupfish raised and protected in an artificial refugium, I placed three female and one male pupfish in each of nine tanks for 1 week until spawning was observed in each tank. The tanks were exposed to three different desiccation treatments (1, 2, and 3 weeks respectively) followed by a rewetting period which simulated the return of water. No pupfish fry hatched from any of the treatments. I then repeated the experiment replacing the natural sand substrate with a dark plastic fiber mat so that eggs could be observed with desiccation for seven days. Again, no pupfish fry hatched. My results indicate that desert pupfish either do not lay desiccation-resistant eggs, or that my experimental population, which had been in captivity with controlled water levels for more than two decades, had lost that potentially adaptive trait. Unless further experiments demonstrate that certain desert pupfish populations possess the ability to lay desiccation-resistant eggs, drying of pupfish habitat to control AIS is not possible.
