Book excerpt: Mast cells are tissue-localized cells that play an important role in immunity and inflammation. Following an offensive event they act as cellular sensors that via the activation of cell surface receptors launch a cellular response culminating in the release of a whole set of inflammatory mediators and products. This response is initially destined to restore tissue homeostasis, but in case of chronic injury or deregulation also promotes pathology. To further understand the action of mast cells in their environmental context it is necessary to decipher the molecular mechanisms of their activation as well as the ensuing cellular responses. This will allow identification of new strategies to promote their beneficial actions or, at the contrary, to interfere with their pathological consequences. While in the past many studies have focused on responses engaged by high affinity IgE receptor because of its implication in the allergic response, it has become clear that mast cells can be activated by multiple types of receptors initiating an intense molecular crosstalk between receptors and signaling pathways that can either synergize, antagonize and in some cases produce new types of responses. Mast cells can indeed react with an astounding diverse array of cellular responses that sometimes are engaged selectively. This Research Topic will focus on selected articles that shed some new light on the molecular mechanisms of mast cell activation, the possible crosstalk between signaling pathways and the ensuing cellular responses that allow mast cells to act as cellular sensors in tissues.

Once activated, they may have either pro- or anti-inflammatory effects that include pathogen killing, safe disposal of apoptotic cells or tissue renewal. The activation state of macrophages is conceptualized by the so-called M1/M2 model of polarization. M2 macrophages are not simply antagonists of M1 macrophages; rather, they represent a network of tissue resident macrophages with roles in tissue development and organ homeostasis. M2 macrophages govern functions at the interfaces of immunity, tissue development and turnover, metabolism, and endocrine signaling. Dysfunction in M2 macrophages can ruin the healthy interplay between the immune system and metabolic processes, and lead to diseases such as insulin resistance, metabolic syndrome, and type 1 and 2 diabetes mellitus. Furthermore, M2 macrophages are essential for healthy tissue development and immunological self-tolerance. Worryingly, these functions of M2 macrophages can also be disrupted, resulting in tumor growth and autoimmunity. The disease itself is highly complex in nature, with variety of clinical manifestations ranging from pinpoi- sized wheals to extensive angiodema. Complexity is also seen in the diversity of possible eliciting factors, the many different clinical subtypes and the therapeutic responsiveness. Only in recent years has a better understanding of the diversity in the different subtypes led to new classi? cations and new evidence-based guidelines for diagnostics and mana- ment of the disease. While mast cells are in the center of most urticaria reactions, it is now clearly understood that the responsible mediators are not only limited to histamines. The current book appears in a series of books by Springer. In 1986, the ? rst monograph was edited by Professor Henz née Chanewsky. Since then, two updates of the book have appeared in the German language with Professor Henz as ? rst editor and T. Zuberbier, J. Grabbe, and E. Monroe as the co-editors of the most recent English version, published in 1998. All these books have been written as a joint effort of Professor Henz together with her team at the Department of Dermatology at the Virchow Clinic, Humboldt University, Berlin.
