Afin de garantir la reproductibilité et l’efficacité du procédé de nettoyage appliqué à l’équipement, celui-ci fait l’objet d’une validation. Parmi les différentes méthodes de nettoyage, le nettoyage manuel est une méthode de nettoyage complexe à valider étant donné le degré de variabilité d’un technicien à l’autre mais également d’un nettoyage à l’autre. L’objectif de ce travail a consisté à proposer une démarche d’optimisation des procédés de nettoyage manuel dans un site de fabrication de formes sèches. La première partie de cette thèse se consacre à la contamination dans l’industrie pharmaceutique. En effet, y sont présentées les différents contaminants rencontrés ainsi que les moyens permettant de maitriser cette contamination. La seconde partie traite du nettoyage des équipements de production ; elle décrit les paramètres influençant le nettoyage, les agents de nettoyage utilisés ainsi que les différentes méthodes de nettoyage employées en industrie. La troisième partie développe la validation du nettoyage d’un point de vue réglementaire et pratique. Enfin, la dernière partie expose un exemple de démarche pour optimiser des procédures de nettoyage manuel d’équipements de production dans un site de fabrication de formes sèches.

Malgré sa consommation en ressources importante (humaines et matérielles), mais aussi en terme de temps, elle peut être considérée comme un tremplin afin de faire évoluer les pratiques pour améliorer la performance industrielle et optimiser les coûts et les délais alloués à ces opérations de nettoyage, mais elle peut également représenter un gain en terme d'hygiène et de sécurité industrielle. Le projet de validation de nettoyage initié à l'usine de WPS à Dakar Sénégal s'inscrit dans une volonté d'amélioration significative de la conformité aux règles des BPF. Lors de cet exercice de validation, certaines erreurs, limites et difficultés n'ont pu permettre d'aboutir à un projet concluant. La validation de nettoyage est un processus qualité qui se doit de s'inscrire dans une démarche d'amélioration continue.
