  • Pages : 308 pages

Download or read book Flow Cytometry written by Marion G. Macey and published by Blackwell Science Incorporated. This book was released on 1994 with total page 308 pages. Available in PDF, EPUB and Kindle. Book excerpt: Flow cytometers were developed in the early 1960s and used initially by research immunologists to sort and analyse cell subpopulations. The system analyses cells as they move in a liquid stream, through a laser beam, over an electronic sensing area. The light-scattering properties of the individual cells allow the identification of different cell types within a mixed population. Recent advances in technology, including the use of monoclonal antibodies, fluorescent dyes and powerful but cheap computers, mean that flow cytometry is now a routine part of clinical practice for haematologists and immunologists. The identification of cell subsets, leukaemia and various lymphomas is now commonplace. Cell cycle analysis and the measurement of DNA aberrations is now clinically feasible. The greater sensitivity of the new generations of bench-top flow cytometers has enabled the identification of bacteria from various sources, including water, sewage and body fluids and this has resulted in a dramatic increase in the use of these machines in microbiology. The editor has spent several years establishing the appropriate techniques for evaluating the ex vivo expression of antigens on leukocytes and has assembled a distinguished team of international contributors who provide practical advice on the application, scope and limitations of flow cytometry. This book will be of use to research workers, to clinical laboratory scientists and to clinicians who require an introduction and authoritative reference to flow cytometry, with an emphasis on clinical application. The distinguished team of contributors includes Richard Camplejohn, Leon Terstappen, Scott Cram and Terry Hoy, who have been at the forefront of flow cytometry development.

Book Clinical Applications of Monoclonal Antibodies

Download or read book Clinical Applications of Monoclonal Antibodies written by Ron Hubbard and published by Springer Science & Business Media. This book was released on 2013-11-11 with total page 254 pages. Available in PDF, EPUB and Kindle. Book excerpt: Immunology has come a long way in the hundred or so years since the general concepts were first enuciated by Metchnikoff, Ehrlich, Von Bebring and others, One of the landmarks in this progress was the invention and development of monoclonal antibody secreting hybridomas by Milstein and bis co-workers in Cambridge. Unlike most modern inventions of this importance that of monoclonal antibody production was made available to the scientific community tbroughout the world unimpeded by patent protection. This may explain tbe unusual rapidity witb which it has been applied to the benefit of mankind in general. This book, representing as it does the proceedings of tbe first International Symposium to be held on the clinical appli cations of monoclonal antibodies, shows just how much bas been achieved within the space of little more than a decade. The enormaus promise of monoclonal antibody technology, which became apparent soon after its discovery, has already progressed a long way towards fulfillment. The contributors to tbis volume, all of whom are actively engaged in monoclonal antibody development and application, represent the state of the art. Professor Vincent Marks V INTRODUCTION It has been some twelve years since the pioneering experiments of Köhler and Milstein led to the discovery of monoclonal antibodies. Single molecular species antiborlies with desired specificities could be produced by the fusion of antibody - producing cells with neoplastic cells.

Book excerpt: Flow cytometry is a state-of-the-art technology that is widely used in biological research and clinical diagnostics. It is a high-throughput platform that allows the quantification, differentiation, and functional study of cells or cell like particles in suspension. Various cell components can be targeted with fluorescently labelled antibodies or fluorescent dyes to allow measurement and analysis of the physical, chemical, and biological properties of individual cells within homogenous or heterogeneous populations. Since its commercialization more than five decades ago, flow cytometry has advanced and become an very important analytical tool in translational research. With the advancement of instrument technology, an increasing number of fluorescent dyes, and an expanding range of monoclonal antibodies, the applications of flow cytometry in applied research continue to grow. The use of multiparametric flow cytometry in translational research provides the ability to rapidly identify different cell populations and to simultaneously measure multiple parameters of single cells for efficiently assessing immune status, decrease/increase of specific immune cell populations, cell activation status, etc. of different cells in preclinical and clinical studies. The primary goal of any research work is to take research results from the "bench to-bed and back". The use of flow cytometry in applied research advances the development of new diagnostic tests or drugs for cancer treatment, immune monitoring, etc. that help in patient care. Today, every biological scientist needs to have basic knowledge of flow cytometry in order to utilise this technology properly in their own research and to understand other’s research work. The present book has been designed to give the knowledge of flow cytometry and its applications to the researchers and teachers. It will allow the readers to utilize the technology in an appropriate way in their research work. This book has describing various applications of flow cytometry like cell health monitoring, immunophenotyping, cell sorting, stem cell characterization, micro-vesicle analysis etc.

Book Basic and Clinical Applications of Flow Cytometry

Download or read book Basic and Clinical Applications of Flow Cytometry written by Frederick A. Valeriote and published by Springer Science & Business Media. This book was released on 2012-12-06 with total page 325 pages. Available in PDF, EPUB and Kindle. Book excerpt: The focus of this symposium was on the present and future capabilities of flow cytometry for both medical and biological applications in cancer. This technology began with quite modest instrumentation, with limited capabilities to answer biological questions. Today, both the clinical workhorses and the powerful multi-laser, multi-detector, sorting machinery, coupled with sophisticated computers and storage devices and the increasing storehouse of markers and dyes, are taking us to the limit and beyond in finding answers to the cause and cure of cancer. In the past, both normal hematopoietic tissue and leukemias have been the tissue samples of choice in the application of flow cytometry, and some of the most recent applications with these tissues are presented here. However, the book also discusses the increasingly sophisticated disaggregation techniques which allow investigators the possibility to train their lasers on solid tumors. Not only can we use flow cytometry with associated fluorescent markers to understand the biology of cancer, but also the wide array of existing and developing markers provides us with important diagnostic tools in the detection of cancer early in either the malignant or relapse process. And the field comes full circle, with the use of the technology for gene mapping and other genetic studies to unlock the basic malignant process.

Book General Methods in Biomarker Research and their Applications

Download or read book General Methods in Biomarker Research and their Applications written by Victor R. Preedy and published by Springer. This book was released on 2015-08-14 with total page 0 pages. Available in PDF, EPUB and Kindle. Book excerpt: In the past decade there has been a major sea change in the way disease is diagnosed and investigated due to the advent of high throughput technologies, such as microarrays, lab on a chip, proteomics, genomics, lipomics, metabolomics etc. These advances have enabled the discovery of new and novel markers of disease relating to autoimmune disorders, cancers, endocrine diseases, genetic disorders, sensory damage, intestinal diseases etc. In many instances these developments have gone hand in hand with the discovery of biomarkers elucidated via traditional or conventional methods, such as histopathology or clinical biochemistry. Together with microprocessor-based data analysis, advanced statistics and bioinformatics these markers have been used to identify individuals with active disease or pathology as well as those who are refractory or have distinguishing pathologies. New analytical methods that have been used to identify markers of disease and is suggested that there may be as many as 40 different platforms. Unfortunately techniques and methods have not been readily transferable to other disease states and sometimes diagnosis still relies on single analytes rather than a cohort of markers. There is thus a demand for a comprehensive and focused evidenced-based text and scientific literature that addresses these issues. Hence the formulation of Biomarkers in Disease. The series covers a wide number of areas including for example, nutrition, cancer, endocrinology, cardiology, addictions, immunology, birth defects, genetics and so on. The chapters are written by national or international experts and specialists.

Book Monoclonal Antibody Production

Download or read book Monoclonal Antibody Production written by National Research Council and published by National Academies Press. This book was released on 1999-05-06 with total page 74 pages. Available in PDF, EPUB and Kindle. Book excerpt: The American Anti-Vivisection Society (AAVS) petitioned the National Institutes of Health (NIH) on April 23, 1997, to prohibit the use of animals in the production of mAb. On September 18, 1997, NIH declined to prohibit the use of mice in mAb production, stating that "the ascites method of mAb production is scientifically appropriate for some research projects and cannot be replaced." On March 26, 1998, AAVS submitted a second petition, stating that "NIH failed to provide valid scientific reasons for not supporting a proposed ban." The office of the NIH director asked the National Research Council to conduct a study of methods of producing mAb. In response to that request, the Research Council appointed the Committee on Methods of Producing Monoclonal Antibodies, to act on behalf of the Institute for Laboratory Animal Research of the Commission on Life Sciences, to conduct the study. The 11 expert members of the committee had extensive experience in biomedical research, laboratory animal medicine, animal welfare, pain research, and patient advocacy (Appendix B). The committee was asked to determine whether there was a scientific necessity for the mouse ascites method; if so, whether the method caused pain or distress; and, if so, what could be done to minimize the pain or distress. The committee was also asked to comment on available in vitro methods; to suggest what acceptable scientific rationale, if any, there was for using the mouse ascites method; and to identify regulatory requirements for the continued use of the mouse ascites method. The committee held an open data-gathering meeting during which its members summarized data bearing on those questions. A 1-day workshop (Appendix A) was attended by 34 participants, 14 of whom made formal presentations. A second meeting was held to finalize the report. The present report was written on the basis of information in the literature and information presented at the meeting and the workshop.
