Book excerpt: Named for the enlarged, inclusion-bearing cells characteristic of infection by these viruses, cytomegaloviruses present a significant challenge to both microbiologist and immunologist. Although most primary infections in humans are subclinical, cytomegalovirus can be associated with a wide spectrum of disease, particularly when infection occurs in the immuno compromised individual or as a result of congenital or perinatal infection. Although reinfection with cytomegalovirus has been demonstrated, most recurrent and persistent infections result from the reactivation of latent virus. Cytomegaloviruses, like other members of the Herpesviridae family, have the capacity to establish latency after a primary infection but the mechanisms for establishing the nonreplicating but reactivat able state have not been defined. The factors responsible for the spectrum of manifestations of cytomegalovirus infection are largely undetermined but host immunological function, route of infection, and size of inoculum all contribute to the extent and severity of disease. Cytomegaloviruses have the largest genomes in the herpes virus family, approximately 240 kilo base pairs, providing a potential coding capacity for more than 200 proteins of which less than one-fourth have been mapped and described. There are many similarities to other herpes viruses in genome structure and gene expression; for example, three temporal classes of genes can be identified as rx (immediate-early), f3 (early), and y (late) products. The first five chapters of this volume review and describe recent developments in understanding the trans cription and regulation of these gene classes.

Book excerpt: Lumpy skin disease (LSD) is a vector-borne disease of cattle and Asian water buffalo that is included on the OIE (World Organisation for Animal Health) list of notifiable diseases. In July 2019 LSD was introduced to Bangladesh, China and India and then spread to Nepal and Bhutan and in 2020 to various provinces of China and India. A qualitative risk assessment was conducted to assess the likelihood of introduction and/or spread of LSD in 23 countries in South, East and Southeast Asia based on information available up to 31 October 2020. The economic impact of LSD for South, East and Southeast countries was estimated to be up to USD 1.45 billion in direct losses of livestock and production. These losses may be higher, due to the severe trade implications for infected countries. This document provides an overview of LSD control approaches, including prevention. The cost-effectiveness estimation demonstrates a strong economic justification for vaccination and advocates for a regional approach to harmonize control measures.

Introduction Dengue fever is a mosquito-transmitted disease of man which has afflicted untold millions of people over the past two centuries. It is caused by viruses classified as a subgroup of the group B togaviruses. Along with other members of that group as well as group A, the dengue viruses have been investigated intensively during recent years. Certain unique aspects of their structure, composition, antigenicity, replication, and antigenic relationships have established the togavirus family as quite distinct from other families of enveloped RNA viruses (see recent review of PFEFFERKORN and SHAPIRO, 1974). The basic studies leading to this conclusion have coincided with epidemiological field investigations which have resulted in a continuing increase in the number of viruses now designated as group A or B togaviruses. This, in turn, has led to a growing appreciation of their immense importance as actual or potential pathogens of man and beast.

Book excerpt: The Geographical Distribution of Animal Viral Diseases attempts to shed some light on the global distribution of 110 different viral diseases, mainly of livestock and companion animals. The world literature was screened for 110 different viruses, and maps were prepared. These maps delineate the global distribution of pathogenic viruses based on authenticated reports from a variety of reliable sources. Four viruses were categorized as affecting more than one species to a significant degree (astrovirus, rabies, rotaviruses, and Rift Valley fever). The largest number of maps involved viruses that affect humans. Of the 28 viruses a large number were from the California encephalitis group. Ten of the 28 viruses were reported only in the Eastern Hemisphere, 14 only in the Western Hemisphere, and four were worldwide. Birds were the next most frequently affected group with the 15 viruses, followed by pigs with 14 viruses. Overall the vector-borne viruses appear to have much sharper and clear-cut geographical boundaries than the others.
