For many tumors, this complex network of cytokine and immune interactions represent a formidable means of escape from immune surveillance. These cytokine networks are particularly important in pancreatic ductal adenocarcinoma (PDA), where a prominent infiltration of immunosuppressive immune populations could be found. Myeloid-derived suppressor cells (MDSCs) have previously been shown to be potent suppressors of anti-tumor immunity in PDA, but the cytokine networks regulating their recruitment to the tumor microenvironment remain incompletely understood. Here, I found that CXCR2 ligand expression is specifically correlated with enrichment of the granulocytic subset of MDSCs (G-MDSCs) in human PDAs. Using a genetically engineered mouse model of PDA, I showed that CXCR2 is required for G-MDSC trafficking to the tumor microenvironment, but not necessary for their systemic differentiation and expansion. The specific lack of G-MDSCs in the tumor microenvironment led to a T cell-dependent inhibition of tumor growth. Expression of CXCR2 ligands in PDA tumor cells can be potently induced by NF-?B activation. These findings describe a cytokine network in PDA where inflammatory signals in the tumor microenvironment drive the expression of CXCR2 ligands and the recruitment of immunosuppressive G-MDSCs. To discover other potentially important cytokine networks, I developed a novel analysis pipeline to reconstruct and compare cytokine networks from whole tumor gene expression data. Using expression of cytolytic genes as a gauge for anti-tumor immune activity, I found that PDA patients with high cytolytic activity have a slight survival advantage compared to those with lower activity. While macrophages were the most influential in tumors with low cytolytic activity, tumors with high cytolytic activity were characterized by increased activity of NK cells, recruitment of B cells, and increased importance of CD8 T cells, CD4 T helper cells, and B cells, among others. I further highlighted the cytokines that might be associated with these immune populations. Therefore, my analysis identified potentially important components of the cytokine network associated with high and low cytolytic activity. Collectively, the work in this thesis suggests that cytokine networks are crucial for maintaining an immunosuppressive microenvironment in cancer. Furthermore, disrupting key components of these networks can tip the balance in favor of cancer immunosurveillance.

CRC is not only a genetic disease with several mutations in specific oncogenes and/or tumor suppressor genes such as APC, KRAS, PIC3CA, BRAF, SMAD4 or TP53 but also a multifactorial disease including environmental factors. Cancer cells communicate with their environment mostly via soluble factors such as cytokines, chemokines or growth factors to generate a favorable tumor microenvironment (TME). The TME, a heterogeneous population of differentiated and progenitor cells, plays a critical role in regulating tumor development, growth, invasion, metastasis and therapy resistance. In this context, cytokines from cancer cells and cells of the TME influence each other, eliciting an inflammatory milieu that can either enhance or suppress tumor growth and metastasis. Additionally, several lines of evidence exist that the composition of the microbiota regulates inflammatory processes, controlled by cytokine secretion, that play a role in carcinogenesis and tumor progression. In this review, we discuss the cytokine networks between cancer cells and the TME and microbiome in colorectal cancer and the related treatment strategies, with the goal to discuss cytokine-mediated strategies that could overcome the common therapeutic resistance of CRC tumors

The mechanisms of escape are multiple and diverse and do not necessarily require the loss by tumor cells of expression of antigens recognized by the immune system. More commonly, tumor-specific T cells are rendered dysfunctional by a number of soluble and cell surface-bound molecules that are produced or modified by the action of cancer cells or suppressive and regulatory immune cells recruited to the tumor. Overall, the obstacles that prevent tumor rejection are active at several levels, affecting T-cell priming, functional differentiation, recruitment to the tumor, survival and effectors’ function inside the tumor. The multifaceted nature of these immunosuppressive networks represents a formidable obstacle to the success of immunotherapy. Nevertheless, the recent achievements of therapeutics targeting key checkpoint receptors highlight the great potential of strategies that are based on selective disruption of immunosuppressive networks.

Book excerpt: A link between inflammation and cancer has been established many years ago, yet it is only recently that the potential significance of this connection has become apparent. Although several examples of chronic inflammatory conditions, often induced by persistent irritation and/or infection, developing into cancer have been known for some time, there has been a notable resistance to contemplate the possibility that this association may apply in a causative way to other cancers. Examples for such progression from chronic inflammation to cancer are colon carcinoma developing with increased frequency in patients with ulcerative colitis, and the increased incidence of bladder cancer in patients suffering from chronic Schistosoma infection. Inflammation and cancer have been recognized to be linked in another context for many years, i.e., with regards to pathologies resembling chronic lacerations or 'wounds that do not heal.' More recently, the immunology of wound healing has given us clues as to the mechanistic link between inflammation and cancer, in as much as wounds and chronic inflammation turn off local cell-mediated immune responses and switch on growth factor release as well the growth of new blood vessels - angiogenesis. Both of these are features of most types of tumours, which suggest that tumours may require an immunologically shielded milieu and a growth factor-rich environment.
