Book Methods in Plant Electron Microscopy and Cytochemistry

Download or read book Methods in Plant Electron Microscopy and Cytochemistry written by William V. Dashek and published by Springer Science & Business Media. This book was released on 2000-06-29 with total page 297 pages. Available in PDF, EPUB and Kindle. Book excerpt: Hands-on experimentalists describe the cutting-edge microscopical methods needed for the effective study of plant cell biology today. These powerful techniques, all described in great detail to ensure successful experimental results, range from light microscope cytochemistry, autoradiography, and immunocytochemistry, to recent developments in fluorescence, confocal, and dark-field microscopies. Important advances in both conventional and scanning electron microscopies are also fully developed, together with such state-of-the-art ancillary techniques as high-resolution autoradiography, immunoelectron microscopy, X-ray microanalysis, and electron systems imaging. Easy-to-use and up-to-date, Methods in Plant Electron Microscopy and Cytochemistry offers today's plant scientists a first class collection of readily reproducible light and electron microscopical methods that will prove the new standard for all working in the field.

Book Autoradiography and Immunocytochemistry

Download or read book Autoradiography and Immunocytochemistry written by M.A. Williams and published by Elsevier. This book was released on 2013-10-22 with total page 238 pages. Available in PDF, EPUB and Kindle. Book excerpt: Autoradiography and Immunocytochemistry sets out in detail the preparation of autoradiographs and methods of preparing and use of immunological staining reagents. This book begins with an introduction to the demand for autoradiography and immunocytochemistry and their most advantageous use, followed by a discussion on the immunocytochemistry at the electron microscopical (EM) level in relation to the preparation of labeled antibodies. Other topics include the general remarks on “staining with antibodies; applying Fer-Ab conjugates to ultrathin sections; and controls in immunoferritin staining. The multistep antibody staining methods with unmodified proteins; control observations in staining experiments using lectins; and pattern analysis in EM immunocytochemistry are also covered. This text concludes with a description of the preparation of electron microscope autoradiographs, including the radioisotopes in EM autoradiography, preparation of radioactively-labeled tissues and cells, and methods of applying nuclear emulsions. This publication serves as a detailed laboratory guide for researchers and workers to successfully conduct electron microscope techniques.

Download or read book Biological Electron Microscopy written by Michael J. Dykstra and published by Springer Science & Business Media. This book was released on 2011-06-28 with total page 537 pages. Available in PDF, EPUB and Kindle. Book excerpt: Electron microscopy is frequently portrayed as a discipline that stands alone, separated from molecular biology, light microscopy, physiology, and biochemistry, among other disciplines. It is also presented as a technically demanding discipline operating largely in the sphere of "black boxes" and governed by many absolute laws of procedure. At the introductory level, this portrayal does the discipline and the student a disservice. The instrumentation we use is complex, but ultimately understandable and, more importantly, repairable. The procedures we employ for preparing tissues and cells are not totally understood, but enough information is available to allow investigators to make reasonable choices concerning the best techniques to apply to their parti cular problems. There are countless specialized techniques in the field of electron and light microscopy that require the acquisition of specialized knowledge, particularly for interpretation of results (electron tomography and energy dispersive spectroscopy immediately come to mind), but most laboratories possessing the equipment to effect these approaches have specialists to help the casual user. The advent of computer operated electron microscopes has also broadened access to these instruments, allowing users with little technical knowledge about electron microscope design to quickly become operators. This has been a welcome advance, because earlier instru ments required a level of knowledge about electron optics and vacuum systems to produce optimal photographs and to avoid "crashing" the instruments that typically made it difficult for beginners.

Book Electron Microscopy of Plant Cells

Download or read book Electron Microscopy of Plant Cells written by C Hawes and published by Academic Press. This book was released on 2012-12-02 with total page 477 pages. Available in PDF, EPUB and Kindle. Book excerpt: Electron Microscopy of Plant Cells serves as manual or reference of major modern techniques used to prepare plant material for transmission and scanning electron microscopy. There have been other books that generally discuss electron microscope methodology. This book focuses on problem areas encountered through the presence of tough cell walls and large central vacuole. It details preparative techniques for botanical specimens. Each of the nine chapters of this book covers the basic principles, useful applications, and reliable procedures used on the method of electron microscopy. Other topics discussed in each chapter include the general preparation and straining of thin sections, quantitative morphological analysis, and enzyme cytochemistry. This book also explains the immunogold labelling, rapid-freezing methods, and ambient- and low-temperature scanning electron microscopy among others. This book will be invaluable to general scientists, biologists, botanists, and students specializing in plant anatomy.
