Book excerpt: This thesis addresses data-driven analysis as well as prediction-based design of aperiodic sampling in control systems. In many modern control applications, the traditional view of sampling and controlling in a periodic fashion must be abandoned due to varying transmission delays, data loss or uneven computational latencies. The effect of these undesired aperiodicities on the closed loop must thus be analyzed. In addition, a targeted aperiodic design of the sampling pattern, e.g. via event-triggered control (ETC), often allows for a better usage of bandwidth than periodic concepts. We consider both perspectives in this thesis. In particular, first we provide methods to analyze stability of control systems under arbitrary aperiodic sampling directly from recorded data. Previous approaches to this end required model knowledge, which might be challenging to obtain. Second, we propose a prediction-based approach for design of the aperiodic sampling pattern, called rollout ETC. This approach allows for predictability of the required communication resources and a guarantee of improved sample efficiency, which were both difficult to address in existing ETC schemes.

Book excerpt: Antimicrobial resistance (AMR) has emerged as a major public health issue, attracting increased attention from governments and researchers. The spread and transmission of antimicrobial resistant bacteria/genes is not only a clinical problem, but also poses unprecedented challenges in the animal husbandry industry and is closely linked to the environment. The clinically important antimicrobial resistant bacteria and their AMR genes, such as carbapenemase-producing Gram-negative Bacilli with blaNDM, blaKPC, blaIMP, blaVIM genes, colistin-resistant Gram-negative bacteria with mcr genes, methicillin-resistant Staphylococcus aureus with mec genes, vancomycin-resistant Enterococcus with van genes, and tigecycline-resistant bacteria with tet(X) genes have been detected not only in health-care facilities but also in farms and environments. The concept of “One Health” emphasizes the interconnectedness of human, animal, and ecosystem health, with antimicrobial-resistant bacteria/genes occupying a unique position in close contact with all aspects.

Given the critical and interdependent nature of AMR in public health, animals and the environment, it is logical and necessary to take a "One Health" approach to address this issue. Clinically important antimicrobial resistant bacteria and genes have been reported and shown to be disseminated in humans, animals and the environment over the last few decades. The majority of these studies, however, only described resistant bacteria or genes from a single source and region, with no systematic or global conceptualization. The research on the transmission of AMR bacteria/genes has been limited and insufficient. A growing number of AMR-related proposals and policies have emerged, but few AMR control policy studies have been reported due to a lack of understanding of each participant's perspectives, values and goals. In this context, studies on the prevalence, transmission and control of clinically important AMR within the "One Health" framework are in line with demand.

Book excerpt: The performance of model predictive control (MPC) largely depends on the accuracy of the prediction model and of the constraints the system is subject to. However, obtaining an accurate knowledge of these elements might be expensive in terms of money and resources, if at all possible. In this thesis, we develop novel learning-based MPC frameworks that actively incentivize learning of the underlying system dynamics and of the constraints, while ensuring recursive feasibility, constraint satisfaction, and performance bounds for the closed-loop. In the first part, we focus on the case of inaccurate models, and analyze learning-based MPC schemes that include, in addition to the primary cost, a learning cost that aims at generating informative data by inducing excitation in the system. In particular, we first propose a nonlinear MPC framework that ensures desired performance bounds for the resulting closed-loop, and then we focus on linear systems subject to uncertain parameters and noisy output measurements. In order to ensure that the desired learning phase occurs in closed-loop operations, we then propose an MPC framework that is able to guarantee closed-loop learning of the controlled system. In the last part of the thesis, we investigate the scenario where the system is known but evolves in a partially unknown environment. In such a setup, we focus on a learning-based MPC scheme that incentivizes safe exploration if and only if this might yield to a performance improvement.

Book excerpt: Fruits and vegetables are an important part of a healthy diet and are protective against many chronic health conditions. Yet, fresh fruits and vegetables have been consistently implicated in food safety incidents involving microbiological hazards around the globe for decades. In response to requests of the Codex Committee on Food Hygiene concerning microbiological hazards in fresh fruits and vegetables and to update and expand the information available in Microbiological hazards in fresh leafy vegetables and herbs (MRA14), which was published in 2008, FAO and WHO convened a series of expert meetings in 2021 to 2022. The purpose of the meetings was to collect, review and discuss relevant measures to control microbiological hazards from primary production to point of sale in fresh, ready-to-eat (RTE) and minimally processed fruits and vegetables, including leafy vegetables. The experts made an effort to update and include any recent trends in commodity and pathogen pairing or pathogen occurrence and presence with a focus on emerging and neglected pathogens. The primary production in open fields was investigated by considering the location, adjacent land use, topography, and climate; prior land use; water; wildlife, animal and human intrusion; soil amendments; and harvest and packing. The experts also worked on: primary production in protected facilities; minimal processing; transport, distribution, and point of sale; and also the gaps in mitigation and interventions measures. The advice herein is useful for both risk assessors and risk managers, at national and international levels and those in the food industry working to control the relevant hazards in the fresh fruits and vegetables. the development of improved mitigation and intervention measures.
