Available in PDF, EPUB and Kindle. Book excerpt: Treatment of Parkinson's Disease (PD) via deep brain stimulation (DBS) has gained much attention in recent years. The treatment uses a surgically implanted electrode to excite selected areas of a patient's brain with electronic stimulation, often yielding drastic improvements in PD symptoms. The method of stimulation is a simple one; the patient's subthalamic nucleus (STN) or internal segment of the globus pallidus (GPi) is subjected to a constant barrage of high frequency pulses, warranting continuous spiking activity of the area. Until this point, no other method of stimulation has been shown to work in practice. This work examines the possibility of using a less constant, more energy-efficient stimulation pattern, wherein the stimulation waveform is not simply pre-programmed for all time; it is calculated continuously through use of model-based feedback control. Specifically, a computational model of a neuronal network that plays a key role in the emergence of PD symptoms is employed to simulate the activity of the brain under the influence of feedback-controlled stimulation. Two control strategies are developed, and the results of each are presented and discussed. Important aspects pertaining to general simulation-based neuronal control that arise throughout the research are also commented on.

Available in PDF, EPUB and Kindle. Book excerpt: The treatment of Parkinson's disease with electrical stimulation has become common with the application of deep brain stimulation (DBS). This method of applying an open-loop, high frequency pulsed signal to the patients subthalamic nucleus (STN) or global pallidus interior (GPi) succeeds in improving thalamic response to sensorimotor inputs. However, it is a method that can be further improved. One way that this may be possible is through the use of optogenetics. With the application light-sensitive channelrhodopsin-2 (ChR2), an ionic current can be generated by controlled illumination of the neuron.This thesis first analyzes potential computational neuronal models that can be used to successfully design a controller that would improve on current techniques. An analytic solution for the reduced order neuron model is found via an AFT solution technique to verify the numerical simulation of the model and further analyze the dynamics of the system. An optogenetics model is then acquired and observed in detail in order to apply it to the neuron model. Finally, multiple control strategies, including PID, linear compensator application, and pulsed stimulation, are explored and discussed in an effort to alleviate Parkinson's symptoms.

Book excerpt: Millions of people worldwide are affected by neurological disorders which disrupt the connections within the brain and between brain and body causing impairments of primary functions and paralysis. Such a number is likely to increase in the next years and current assistive technology is yet limited. A possible response to such disabilities, offered by the neuroscience community, is given by Brain-Machine Interfaces (BMIs) and neuroprostheses. The latter field of research is highly multidisciplinary, since it involves very different and disperse scientific communities, making it fundamental to create connections and to join research efforts. Indeed, the design and development of neuroprosthetic devices span/involve different research topics such as: interfacing of neural systems at different levels of architectural complexity (from in vitro neuronal ensembles to human brain), bio-artificial interfaces for stimulation (e.g. micro-stimulation, DBS: Deep Brain Stimulation) and recording (e.g. EMG: Electromyography, EEG: Electroencephalography, LFP: Local Field Potential), innovative signal processing tools for coding and decoding of neural activity, biomimetic artificial Spiking Neural Networks (SNN) and neural network modeling. In order to develop functional communication with the nervous system and to create a new generation of neuroprostheses, the study of closed-loop systems is mandatory. It has been widely recognized that closed-loop neuroprosthetic systems achieve more favorable outcomes for users then equivalent open-loop devices. Improvements in task performance, usability, and embodiment have all been reported in systems utilizing some form of feedback. The bi-directional communication between living neurons and artificial devices is the main final goal of those studies. However, closed-loop systems are still uncommon in the literature, mostly due to requirement of multidisciplinary effort. Therefore, through eBook on closed-loop systems for next-generation neuroprostheses, we encourage an active discussion among neurobiologists, electrophysiologists, bioengineers, computational neuroscientists and neuromorphic engineers. This eBook aims to facilitate this process by ordering the 25 contributions of this research in which we highlighted in three different parts: (A) Optimization of different blocks composing the closed-loop system, (B) Systems for neuromodulation based on DBS, EMG and SNN and (C) Closed-loop BMIs for rehabilitation.

Book excerpt: In the first part of this thesis, motivated by the development of deep brain stimulation for Parkinson's disease, we consider the problem of reducing the synchrony of a neuronal population via a closed-loop electrical stimulation. This, under the constraints that only the mean membrane voltage of the ensemble is measured and that only one stimulation signal is available (mean-field feedback). The neuronal population is modeled as a network of interconnected Landau-Stuart oscillators controlled by a linear single-input single-output feedback device. Based on the associated phase dynamics, we analyze existence and robustness of phase-locked solutions, modeling the pathological state, and derive necessary conditions for an effective desynchronization via mean-field feedback. Sufficient conditions are then derived for two control objectives: neuronal inhibition and desynchronization. Our analysis suggests that, depending on the strength of feedback gain, a proportional mean-field feedback can either block the collective oscillation (neuronal inhibition) or desynchronize the ensemble.In the second part, we explore two possible ways to analyze related problems on more biologically sound models. In the first, the neuronal population is modeled as the interconnection of nonlinear input-output operators and neuronal synchronization is analyzed within a recently developed input-output approach. In the second, excitability and synchronizability properties of neurons are analyzed via the underlying bifurcations. Based on the theory of normal forms, a novel reduced model is derived to capture the behavior of a large class of neurons remaining unexplained in other existing reduced models.
