The 1994 meeting was the first in this series devoted to the cytoskeleton. This complex system enables the eukaryotic cell to form discrete contacts with neighboring cells and the extracellular matrix, to differentiate, to move, change shape, transport organelles, and proliferate. These diverse tasks are performed by three distinct fibrillar networks: microfilaments, microtubules, and intermediate filaments, which are composed of structural and regulatory elements. The precise interplay between the components in time and space determines which of the various functions is performed. The rapid progress made in this field is best exemplified by the recent unraveling of the molecular mechanism of intracellular movement. Here, the modem microscopies of today allow the motility to be visualized in realtime. Molecular biology has dissected the functional domains of the motor proteins involved and provided material required for biochemical studies as well as structure analysis by X-ray diffraction and NMR. Thus, it is now possible to integrate cellular behavior and molecular structure in a unifying picture. Other recent advances in the field point to the role of cytoskeletol proteins in human diseases, from allergies to skin blistering, atrophies, and Alzheimers's disease. We have tried to bring together experts from different fields of research who are studying natural processes by which DNA molecules from differ ent sources are linked. It has been known for a long time that such linkage occurs between the chromosomes of bacteriophages and plasmids on the one hand and the chromosome of the bacterial host on the other. This process has been especially well studied in bacteriophage A. Since it is controlled in a complicated way, we began with a lecture by M. ptashne on these regulatory processes. H. Nash described the inte gration of bacteriophage A into the bacterial chromosome. To put this site-specific process into perspective, G. Mosig lectured on genetic recombination in prokaryotes in general and K. Murray described the use of bacteriophage A as an artificial vector for genetic engineering. A different kind of bacteriophage integration is shown by bacteriophage Mu, which is much less specific in its choice of an integration site than A. The properties of this phage were described by P. van de Putte.

Book excerpt: Eukaryotic cells contain a plurality of organelles distinguished by their specific membranes and contents. Their biogenesis occurs by growth and division of preexisting structures rather than de novo. Mitochondria and chloroplasts, which appear to be descended from prokaryotic ancestors, have retained some DNA and the biosynthetic capability for its expression. They synthesize, however, only a few of their proteins themselves. Most of their proteins are synthesized on free ribosomes in the cytoplasm and are only assembled in the correct membrane after synthesis is complete. The biogenesis of peroxisomes and glyoxysomes also appears to occur by an incorporation of proteins synthesized first in the cytoplasm. Other organelles, the Golgi complex, lysosomes, secretory vesicles, and the plasma membrane, are formed in a different manner. Their proteins are assembled in the membrane of the endoplasmic reticulum during trans lation by bound ribosomes and they must then be transported to the correct membrane. The 1980 Mosbach Colloquium was one of the first attempts to discuss the biogenesis of the various organelles in biochemical terms. This was appropriate since the crucial problems now center on the search for signals and receptors that dictate the site of assembly, the route taken, and the final location of a particular organelle protein. The assembly of prokaryotic membranes and the membrane of an animal virus were also discussed, since these simpler systems might shed light on the biogenesis of organelles in eukaryotes.
