Book Identification and Cloning a Novel Tumor Suppressor Gene in Breast Cancer

Download or read book Identification and Cloning a Novel Tumor Suppressor Gene in Breast Cancer written by and published by . This book was released on 1998 with total page 12 pages. Available in PDF, EPUB and Kindle. Book excerpt: On the basis of an extensive body of cytogenetic literature as well as molecular evidence from our laboratory, we hypothesize that a tumor suppressor gene is located on the long arm of Human Chromosome 7 at the q3 1.1 band. Furthermore, we propose that inactivation of this tumor suppressor gene plays a role in the development of breast cancer. The long term objectives of the study are to identify and clone the tumor suppressor gene and to determine its function. The objectives for the requested funding period are to provide functional evidence for the existence of this tumor suppressor gene in breast cancer using microcell fusion and to clone the DNA fragment containing the putative tumor suppressor gene using a novel approach based on the introduction of Yeast Artificial Chromosomes (YACs) into breast cancer cells. This novel approach will facilitate the cloning of putative tumor suppressor genes closing the technical gaps that exist between the identification of deleted regions of genome containing putative tumor suppressor genes and the actual cloning of DNA fragments containing the candidate genes.

Book Novel Oncogenes in Breast Cancer Development

Download or read book Novel Oncogenes in Breast Cancer Development written by and published by . This book was released on 2001 with total page 9 pages. Available in PDF, EPUB and Kindle. Book excerpt: Breast cancer is a multi-step genetic process involving mutations of oncogenes and tumor suppressor genes With a few notable exceptions the genetic determinants that contribute to the development of breast cancer remain unknown We have developed and applied a novel retrovirus-based library screening strategy coupled to a biological assay for growth transformation, to identify novel oncogenes in breast cancer development The approach involves the isolation of mRNA from noninvasive (MCF-7) and invasive (BT549 and Hs578T) human breast carcinoma cell lines to generate full length cDNA sequences, and introduced into a retrovirus expression vector The retrovirus, encoding genes expressed in breast cancers, is infected into Rat-l fibroblasts or RIE-l rat intestinal epithelial cells and screened for the appearance of foci of transformed cells The resulting transformed cells are isolated and the retrovirus-associated cDNA is rescued After sequence analyses, and verification of transforming activity of the isolated sequences, further analyses are done to assess the contribution of the isolate% genes to breast cancer development From these screens, we have identified the FGF receptor 2 and the Raf-l serine/threonine kinase.

Book Cancer Cell Structures Carcinogens and Genomic Instability

Download or read book Cancer Cell Structures Carcinogens and Genomic Instability written by Leon P. Bignold and published by Springer Science & Business Media. This book was released on 2006 with total page 396 pages. Available in PDF, EPUB and Kindle. Book excerpt: Tumors can be induced by a variety of physical and chemical carcinogens. The resulting tumor cells are usually abnormal in their morphology and behavior and transmit their abnormalities to their daughter tumor cells. Most theories of the pathogenesis of tumors suggest that carcinogens in some way cause alterations either of the genomes or of inheritable patterns of gene expression in normal cells, which then cause morphological and behavioral changes. This volume presents a collection of articles aimed at the question by what genetic or epigenetic mechanisms carcinogens can cause morphological abnormalities of tumor cells. It includes reviews of cellular targets of known carcinogens, and presents varying viewpoints of how morphological abnormalities and the actions of carcinogens might be related. The volume will be of interest to all those who are involved in cancer research or in the prevention, diagnosis or management of tumors in humans or animals.

Book RNA Binding Proteins as Novel Oncogenes and Tumor Suppressors in Breast Cancer

Download or read book RNA Binding Proteins as Novel Oncogenes and Tumor Suppressors in Breast Cancer written by and published by . This book was released on 2005 with total page 9 pages. Available in PDF, EPUB and Kindle. Book excerpt: The level of an mRNA depends upon its relative rates of synthesis and degradation. This is particularly important for oncoproteins and cell cycle proteins because their sustained synthesis favors cell growth rather than differentiation, a hallmark of the neoplastic phenotype. This control is exerted via a balance between the action of at least two RNA-binding proteins, AUF1 and HuR. AUF1 targets the degradation of mRNAs like the c-myc proto-oncogene and the cell cycle regulator cyclin D1. By contrast, HuR promotes stabilization of mRNAs. c-myc and cyclin D1 are particularly important, since both play causative roles in mammary tumorigenesis. Phase I of this work is to examine the effects of AUF1 and HuR expression levels on global gene expression in human breast carcinoma cells. Phase II is to assess the roles of AUF1 and HuR in cellular proliferation and tumorigenesis in vivo. During this funding period, we continued Phase I by transfecting expression vectors for inducible overexpression or knocked-down expression of AUF1 or HuR in human breast carcinoma cells and selecting a panel of clones. We also began identification of transcripts that are binding targets of AUF1 using mRNP immunoprecipitation (RIP) and cDNA microarray analyses of purified mRNAs.

Book Her 2

    Book Details:
  • Author : Robert Bazell
  • Publisher : Random House
  • Release : 2011-04-27
  • ISBN : 0307764982
  • Pages : 241 pages

Download or read book Her 2 written by Robert Bazell and published by Random House. This book was released on 2011-04-27 with total page 241 pages. Available in PDF, EPUB and Kindle. Book excerpt: Two years after she underwent a mastectomy and chemotherapy, Barbara Bradfield's aggressive breast cancer had recurred and spread to her lungs. The outlook was grim. Then she took part in Genentech's clinical trials for a new drug. Five years later she remains cancer-free. Her-2 is the biography of Herceptin, the drug that provoked dramatic responses in Barbara Bradfield and other women in the trials and that offers promise for hundreds of thousands of breast cancer patients. Unlike chemotherapy or radiation, Herceptin has no disabling side effects. It works by inactivating Her-2/neu--a protein that makes cancer cells grow especially quickly-- produced by a gene found in 25 to 30 percent of all breast tumors. Herceptin caused some patients' cancers to disappear completely; in others, it slowed the progression of the disease and gave the women months or years they wouldn't otherwise have had. Herceptin is the first treatment targeted at a gene defect that gives rise to cancer. It marks the beginning of a new era of treatment for all kinds of cancers. Robert Bazell presents a riveting account of how Herceptin was born. Her-2 is a story of dramatic discoveries and strong personalities, showing the combination of scientific investigation, money, politics, ego, corporate decisions, patient activism, and luck involved in moving this groundbreaking drug from the lab to a patient's bedside. Bazell's deft portraits introduce us to the remarkable people instrumental in Herceptin's history, including Dr. Dennis Slamon, the driven UCLA oncologist who played the primary role in developing the treatment; Lily Tartikoff, wife of television executive Brandon Tartikoff, who tapped into Hollywood money and glamour to help fund Slamon's research; and Marti Nelson, who inspired the activists who lobbied for a "compassionate use" program that would allow women outside the clinical trials to have access to the limited supplies of Herceptin prior to FDA approval of the drug. And throughout there are the stories of the heroic women with advanced breast cancer who volunteered for the trials, risking what time they had left on an unproven treatment. Meticulously researched, written with clarity and compassion, Her-2 is masterly reporting on cutting-edge science.

Book Cancer as a Metabolic Disease

Download or read book Cancer as a Metabolic Disease written by Thomas Seyfried and published by John Wiley & Sons. This book was released on 2012-05-18 with total page 482 pages. Available in PDF, EPUB and Kindle. Book excerpt: The book addresses controversies related to the origins of cancer and provides solutions to cancer management and prevention. It expands upon Otto Warburg's well-known theory that all cancer is a disease of energy metabolism. However, Warburg did not link his theory to the "hallmarks of cancer" and thus his theory was discredited. This book aims to provide evidence, through case studies, that cancer is primarily a metabolic disease requring metabolic solutions for its management and prevention. Support for this position is derived from critical assessment of current cancer theories. Brain cancer case studies are presented as a proof of principle for metabolic solutions to disease management, but similarities are drawn to other types of cancer, including breast and colon, due to the same cellular mutations that they demonstrate.

Book Cancer Evolution

    Book Details:
  • Author : Charles Swanton
  • Publisher : Perspectives Cshl
  • Release : 2017
  • ISBN : 9781621821434
  • Pages : 350 pages

Download or read book Cancer Evolution written by Charles Swanton and published by Perspectives Cshl. This book was released on 2017 with total page 350 pages. Available in PDF, EPUB and Kindle. Book excerpt: Tumor progression is driven by mutations that confer growth advantages to different subpopulations of cancer cells. As a tumor grows, these subpopulations expand, accumulate new mutations, and are subjected to selective pressures from the environment, including anticancer interventions. This process, termed clonal evolution, can lead to the emergence of therapy-resistant tumors and poses a major challenge for cancer eradication efforts. Written and edited by experts in the field, this collection from Cold Spring Harbor Perspectives in Medicine examines cancer progression as an evolutionary process and explores how this way of looking at cancer may lead to more effective strategies for managing and treating it. The contributors review efforts to characterize the subclonal architecture and dynamics of tumors, understand the roles of chromosomal instability, driver mutations, and mutation order, and determine how cancer cells respond to selective pressures imposed by anticancer agents, immune cells, and other components of the tumor microenvironment. They compare cancer evolution to organismal evolution and describe how ecological theories and mathematical models are being used to understand the complex dynamics between a tumor and its microenvironment during cancer progression. The authors also discuss improved methods to monitor tumor evolution (e.g., liquid biopsies) and the development of more effective strategies for managing and treating cancers (e.g., immunotherapies). This volume will therefore serve as a vital reference for all cancer biologists as well as anyone seeking to improve clinical outcomes for patients with cancer.

Book Heat Shock Proteins in Cancer

    Book Details:
  • Author : Stuart K. Calderwood
  • Publisher : Springer Science & Business Media
  • Release : 2007-09-09
  • ISBN : 1402064012
  • Pages : 399 pages

Download or read book Heat Shock Proteins in Cancer written by Stuart K. Calderwood and published by Springer Science & Business Media. This book was released on 2007-09-09 with total page 399 pages. Available in PDF, EPUB and Kindle. Book excerpt: Heat shock proteins are emerging as important molecules in the development of cancer and as key targets in cancer therapy. These proteins enhance the growth of cancer cells and protect tumors from treatments such as drugs or surgery. However, new drugs have recently been developed particularly those targeting heat shock protein 90. As heat shock protein 90 functions to stabilize many of the oncogenes and growth promoting proteins in cancer cells, such drugs have broad specificity in many types of cancer cell and offer the possibility of evading the development of resistance through point mutation or use of compensatory pathways. Heat shock proteins have a further property that makes them tempting targets in cancer immunotherapy. These proteins have the ability to induce an inflammatory response when released in tumors and to carry tumor antigens to antigen presenting cells. They have thus become important components of anticancer vaccines. Overall, heat shock proteins are important new targets in molecular cancer therapy and can be approached in a number of contrasting approaches to therapy.
