Book excerpt: During the past several decades, much research effort has gone into the elucidation of the role of neuroendocrine systems as secretory and metabolic regulators of cells of a variety of organs and structures, including the testes, ovaries, adrenals, thyroid, pituitary gland, and mammary glands. However, the role of cells comprising such organs and structures in the modulation of neuroendocrine processes has received considerably less is generally less well appreciated. attention and Nonetheless, it is important that we understand the actions on neuroendocrine systems of substances that reach the brain by way of the vasculature, including hormones, cytokines, toxins, amino acids, drugs, and similar agents. In order to analyze the present state of knowledge on this topic, experimental scientists and clinicians, whose shared interests include actions of circulating agents on the brain, met at a satellite symposium of the XXXI International Congress of Physiological Sciences. This symposium, entitled Circulating Regulatory Factors and Neuroendocrine Function, was held in Smolenice Castle, Czechoslovakia, June 26-July 1, 1989, and reviews delivered at this symposium as invited presentations are published in this volume. Presentations given as free communications have been published separately and are available in Endocrinologia Experimentalis 24: 1-273, 1990.

Progress in this field has been evident on many fronts, with novel information rapidly accumulating in the literature regarding the chemistry and molecular biology of this enzyme and its role in many important physiological processes. These include cellular signal transduction via the G-protein cycle, and in the generation of many cellular mediators, such as the platelet activating factor (PAF) and the eicosanoids that participate in the initiation and propagation of inflammation, to mention a few. This symposium was organized to obtain an overview of current investigations on this enzyme from the standpoint of its chemistry, molecular biology and physiology. Another important focus of this symposium concerns the regulation of PLA2, including endogenous and synthetic inhibitors and activators of this enzyme. To review these important areas in PLA2 research we invited scientists who made significant contributions in this field. The papers in this volume are organized to emphasize the recent advances in several areas of investigation, including: (I) the structure and mechanism of action of PLA2, (2) mechanism of activation of PLA2, (3) molecular biology, physiology and endogenous inhibitors of this enzyme and finally, (4) clinical investigations emphasizing the pathophysiological role of this enzyme in human diseases. Book excerpt: Interest in interleukin-1 (IL-1) has increased dramatically over the last decade, but has been largely restricted to immunologists, cell biologists and those studying inflammation and cancer. However, it has recently been recognized that the brain directly controls or modulates many aspects of immune function, while molecules classically associated with the immune system, such as interleukin-1, are synthesised within the brain and act directly on the central nervous system to modify local and systemic functions. Thus, this topic is relatively new to neurobiologists, and this book is the first comprehensive description of current knowledge on interleukin-1 in the brain, including its location, synthesis and receptors, actions on behaviour, fever, metabolism, neuroendocrine function, electrical activity of the brain, nerve growth factor, and relationship to clinical indications. The book is organised into three sections. The first reviews the data available on the neural localisation of IL-1 and the nature of its central receptors. The main part of the book examines the different neural effects of IL-1 and the mechanisms which are involved in these actions, comparing IL-1 where possible to other inflammatory cytokines which also have neurotrophic effects. Book excerpt: The traditional concept of a neuroendocrine mechanism for regulation of growth hormone (GH) secretion is based in large part on the work of Roger Guillemin. The work of Dr. Guillemin, who was awarded the 1977 Nobel Prize in Physiology and Medicine, supported the view that quantita tive change in GH secretion was the net result of pituitary stimulation and inhibition by the hypothalamic neurohormones, GH releasing hormone (GHRH), and somatostatin (somatotropin release inhibiting factor; SRIF), respectively. During the 1970s, another endocrine research pioneer, Dr. Cyril Bowers, discovered that structural modification of enkephalin re sulted in a family of peptides with GH releasing properties. These com pounds, simply called GH releasing peptide (GHRP), were originally thought to mimic GHRH. However, upon subsequent investigation they were found to supplement the activity of the natural hormone through a different mechanism. Nearly two decades after their discovery, the differ ences between GHRP and GHRH have been described by many different laboratories throughout the world. The complementary GH secretagogues have different binding sites, second messengers, and effects on gene expres sion. Based on these differences, it has been suggested that expansion of the original two hormone mechanisms for GH regulation to include a third molecule may be appropriate, even though the naturally occurring ana logue of GHRP has not yet been identified. Despite our lack of knowledge concerning the natural product mimicked by GHRP, clinical development of the new family of GH secretagogues for diagnostic and therapeutic purposes has begun in earnest.

Vaslamatzis deals with a “Framework for a new dialogue between psychoanalysis and neurosciences”. In this chapter the author describes three points of epistemological congruence: firstly, dualism is no longer a satisfactory solution; secondly, cautions for the centrality of interpretation (hermeneutics); and, thirdly, the self-criticism of neuroscientists. David W.Mann in his contribution “The mirror crack’d: dissociation and reflexivity in self and group phenomena” tries to show how reflexive processes generate each of three levels of the human system (self, relationships, group) and integrate them one to another, while dissociative processes tend throughout to pull them apart. Health and illness within the self, the relationship and the group can be understood as special states of the dynamic equilibria between these cohesive and dispersive trends. In “Sleep, memory and plasticity” Matthew P. Walker and Robert Stickgold outline a review of the researches following the discovery of rapid eye movement (REM) and non-REM (NREM) sleep, and specifically of those that began testing the hypothesis that sleep, or even specific stages of sleep, actively participated in the process of memory development. The last two chapters, “Clinical implications of neuroscience research in PTSD” by Bessel A. Van Der Kolk, and “Dysregulation of the right brain: a fundamental mechanism of traumatic attachment and the psychopathogenesis of PTSD” by Allan N. Schore, demonstrate how the psychopathology of traumatic conditions can be a fertile field of dialogue between neuroscience and psychoanalysis.
