Book excerpt: Chromosomes, as the genetic vehicles, provide the basic material for a large proportion of genetic investigations, from the construction of gene maps and models of chromosome organization, to the inves tigation of gene function and dysfunction. The study of chromosomes has developed in parallel with other aspects of molecular genetics, beginning with the first preparations of chromosomes from animal cells, through the development of banding techniques, which permitted the unequivocal identification of each chromosome in a karyotype, to the present analytical methods of molecular cytogenetics. Although some of these techniques have been in use for many years, and can be learned relatively easily, most published scientific reports—as a result of pressure on space from editors, and the response to that pressure by authors—contain little in the way of technical detail, and thus are rarely adequate for a researcher hoping to find all the necessary information to embark on a method from scratch. A new user needs not only a detailed description of the methods, but also some help with problem solving and sorting out the difficulties en countered in handling any biological system. This was the require ment to which the series Methods in Molecular Biology is addressed, and Chromosome Analysis Protocols forms a part of this series.

Given the accelerated growth of knowledge in the field of eukaryotic gene expression, it seemed timely to hold a NATO Advanced Study Institute to discuss current developments in this area of biology and to evaluate the potential of emerging technologies such as gene transfer, recombinant DNA cloning and quantitative high resolution two-dimensional gel electrophoresis. The initial articles in t~is volume describe various differentiation models and address questions such as the relationships between differentiation and cell proliferation, biochemical changes accompanying differen tiation, expression of differentiated gene products and their regulation as well as gene organization of cytoskeletal proteins. The second section describes properties of neoplastic cells, surveys current assays for transformation and offers some new insights into the mechanisms involved in carcinogenesis. The third part is dedicated to viral oncogenesis and to the role of onco genes in cell transformation. Particular emphasis is given to the role of tyrosine kinases in cell transformation. The concluding section deals with various aspects of gene expression in normal and transformed cells with special emphasis given to studies using two dimensional gel electrophoresis, cell hybridization, gene transfer and immunological techniques.

Book excerpt: Developmental cancer products (oncodevelopmental markers, ODM) not only serve as diagnostic and prognostic indicators but also may be used to study the nature of the carcinogenic process and the biology of tumors. For many years oncologists have searched for markers of cancer cells that would permit unequivocal recognition of cancer in contrast to noncancerous tissue. The earliest and still most widely used method of identification of cancer tissue or cells is the structural resemblance of cancer tissue to fetal or immature tissue. Pathologists not only recognize cancer by its morphologic similarity to fetal tissues, but also in many instances can relate the behavior of a given tumor to the degree of tissue differentiation. Thus, poorly differentiated tumors that resemble fetal tissue generally grow more rapidly and metastasize earlier than do well-differentiated tumors that more closely resemble adult tissue. In recent years the commonality of fetal and cancer tissue has been extended to products of tumor cells that, can be analyzed by biochemical, immunological, or physiological techniques. Increas ingly, products of cancer cells similar to fetal products are being identified and studied. These products range from cell-surface markers (fetal or differentiation antigens), placental proteins, hormones, and isoenzymes to a multitude of products, such as carcinoembryonic antigen (CEA), alphafetoprotein (AFP), lymphocyte markers, and nucleic acids, such as tRNA, that are produced in small amounts by v vi PREFACE continually differentiating cells in the adult but in much greater amounts by tumors.
