When Marlowe was killed after being accused of blasphemy, he left the poem unfinished to be later completed by George Chapman. The poem follows the love story between the two Greek mythological characters Hero and Leander which was first narrated in the works of the two ancient poets Musaeus Grammaticus and Ovid. Hero is a priestess of the goddess Venus to whom she has made a vow of chastity, yet she falls in love with Leander, a young man from Abydos on the opposite side of the narrow sea passage known as the Hellespont. Leander, who promises to cross the Hellespont every night to reach his beloved, gets in trouble with Neptune, the god of the seas. When he eventually comes at Hero's door, he has to make use of his guile and physical beauty to seduce his coy beloved. At this point starts Chapman's contribution to accompany the two lovers until Leander drowns in the sea on his way to Hero. Watching her lover's dead body from her high tower, the latter decides to throw herself and die by his side.

At a festival in honor of her deity, Venus and Adonis, she is seen by Leander, a youth from Abydos on the opposite side of the Hellespont. Leander falls in love with her, and she reciprocates, although cautiously, as she has made a vow of chastity to Venus.

The poem was first published posthumously, five years after Marlowe's demise. Hero and Leander is the Greek myth relating the story of Hero, a priestess of Aphrodite who dwelt in a tower in Sestos on the European side of the Hellespont (today's Dardanelles), and Leander, a young man from Abydos on the opposite side of the strait. Leander fell in love with Hero and would swim every night across the Hellespont to be with her. Hero would light a lamp at the top of her tower to guide his way. Succumbing to Leander's soft words and to his argument that Venus, as the goddess of love, would scorn the worship of a virgin, Hero allowed him to make love to her. These trysts lasted through the warm summer. But one stormy winter night, the waves tossed Leander in the sea and the breezes blew out Hero's light; Leander lost his way and was drowned. When Hero saw his dead body, she threw herself over the edge of the tower to her death to be with him.
