Due to its wide availability and the ease of production, ricin has been exploited as an agent of bioterrorism and biological warfare and is classified as a level B threat. Tons of ricin are produced annually worldwide as a by-product of castor oil, leading to exposure. Currently, there is no proven safe treatment for ricin intoxication. Ricin consists of an active A subunit (RTA) covalently linked to a cell binding B subunit (RTB). RTA depurinates a universally conserved adenine in the sarcin–ricin loop (SRL) of the large rRNA and inhibits protein synthesis. RTA-antibody complexes have been explored as immunotoxins against cancer cells. The articles published in this book address the history of ricin research, how ricin enters cells and traffics to the ribosome, how it inactivates ribosomes, how it induces inflammatory signaling pathways and programmed cell death, the design of therapies against ricin intoxication, and innovative assays for ricin detection.

Book excerpt: Bacillus thuringiensis (Bt) has been used as a biopesticide in agriculture, forestry and mosquito control because of its advantages of specific toxicity against target insects, lack of polluting residues and safety to non-target organisms. The insecticidal properties of this bacterium are due to insecticidal proteins produced during sporulation. Despite these ecological benefits, the use of Bt biopesticides has lagged behind the synthetic chemicals. Genetic improvement of Bt natural strains, in particular Bt recombination, offers a promising means of improving efficacy and cost-effectiveness of Bt-based bioinsecticide products to develop new biotechnological applications. On the other hand, the different Bacillus species have important biotechnological applications; one of them is carried out by producing secondary metabolites, which are the study object of natural product chemistry. The amazing structural variability of these compounds has attracted the curiosity of chemists and the biological activities possessed by natural products have inspired the pharmaceutical industry to search for lead structures in microbial extracts. Screening of microbial extracts reveals the large structural diversity of natural compounds with broad biological activities, such as antimicrobial, antiviral, immunosuppressive, and antitumor activities that enable the bacterium to survive in its natural environment. These findings widen the target range of Bacillus spp., in special B. thuringiensis, besides insecticidal activity and help people to better understand its role in soil ecosystem.

RIPs catalyze the removal of an adenine residue from a conserved loop in the large ribosomal RNA. The adenine residue removed by this depurination is crucial for the binding of elongation factors. Ribosomes modified in this way are no longer able to carry out protein synthesis. Most RIPs exist as single polypeptides (Type 1 RIPs) which are largely non-toxic to mammalian cells because they are unable to enter them and thus cannot reach their ribosomal substrate. In some instances, however, the RIP forms part of a heterodimer where its partner polypeptide is a lectin (Type 2 RIPs). These heterodimeric RIPs are able to bind to and enter mammalian cells. Their ability to reach and modify ribosomes in target cells means these proteins are some of the most potently cytotoxic poisons found in nature, and are widely assumed to play a protective role as part of the host plant’s defenses. RIPs are able to further damage target cells by inducing apoptosis. In addition, certain plants produce lectins lacking an RIP component but which are also cytotoxic. This book focuses on the structure/function and some potential applications of these toxic plant proteins.

It provides a background of knowledge and current ideas in the field and gives ex amples of observations and their interpretations. I understand that some interpre tations and generalizations may be tentative or disputable, but I hope that this will stimulate thinking and discussing better than if I left white spots. The book has a prototype: it is my monograph "Ribosome Structure and Pro tein Biosynthesis" published by the Benjamin/Cummings Publishing Company, Menlo Park, California, in 1986. Here I have basically kept the former order of pre sentation ofthe topics and the subdivision into chapters. The contents ofthe chap ters, however, have been significantly revised and supplemented. The newly writ ten chapters on translational control in prokaryotes (Chapter 16) and eukaryotes (Chapter 17) are added.
