Book excerpt: Recent results from a genetic gain study have illustrated the contribution of plant breeding to the improvement in seed yield of soybean (Glycine max (L.) Merr.). The objective of this research was to characterize the changes in several physiological parameters that have occurred in the released cultivars with improvement of seed yield. Sixty maturity group III and 54 maturity group IV cultivars, released from the 1920's through 2010, were evaluated in dryland and irrigated environments at Manhattan, KS in 2010 and 2011. Genotypes were planted in four-row plots, 3.4 m long, spaced 76 cm apart, arranged in a randomized complete block design with four replications. Genotypes were evaluated for canopy temperature, leaf chlorophyll content, pollen germination, leaf chlorophyll fluorescence, leaf antioxidants, and yield components. Canopy temperature measurements were captured between 1000h and 1400h using an infrared camera multiple times from R1 continuing through R6. Leaf chlorophyll content was measured using a SPAD meter several times from R1 through R6. In vitro pollen germination was measured using incubation temperatures of 28 and 34°C, beginning at late R1 through the end of flowering. Leaf chlorophyll fluorescence was measured beginning at R1 through R6. Leaf antioxidants were analyzed for total antioxidant capacity and electrolyte leakage by collecting leaves from the top 3rd to 5th trifoliate at R4 and R6. Yield components were analyzed from a hand harvested 0.33 m section of one border row. Genotypes differed significantly for canopy temperature, leaf chlorophyll content, pollen germination, and yield components. No significant differences were found for leaf chlorophyll fluorescence or leaf antioxidants. Seed yield increased with year of release. Canopy temperature was negatively correlated and leaf chlorophyll content was positively correlated with year of release in both maturity groups. No significant correlation with year of release was found for in vitro pollen germination or electrolyte leakage. Leaf chlorophyll fluorescence, yield components, and total antioxidant capacity was positively correlated in the maturity group IV genotypes with year of release. Evaluation of these parameters may serve as a basis to select for seed yield, or to assess the abiotic stress tolerance of a genotype.

Soybean was domesticated in China, and since last 4-5 decades it has become one of the most widely grown crops around the globe. The crop is grown on an anticipated 6% of the world’s arable land, and since the 1970s, the area in soybean production has the highest percentage increase compared to any other major crop. It is a major crop in the United States, Brazil, China and Argentina and important in many other countries. The cultivated soybean has one wild annual relative, G. soja, and 23 wild perennial relatives. Soybean has spread to many Asian countries two to three thousand years ago, but was not known in the West until the 18th century. Among the various constraints responsible for decrease in soybean yields are the biotic and abiotic stresses which have recently increased as a result of changing climatic scenarios at global level. A lot of work has been done for cultivar development and germplasm enhancement through conventional plant breeding. This has resulted in development of numerous high yielding and climate resilient soybean varieties. Despite of this development, plant breeding is long-term by nature, resource dependent and climate dependent. Due to the advancement in genomics and phenomics, significant insights have been gained in the identification of genes for yield improvement, tolerance to biotic and abiotic stress and increased quality parameters in soybean. Molecular breeding has become routine and with the advent of next generation sequencing technologies resulting in SNP based molecular markers, soybean improvement has taken a new dimension and resulted in mapping of genes for various traits that include disease resistance, insect resistance, high oil content and improved yield. This book includes chapters from renowned potential soybean scientists to discuss the latest updates on soybean molecular and genetic perspectives to elucidate the complex mechanisms to develop biotic and abiotic stress resilience in soybean. Recent studies on the improvement of oil quality and yield in soybean have also been incorporated.

It fixes atmospheric nitrogen with the symbiotic relationship of rhizobia bacteria that inhabit root nodules. The global population is expected to exceed 9.7 billion by 2050, resulting in increased food demand, particularly for protein. In Canada, soybean is the third largest field crop in terms of farm cash receipts, and its production has recently grown significantly in the Canadian Prairies. The changing global climate is predicted to lead large areas of the world to experience extensive drought conditions over the next few decades. Drought will be a critical issue for western Canada as it has been experiencing frequent and severe droughts over the last few decades. Drought has negative impacts on soybean plant development, yield, and symbiotic nitrogen fixation. The main objective of this project is to identify the allelic variations associated with diverse short-season soybean varieties for plant physiological parameters, yield traits, and symbiotic nitrogen fixation under drought stress. A greenhouse pot experiment with 103 early-maturity Canadian soybean varieties was conducted to determine the above-mentioned traits. Seedlings were inoculated with Bradyrhizobium japonicum USDA 110, and the initial soil moisture content of the growing media was maintained at 80% field capacity (FC). Drought stress was applied after three weeks of plant growth, with half of the plants kept at 30% FC and the remaining at 80% FC until maturity. Plant physiological traits such as photosynthesis, transpiration, stomatal conductance, leaf chlorophyll, water-use efficiency, and intrinsic water use efficiency were collected during the flowering stage. At seed maturity, data on yield and nitrogen fixation-related traits, including number of pods, number of seeds, seed yield, 100-seed weight, seed nitrogen content, % nitrogen derived from the atmosphere (%Ndfa), total seed nitrogen fixed, and carbon isotope discrimination (CID), were collected. Drought stress reduced stomatal conductance and transpiration, resulting in greater water-use efficiency compared to well-watered plants at the flowering stage. In comparison to the well-watered treatment, drought-stressed soybean plants had lower seed yield, yield components (number of pods, number of seeds, 100-seed weight), seed nitrogen content, %Ndfa, and total fixed nitrogen. Specifically, drought reduced yield by 34.7%. %Ndfa by 13.4%, total seed nitrogen by 34.9%, and amount of seed nitrogen fixed by 42.1% compared to the well-watered treatment. Significant genotypic variability among soybean varieties was found for plant physiological parameters, yield parameters, nitrogen fixation traits, and water use efficiency. A Genome-Wide Association Study (GWAS) was conducted using 2.16M SNPs (2,164,465 SNPs) for above mentioned parameters for 30% FC, 80% FC and their relative performance (30% FC / 80% FC). In total, 13 quantitative trait locus (QTL) regions, including multiple candidate genes, were detected as significantly associated with different plant physiological traits, including photosynthesis, stomatal conductance, and water-use efficiency for 30% FC and relative performance. In terms of yield and nitrogen fixation-related characteristics, six QTL regions and candidate genes were identified as significantly correlated with %Ndfa and CID under drought stress and relative performance. These QTL regions may be useful in future breeding efforts to create drought-resistant soybean cultivars.
