Reversible protein phosphorylation is a research field pioneered and developed by Krebs and Fischer. This book contains short reviews and original research papers contributed by Krebs and Fischer's coworkers, both former and current. The contents reflect the two-way interaction between protein phosphorylation and other biomedical research fields. The chapters are grouped into four sections. The first two deal with structure/function aspects of protein kinases and protein mechanisms. Unlike many other research fields, which undergo periods of intense activity and productivity followed by relative calm, the protein phosphorylation field enjoyed continued growth both in scope and intensity, and the pace of this growth has increased markedly in recent years. The first meeting with the topic "Signal Trans duction and Protein Phosphorylation" was held on the island of Spezai, Greece, in September 1986. The second one took place in Chateau La Londe, France, in September 1989 on "Cellular Regulation by Protein Phosphorylation", the third one on " Tyrosine Phosphorylation/Dephosphorylation and Downstream Signaling" was in September 1992 in Maratea, Italy. The titles of these books clearly mirror the developments that have taken place in the last decade. Beginning with the recognition that protein phosphorylation is at the center of signaling -clearly established in 1990 -it became apparent that many cellular processes are regulated by this mode. A new focus then emerged when it was recognized that growth factors are bound to corresponding receptors trigger protein tyrosine phosphorylation which controls cell prolifera tion. This was the topic of the third meeting in this series. It is now evident that further progress depends on understanding the three dimensional structure of the proteins involved. It goes without saying, for example, that understanding the location of proteins by adaptor proteins is only possible on the basis of the three dimensional protein structure. Therefore, the fourth meeting in this series concentrated on the protein structure of signaling molecules as well as on the elucidation of the principles of protein domain interactions.

Then follow a few reviews dedicated to specific cell cycle regulators: the 14-3-3 protein (chapter 5), the cdc7/Dbf4 protein kinase (chapter 6), the two products of the pI6/CDKN2A locus and their link with Rb and p53 (chapter 7), the Ph085 cyclin-dependent kinases in yeast (chapter 9), the cdc25 phophatase (chapter 10), RCCI and ran (chapter 13). The intriguing phosphorylation dependent prolyl-isomerization process and its function in cell cycle regulation are reviewed in chapter 8.

One paper describes that the amino acid pool size and content are governed by the conditioning of intracellular proteins and the metabolic activity of the cell. The paper also suggests that intracellular pools do not regulate protein metabolism. The collection can prove beneficial for biochemists, micro-biologists, cellular researchers, and academicians involved in the study of cellular biology or physiology.

As this meeting attracted many investigators involved in cyclic nucleotides and calcium role in intracellular regulation, it was thought that this opportunity could be used to organize, prior to the Congress, an in-depth introductory course on the subject. This was carried out as a NATO Advanced Study Institute. The participants included Ph. D. students and M. D. s engaged in a research training, but also fully trained and well known researchers who wanted a refresher course on the whole subject. During the course, most of the participants and lecturers asked to be provided with a text summarizing the basic lectures of the course. This book was therefore conceived as a basic textbook on the regulation and action of intracellular signal molecules, concentrating mainly on cyclic nucleotides and calcium. It was deliberately kept at a basic level. We would therefore be happy if it could be used as an introduction for interested M. D. s or Ph. D. s working in other fields or entering this field and as a general refresher for researchers interested in the subject. For this reason, very general schemes have been asked of the authors, along with reading lists of available reviews rather than extensive bibliographies.
