Book ACTH Action in the Adrenal Cortex From Molecular Biology to Pathophysiology

Download or read book ACTH Action in the Adrenal Cortex From Molecular Biology to Pathophysiology written by Nicole Gallo-Payet and published by Frontiers Media SA. This book was released on 2017-07-27 with total page 421 pages. Available in PDF, EPUB and Kindle. Book excerpt: By stimulating adrenal gland and corticosteroid synthesis, the adrenocorticotropic hormone (ACTH) plays a central role in response to stress. In this Research Topic, a particular attention has been given to the recent developments on adrenocortical zonation; the growth-promoting activities of ACTH; the various steps involved in acute and chronic regulation of steroid secretion by ACTH, including the effect of ACTH on circadian rhythms of glucocorticoid secretion. The Research Topic also reviews progress and challenges surrounding the properties of ACTH binding to the MC2 receptor (MC2R), including the importance of melanocortin-2 receptor accessory protein (MRAP) in MC2R expression and function, the various intracellular signaling cascades, which involve not only protein kinase A, the key mediator of ACTH action, but also phosphatases, phosphodiesterases, ion channels and the cytoskeleton. The importance of the proteins involved in the cell detoxification is also considered, in particular the effect that ACTH has on protection against reactive oxygen species generated during steroidogenesis. The impact of the cellular microenvironment, including local production of ACTH is discussed, both as an important factor in the maintenance of homeostasis, but also in pathological situations, such as severe inflammation. Finally, the Research Topic reviews the role that the pituitary-adrenal axis may have in the development of metabolic disorders. In addition to mutations or alterations of expression of genes encoding components of the steroidogenesis and signaling pathways, chronic stress and sleep disturbance are both associated with hyperactivity of the adrenal gland. A resulting effect is increased glucocorticoid secretion inducing food intake and weight gain, which, in turn, leads to insulin and leptin resistance. These aspects are described in detail in this Research Topic by key investigators in the field. Many of the aspects addressed in this Research Topic still represent a stimulus for future studies, their outcome aimed at providing evidence of the central position occupied by the adrenal cortex in many metabolic functions when its homeostasis is disrupted. An in-depth investigation of the mechanisms underlying these pathways will be invaluable in developing new therapeutic tools and strategies.

Book Adrenal Cortex From Physiology to Disease

Download or read book Adrenal Cortex From Physiology to Disease written by Pierre Val and published by Frontiers Media SA. This book was released on 2016-07-18 with total page 91 pages. Available in PDF, EPUB and Kindle. Book excerpt: The adrenal gland plays essential roles in the control of body homeostasis, stress and immune responses. The adrenal cortex represents up to 90% of the gland and is specialised in the production of mineralocorticoids, glucocorticoids and adrenal androgens. This production is tightly coordinated and results from a unique zonal organisation. Although our knowledge of the molecular mechanisms controlling adrenal steroidogenesis is quite extensive, for decades, the mechanisms of adrenal cortex development, cellular homeostasis and renewal have remained elusive. The advent of new high-throughput technologies and sophisticated genetic approaches has brought tremendous progress in our understanding of how the adrenal cortex achieves and maintains its particular organisation. The aim of this Frontiers in Endocrinology Topic is to provide readers with a snapshot of our current knowledge on adrenal physiology and how deregulations of these processes result in adrenal diseases. This includes but is not limited to, basic research on adrenal development, cell lineage identification, progenitor cells, tissue renewal, control of differentiation and zonation and clinical research on the identification of disease-related genes.

The major contributions of modern biochemistry and molecular genetics in our current understanding are visible throughout, providing a unique overview of adrenocortical biology. This book is of great interest to general pediatricians, paediatric endocrinologists, adult endocrinologists, clinical geneticists and basic scientists working in areas such as developmental biology and molecular steroidogenesis.

Book The Origin of a New Progenitor Stem Cell Group in Human Development

Download or read book The Origin of a New Progenitor Stem Cell Group in Human Development written by Hubert Wartenberg and published by Springer. This book was released on 2018-12-12 with total page 90 pages. Available in PDF, EPUB and Kindle. Book excerpt: This book presents an overview of a new group of progenitor stem cells. Their origins were originally detected in light- and electron microscopical sections of plastic embedded human embryonal and fetal organs. The distribution of these cells can result in a large quantity of ectopic stem cells. The observation of these ectopic stem cells leads us to postulate the existence of a basic wave of stem cell progenitors. This process proceeds earlier and independently from germ cell migration. This first wave of progenitor stem cell migration initially reaches the peri-aortal AMG-region (Aortal-Mesonephric-Gonadal region). From here, some of these progenitor cells enter the aorta, are distributed through the vascular system and become the embryonal stem cells (reserve cells) in many peripheral tissues. A second part delivers progenitor cells for the adrenal 'anlage'. Within the peripheral Zona definitiva of the adrenal cortex the progenitor cells multiplicate and subsequently leave the adrenal cortex through a gate of the capsule. The progenitor cells arrive at the pre-aortal sympathic plexus ('second brain'). Within the plexus ganglia they form new organ-like clusters, the paraganglia. In a second process the progenitor cells (Chromaffine cells) enter sympathetic nerve bundles. Here an 'axonal migration' starts, which guides the progenitor cells to several peripheral organs. During this migration process the stem cells develop into their final state. In the adrenal cortex the ingrowing nerves guide the stem cells to the adrenal medulla. In the pancreas the ingrowing nerve fibers guide the stem cells to the islets. These newly detected stem cells represent a specific developmental step which is found only in humans.

Available in PDF, EPUB and Kindle. Book excerpt: Written by experts and specialized investigators, this book presents a detailed overview of the recent progress in our understanding of the adrenal cortex and its pivotal roles in homeostasis. Genes, molecules and cell compartments directly or indirectly involved in the complex steroidogenesis pathway as well as the resulting end-hormones glucocorticoids, mineralocorticoids and androgens are analyzed. Furthermore, the defects of the genes responsible both for common and rare adrenal disorders are presented. The interactions of the adrenal cortices with the adrenal medulla and their importance in the integration of adrenocortical and adrenomedullary function are discussed. The complex molecular pathophysiology of congenital adrenal hyperplasia is presented; long-term effects of the disorder and the still controversial antenatal therapy are examined. Finally, expert review chapters discuss autoimmune Addison disease and the adrenoleukodystrophy/adrenomyeloneuropathy syndrome.Presenting novel research findings in adrenal gland physiology and pathophysiology, this book is a useful tool not only for pediatric endocrinologists, but also for clinicians and researchers studying human development, organogenesis, mitochondrial biology, nuclear receptors, stress biology and pharmacology.
