Book excerpt: The capacity of macrophages and neutrophils to support the reactivity of purified lymphocytes in the mixed leukocyte culture (MLC) reaction was compared: only the macrophage was active. A second study consisted of experiments probing the function of transplantation (H) antigens in the elicitation of the MLC reaction, in the generation of cytotoxic lymphocytes (CL) during MLC, and as target of CL in the cell mediated lympholysis (CML) reaction. Only macrophages and lymphocytes were active in eliciting a MLC whereas neutrophils and platelets which do contain H antigens as well as microsomes and solubilized H antigens obtained from lymphocytes were without effect. However addition of these preparations to third party, unrelated MLC reactions did induce CL specific, to target cells bearing the antigens utilized suggesting that the target of CL generated during MLC is H antigen. This suggestion was further supported by the demonstration that H antigens could specifically inhibit the CML reaction. The results described support the view that substances other than H antigens, present on macrophages and lymphocytes, are responsible for elicitation of MLC. Furthermore they suggest that the target of the CL induced in vitro is H antigen, and that the cell reactive in MLC and in CML may not be one and the same.
