Book Dendritic Cell Control of Immune Responses

Download or read book Dendritic Cell Control of Immune Responses written by Penelope Anne Morel and published by Frontiers Media SA. This book was released on 2016-07-27 with total page 123 pages. Available in PDF, EPUB and Kindle. Book excerpt: Dendritic cells (DC) are among the first cells to encounter pathogens and damage in peripheral tissues and, upon activation, DC migrate to lymph nodes where they activate and educate T cells to initiate and shape the immune response. DC present pathogen-derived antigen to T cells and drive T cell differentiation into particular effector cells through the expression and secretion of co-stimulatory molecules and cytokines respectively. The study of DC biology has included the identification of multiple DC subsets in tissues and lymphoid organs, the differentiation and plasticity of DC subsets, the functional consequences of DC interaction with pathogen, control of DC migratory properties and the impact of DC on T cell activation and differentiation. In recent years sophisticated systems biology approaches have been developed to deepen our understanding of DC function. These studies have identified differences between DC subsets located in various tissues and critical factors that drive the outcome of the interaction between DC and T cells. DC are currently being used in in various clinical therapeutic settings, including as vaccines for cancer and autoimmune disease. A clear understanding of DC factors that contribute to specific immune responses is vital to the success of DC based therapies. This research topic will give a comprehensive overview of current issues in DC biology and provides an update on the clinical uses of DC in the therapy of autoimmunity and cancer.

Book Biophysical control of microfibril orientation in plant cell walls

Download or read book Biophysical control of microfibril orientation in plant cell walls written by J.D. Boyd and published by Springer Science & Business Media. This book was released on 2012-12-06 with total page 210 pages. Available in PDF, EPUB and Kindle. Book excerpt: Within the extreme diversity of aquatic and terrestrial plant genera, each has characteristic cell wall forms. A number of hypotheses have been advanced to explain differences in microfibril arrangements across anyone such wall. Of those, only the 'multinet' theory, which involves the postulation of reorientation of microfibrils caused by cell extension, now has a substantial number of ad herents. However, many scientists are sceptical of its validity; obviously it is incompatible with various observed microfibril arrangements. The tenet of this study is that any such hypothesis can be valid only if it is applicable to all plant forms and wall types. Initially, reanalyses are made of data claimed to confirm justification for multi net postulations. The results show that previous deductions from those data, in support of multinet, are subject to serious challenge. Similarly, a re-examination of the observations, which inspired the multinet theory, shows they have a more logical explanation. Herein, it is concluded that cell wall development involves biophysical factors, which neces sarily prevent multinet's postulated large reorientations of microfibrils, after their formation. Unfortunately the previously most recent published theory, which is based on the absence of reorientation during extension, fails to answer the fundamental question of how alternating orientations between lamellae are controlled, or explain variations in thickness of wall layers. Extensive published data are used to identify forces involved in cell wall development.

Book Physico Chemical Control of Cell Function

Download or read book Physico Chemical Control of Cell Function written by Cesare Gargioli and published by Frontiers Media SA. This book was released on 2019-10-17 with total page 272 pages. Available in PDF, EPUB and Kindle. Book excerpt: Extracellular Matrix (ECM) has been considered for a long time merely a scaffold sustaining cell and tissue function. Despite this simplistic view shared by many, nowadays ECM and their mechanic-physical and chemical characteristic acquired a progressive larger role actively regulating cell life: survival, proliferation, gene expression and differentiation. The interplay between cells and the ECM is continuously controlled at the cell level in a dynamic way. While cells synthesize the raw components of the ECM, this in turn impacts on cell function by providing chemical, topographical and mechanical hints. Such stimuli have been proven to control several aspects of cell function, including survival, proliferation, differentiation and migration. The molecular pathways activated by cells in response to the physical cues arising from the ECM are being disclosed and thus the possibility to control cell function through materials design is becoming more realistic. Current in vitro protocols, relying in 2D cell culture system, entail reductionist approaches to the complexity of cell-ECM interaction and result in cells rapidly losing their distinctive functions in culture. Understanding and replicating the 3D microenvironmental cues affecting cell function appears as a mandatory requirement for the development of next-generation biomaterials, as well as for the establishment of more physiologically relevant and predictive in vitro models of diseases. Such an effort will require a multidisciplinary approach at the convergence of biophysics, biology, nanotechnology, and bioengineering.
