Metals based catalytic amination of alcohols via the so-called “hydrogen borrowing” mechanism is a relative efficient and environmental benign way for synthesis of different primary amines. However, there are still some challenges exist, such as high cost of noble metals, low selectivity of primary amine products, and poor stability and recyclability for industrial applications. To solve these problems, different strategies have been applied in this thesis. Carbon deposition produced by catalyst pre-treatment with alcohols under the optimized conditions has been employed for major enhancement of the selectivity of alcohol amination to primary amines (from 30-50 to 80-90%), which arises from steric hindrance in hydrogenation of bulky secondary imines as intermediate products over partially carbon-decorated cobalt nanoparticles. An efficient approach to protect cobalt catalyst from catalytic deactivation by liquid bismuth promotion with different loading content was disclosed for selectively amination of 1-octanol. The N-alkylation of amines by alcohols over a cheap and efficient heterogeneous catalyst-titanium hydroxide was also proposed. The catalyst with mild Brönsted acidity provides the selectivity higher than 90% to secondary amines for functionalized aromatic and aliphatic alcohols and amines at high catalytic activity and stability.

The two main strategies 'hydrogen' addition vs carbanion addition have had significant impact for the synthesis of [alpha]-chiral amines. Comparison of the literature pertaining to these two different approaches shows them to generally be complementary. From this perspective, the main goal of the described research in this thesis was to develop improved methodology for the preparation of enantiomerically pure [alpha]-chiral amines based on asymmetric reductive amination and carbanion addition methods. Chapter 2 describes a two-step procedure for [alpha]-chiral primary amine synthesis based on reductive amination. The first and key step is the asymmetric reductive amination of a prochiral ketone with a-methylbenzylamine (a-MBA). Depending on the ketone examined, the correct combination of heterogeneous hydrogenation catalyst, solvent, and temperature is crucial for allowing high yield and de with practical reaction times under hydrogenation. Chapter 3 describes the first ytterbium promoted reductive amination reaction which shows significantly enhanced diastereoselectivity as compared to all previous methods. Furthermore, an advantage of this new Lewis acid based reductive amination procedure, verses the traditional Brønsted based methods is the suppression of alcohol by-product formation, is discussed in this chapter. A one-pot asymmetric sequential amination-alkylation (carbanion addition to in situ formed aldimine) method for the synthesis of alkyl-alkyl' [alpha]-chiral primary amines from aldehydes is described in the chapter 4. In situ aldimine formation from aliphatic aldehydes with (R)- or (S)-[alpha]-MBA [catalyzed by 5 mol-% Ti(OiPr)4] followed by reaction with a methyl, ethyl, or n-butyl cuprate complex, in the presence of boron trifluoride, provides good yield and diastereomeric excess.

Abstract: Nowadays, both secondary and tertiary amines are playing vital roles in modern chemical industry as well as pharmaceutical industry. Various synthetic methods of amines have also been reported due to their promising applications. In this thesis, a simple and widely applicable one-pot aza-Wittig/reduction method is reported for synthesizing both secondary and tertiary amines. Firstly, we focused on tandem aza-Wittig/reduction reactions for synthesizing secondary amines. We noticed that an aza-Wittig reaction involving an iminophosphrane which is the product from a Staudinger reaction, followed by addition of an aldehyde can be used to construct a C=N double bond in the product. By making use of the PPh3O (byproduct of aza-Wittig reactions) as the catalyst along with trichlorosilane as the reducing agent, the imine products in the aza-Wittig reactions can be converted into amines in one pot. In addition to using PPh3, highly-efficient heterogeneous polystyrene-based rasta resin-supported PPh3 was also examined in our study. Secondly, in order to broaden the utility of this one-pot method, instead of using relatively dangerous organic azides as substrates, in situ azide formation reaction was added to the flow chart. By reacting alkyl bromides with sodium azide or tertabutylammonium azide can give various azides in one-pot. Thirdly, the secondary amines formed using our one-pot procedure can undergo reductive amination reaction with aldehyde to give diverse tertiary amines as the final products in one pot. In this reaction, trichlorosilane reduction was performed twice by utilizing the phosphine oxide byproduct from the aza-Wittig reaction as a catalyst. Finally, chemistry regarding tandem one-pot aza-Wittig reaction by using catalytic amount of phosphine oxide was discussed. Instead of using organic azides, various aryl isocyanates were used as substrates for the reaction, with the help of catalytic amount of a five-membered ring phosphine oxide to give high yields of secondary amines. Chemistry studied in the thesis stands as solid proof for the superiority of our method. Tandem one-pot reactions improved the efficiency of our method by avoiding lengthy workup and purification steps.
