Book excerpt: The extracellular matrix (ECM) is a complex network of proteins that surrounds cells in all tissues, providing structural and biochemical cues that influence a multitude of cellular functions. In the heart, the ECM provides the local support and context for highly dynamic cardiomyocytes that are continuously contracting to pump blood throughout the body. However, the cardiac phenotype changes drastically throughout fetal development and postnatal maturation. Fetal cardiomyocytes are small, proliferative, glycolytic cells with disorganized sarcomeric structures. In the adult, however, cardiomyocytes become elongated, aligned, post-mitotic cells that rely on oxidative respiration and have highly organized sarcomeres. These ontogenic changes provide a dramatic increase in contractile efficiency, but sacrifice the heart's proliferation-dependent ability to regenerate after injury. As a result, heart disease is a major cause of death worldwide, and has warranted a wealth of research efforts to model cardiac tissue function and disease. During cardiac development and maturation, the ECM also undergoes age-dependent changes that can impact various cardiac functions, but the extent of its influence is poorly understood. To model the age-specific cardiac ECM in vitro, we created cell-derived matrices (CDMs) from cardiac fibroblasts to provide the structural, biophysical and chemical context of cardiac-specific ECM. In doing so, we sought to avoid certain caveats of established CDM protocols, which enhance matrix adhesion and thickness via introduction and promotion of singular matrix proteins, skewing the matrix composition, and confounding comparisons made between CDMs. We, therefore, developed a protocol that enhances matrix adhesion and deposition, respectively, by combining an L-polydopamine coating and macromolecular crowding to produce CDMs composed of cell-produced ECM. This methodology was applied to the study of age-dependent phenotypic and functional changes observed in the heart by comparing the morphologic, metabolic and proliferative responses of cardiomyocytes to CDMs produced by fetal and adult cardiac fibroblasts. Additionally, mass spectrometry proteomics identified the enrichment of ECM proteins in fetal and adult CDMs, enabling loss-of-function studies via genetic silencing of enriched proteins during CDM production. Using this strategy, we identified collagen VI as necessary for maximal oxidative respiration in stem cell-derived cardiomyocytes, which is a key component of cardiomyocyte maturation. Furthermore, we conducted a screen of all age-specific enriched CDM proteins for their influence over DNA synthesis in cardiomyocytes, and found that laminin Îł2, plasmin-mediated signaling in fibroblasts, and fibrosis-associated proteins exerted inhibitory effects on proliferation. Overall, these results demonstrate the development and application of age-specific cardiac CDMs to dissecting the influence of ECM proteins on cardiomyocyte function.

Book excerpt: Myocardial infarction (MI) is the leading cause of death among men and women in the United States. Once a person suffers from a MI, the heart wall will undergo a dynamic and time dependent change, as it goes through the inflammatory phase, proliferative phase, and healing phase. During these phases, the necrotic tissue is removed, and the extracellular matrix (ECM) that holds the cardiomyocytes is altered by an increase in type I collagen, which leads to a scar formation in the infarcted area. The goal of this dissertation is to better understand the role of the cardiac ECM biomechanics in heart physiology, pathophysiology (MI), and regeneration. Three Aims were hence pursued. In Aim 1, we investigated cardiac ECM architecture in intact acellular hearts using diffusion tensor-magnetic resonance imaging (DT-MRI); additionally, we characterized the biomechanical and structural properties of cardiac ECM at different anatomical locations of the left ventricle wall. In Aim 2, we characterized the biomechanical and structural properties of scar ECM during the acute to chronic stages of MI using a rat heart model, in order to better understanding the time course changes in scar ECM biomechanics/microenvironment. In Aim 3, we determined if large mammals (pig heart model) have the capability to fully regenerate a resected piece of the heart apex during the neonatal stage, in which cardiac ECM is still in a developmental phase. The hope is to apply the obtained knowledge in cardiac ECM biomechanics to improve the effectiveness and efficiency of treatments, such as stem cell injection, scar tissue repairing, and regenerative intervention.

Book excerpt: The extracellular matrix (ECM) is a complex composition of proteins, proteoglycans, and glycosaminoglycans. Biological scaffolds composed of ECM have been used to help repair a variety of tissues in the body. It has been shown that an extracellular matrix material can be generated by decellularizing cardiac tissue and processed into injectable formulations for cardiac tissue engineering. When brought to physiological conditions, the matrix material will self-assemble into a porous hydrogel. The objective of the present study is to prepare the gel form of the myocardial matrix, characterize and tailor material properties of the gel, compare material properties to a human derived myocardial matrix gel, and finally evaluate the ability of the gel to support the growth of smooth muscle cells in vitro. Material properties including viscosity, swelling ratio, gelation kinetics, and stiffness were assessed. The results of the study show that material properties of the myocardial matrix can be tailored by adjusting gelation conditions. Changes in final pH, salt concentration, and protein concentration had effects on viscosity and gelation kinetics but not swelling ratio. This study also shows that there are differences in material properties between that of the porcine myocardial matrix and the human myocardial matrix. Finally the study demonstrated that myocardial matrix supported the adhesion and proliferation of rat aortic smooth muscle cells under static conditions.
