Book excerpt: Brucellosis is endemic in free-ranging elk (Cervus elaphus) and bison (Bison bison) in the Greater Yellowstone Area (GYA; USA). It is possible that an oral brucellosis vaccine could be developed and disseminated in the GYA to reduce disease transmission. Should this occur, non-target species other than elk and bison may come in contact with the vaccine resulting in morbidity or mortality. To assess biosafety, bighorn sheep (Ovis canadensis; n=10), pronghorn (Antilocapra americana; n=9), mule deer (Odocoileus hemionus; n=11), moose (Alces alces shirasi; n=10), and coyotes (Canis latrans; n=24) were given a single oral dose of at least 1.0310 10 colony-forming units of Brucella abortus strain RB51 vaccine (RB51). Animals were randomly divided into vaccinated and control groups. Ungulates were captured, blood sampled, and swabs taken from the nares, rectum, and vagina for bacterial culture on day 0, 42, and 84 post-inoculation (PI). On day 42, the vaccinated group became a control group and vice versa in a crossover design. Blood and swab samples were taken from coyotes on days 0, 14, 28, and 42 PI. There was no crossover for the coyote study. Two coyotes from each group were also euthanized and cultured for RB51 on days 42, 84, 168, and 336 PI. Blood samples were analyzed for hematologic changes and antibodies to RB51 using a modi?ed dot-blot assay. No morbidity or mortality as a result of vaccination was observed in any animal. There were no differences in hematologic parameters at any time for ungulate species; vaccinated coyotes had higher hematocrit, hemoglobin, and eosinophil counts (P[less than or equal to]0.006). All individuals, except some moose, seroconverted to RB51. Strain RB51 was cultured from oropharyngeal lymph nodes from one coyote 42 days PI and from a moose 117 days PI. This study suggested that a single oral dose of RB51 was safe in these species.

Global prevalence of human brucellosis remains significant. More than half a million new brucellosis cases from 100 countries are reported annually to the World Health Organization (WHO). The majority of these cases are reported in developing countries.In humans, brucellosis (undulant fever, Malta fever) is characterized by an acute bacteremic phase followed by a chronic stage that may extend over many years and may involve many tissues. It is a systemic disease, and many organ systems (nervous system, heart, skeletal system, bone marrow, etc.) may become involved following hematogenous dissemination. Although eradicated in some countries, it remains one of the most economically important zoonosis worldwide as it is responsible for huge economic losses as well as significant human morbidity in endemic areas. Because of the nonspecific clinical manifestations of human brucellosis and the need for prolonged combination therapy with antibiotics that are not routinely prescribed for other infectious diseases, laboratory confirmation of the diagnosis is of paramount importance for adequate patient management. In addition, evidence of brucellosis has serious public health implications because it discloses exposure to a contaminated source (infected animals or their products, unsafe laboratory practices, or a potential biological warfare attack). This book addresses human brucellosis with stress on symptoms including those related to the less recognized disease localizations, risk of exposure, treatment, and prevention. Light is shed on animal brucellosis as it pertains to human exposure. The book also emphasizes on laboratory procedures in culturing and serologic techniques. Epidemiologic surveillance is among this books subjects as well as veterinary control measures.

Commercially available anthrax vaccines for immunization of humans are known to provide protection of limited duration and may not protect against the respiratory form of the disease. Commercially available live vaccines for animals have been shown to cause disease in certain species. Brucella abortus is a facultative intracellular bacterium that causes chronic infection in animals and humans. As with other intracellular pathogens, cell mediated immune responses (CMI) are crucial in affording protection against brucellosis. B. abortus strain RB51 has been shown to be useful in eliciting protective CMI and antibody responses against Brucella in cattle and other animal species. Since the protective antigen (PA) of B. anthracis is known to induce antibodies, the pag gene encoding PA was expressed in B. abortus RB51, producing a dual vaccine to protect against both brucellosis and anthrax. In a previous study, the entire pag gene was expressed in strain RB51 and following immunization the vaccine induced antibodies against PA in A/J mice. However, PA stability and protective efficacy were less than desirable as only 1/6 were protected.
