Much of this progress has been driven by the development of new techniques for studying membrane components and their interactions. Tradition ally, the investigation of membranes has largely occurred within the domains of biochemistry, physical chemistry, and cell biology; but many of the most significant advances have resulted from an expansion into other disciplines, such as molecular biology, immunology, and the clinical sciences. In these two volumes we have attempted to bring together some of these techniques—to combine the old and the new. Biomembrane Protocols: II. Architecture and Function is concerned exclusively with the architecture and activities of membranes, whereas the companion volume, Biomembrane Protocols: I. Isolation and Analysis, deals with their isolation and compositional analysis. The aim of each chapter is to provide detailed technical and meth odological information that will allow the reader to perform the tech nique successfully, without the need to consult other texts. Detailed reviews of membrane structure and function are not included, except where they are relevant to the choice or efficacy of a particular proce dure. In addition to Materials and Methods sections, each chapter has a Notes section that explains the reasons for taking certain steps and pro vides practical tips for applying the techniques to other systems.

The functions of membrane proteins include cell/cell and cell/extracellular matrix recognition, the reception and transduction of extracellular signals, and the tra- port of proteins, solutes and water molecules. Abnormal membrane protein expression has profound biological effects and may, for example, underlie phenotypic and functional differences between normal and tumour cells. Moreover the accessibility, particularly of plasma proteins traversing the plasma membrane of cells, makes them of particular ut- ity to the therapeutic intervention in disease. Indeed, it is estimated that of all currently licensed pharmaceuticals, approximately 70% target proteins resident in the plasma m- brane. In theory, unbiased technologies such as proteomics have the power to de?ne patterns of membrane protein expression characteristic of distinct states of cellular development, differentiation or disease, and thereby identify novel markers of, or targets for intervention in, disease. However, although about 25% of open reading frames in fully sequenced genomes are estimated to encode integral membrane proteins, global analysis of membrane protein expression has proved problematic. Membrane protein analysis poses unique challenges at the level of extraction, solubilization, and separation in particular, and to a lesser extent of identi?cation and quantitation. These challenges have, however, fostered creativity, in- vation, and technical advances, many of which are brought together in Membrane P- teomics.

Book excerpt: Efforts to describe and model the molecular structure of biological membranes go back to the beginning of the last century. In 1917, Langmuir described membranes as a layer of lipids one molecule thick [1]. Eight years later, Gorter and Grendel concluded from their studies that “the phospholipid molecules that formed the cell membrane were arranged in two layers to form a lipid bilayer” [2]. Danielli and Robertson proposed, in 1935, a model in which the bilayer of lipids is sequestered between two monolayers of unfolded proteins [3], and the currently still accepted fuid mosaic model was proposed by Singer and Nicolson in 1972 [4]. Among those landmarks of biomembrane history, a serendipitous observation made by Alex Bangham during the early 1960s deserves undoubtedly a special place. His fnding that exposure of dry phospholipids to an excess of water gives rise to lamellar structures [5] has opened versatile experimental access to studying the biophysics and biochemistry of biological phospholipid membranes. Although during the following 4 decades biological membrane models have grown in complexity and functionality [6], liposomes are, besides supported bilayers, membrane nanodiscs, and hybrid membranes, still an indisputably important tool for membrane b- physicists and biochemists. In vol. II of this book, the reader will fnd detailed methods for the use of liposomes in studying a variety of biochemical and biophysical membrane phenomena concomitant with chapters describing a great palette of state-of-the-art analytical technologies.
