Book excerpt: Robotics has markedly extended the reach of human beings in sensing, manipulating, and transforming the world around us. One of the most inspiring challenges in science and technology is to extend our capacity with tiny robots towards operations at the micro/nanoscales, a dimension where we can directly interact with fundamental biological building blocks. This thesis is devoted to advancing micro/nanorobotics to extend human being's capacities in following three themes, with a special focus being given to biomedical applications. The first theme focuses on design and fabrication of bio-inspired un-tethered nanorobots with efficient locomotion, adaptive operation, collective regulation, and eventually biological function towards operation in whole blood. The second theme explores the versatility of functional micro/nanorobots to perform diverse tasks including writing (nanolithography), reading (superresolution imaging), destroying (warfare agents), and repairing (surface cracks), all at the micro/nanoscale. The third theme employs self-propelling microrobot as an active delivery technique that autonomously and precisely transports the therapeutic agents inside live animal's gastrointestinal tract, improving therapeutic efficacy for bacterial infection treatment. This technique opens the door for micro/nanorobots as an active delivery platform for medical treatment and is promising for a wide range of personalized diagnostic and therapeutic applications. Ultimately, micro/nanorobots has the potential to change the game of science, engineering, and medicine by extending our capacity at the micro/nanoscale. The novel applications presented in this thesis are just a few examples showing the power of micro/nanorobots, with countless more avenues waiting to be explored towards living and active matter, and eventually artificial intelligence and synthetic life at the micro/nanoscale.

This dissertation aims to demonstrate unique advantages of micro/nanorobot platforms. The first theme focuses on developing the use of ultrasound propelled nanostructures for diverse applications, including "on-the-move" capture and the transport of multiple cargoes and internalization and movement inside live MCF-7 cancer cells. The second theme explores the use of acoustically triggered microcannons. This principle was tested towards mechanochemical blood clot degradation and enhancing drug permeability through the epidermis. The third theme describes the use of biohybrid robotics systems for detoxification and decontamination of environmental pollutants, including bacteria (E. coli), nerve agent (methyl paraoxon) and heavy metal ions (Cd and Pb) from aqueous solutions. In the not-so-distant future, Micro/nanorobots could serve as robust and versatile platform for potentially improving medical diagnosis and treatment.

In the contemporary era of advancing nanomedicine, the utilization of micro/nanorobots in clinical therapy is still in its nascent stages within the realm of modern healthcare. Biomedical and biological environments hold immense promise as platforms for these active agents, showcasing remarkable functionalities and efficacy in vitro, ex vivo, and in vivo. Micro/nanorobots have the capacity to emulate the behaviors of living cells, particularly bacteria, which play a crucial role in microbial infections, thus impacting public health and medical devices. These active agents possess the potential to overcome biological barriers and enable targeted therapies for various healthcare issues, including the prevention and treatment of diseased tissues and biofilms, which will significantly enhance the minimally invasive operations and remote treatments for the next-generation human healthcare system. The objectives of this research topic are threefold: (1) to investigate the novel functionalities of micro/nanorobots in biological contexts, (2) to unravel the underlying principles of cell, tissue, and organ development, and (3) to innovate active therapeutic approaches for addressing diseased tissues and microbial biofilms
