The book will conclude with a discussion of modeling approaches to biologic cycles and chronotherapy, for future research and translation.

They are driven by a group of genes called clock genes that have been widely observed in plants, animals and even in bacteria. In mammals, the core clock genes are rhythmically expressed in both the suprachiasmatic nucleus (SCN), the master clock residing in the hypothalamus, and almost all peripheral tissues where they control numerous target genes in a circadian manner, and thus affect many physiological and biochemical processes. Evidence suggests that disruption of the circadian rhythms (or desynchronization) is a significant risk factor for the development of metabolic diseases, cardiovascular diseases, cancer and sleep disorders. Evidence also suggests that the disruption suppresses immune function and increases vulnerability to infectious diseases. Restoring or strengthening the circadian rhythm may be therapeutic for these conditions. This becomes exceptionally important in modern societies because many people are suffering from frequent desynchronization due to shift working, exposure to artificial light, travel by transmeridian air flight, and involvement in social activities. Besides, the temporal variations in the incidence and severity of many diseases, such as the onset of cardiovascular events, chronic obstructive pulmonary disease (COPD), inflammatory diseases and mental disorders have also drawn increasing attention to the circadian clock. The circadian rhythms affect not only the health status, but also the drug efficiency. The effects (and side effects) of many drugs vary with biological timing. The tolerance of many medications displays circadian variation as well. The timing of medical treatment in coordination with the body clock may significantly increase the desired effects of drugs, and lower the dose and toxicity. In addition, circadian rhythms can also be modulated by some therapeutic drugs, for example, melatonin and modafinil, which are used to treat circadian rhythm sleep disorders. In this Research Topic, we assemble a series of critical review and research articles that focus on the therapeutic implications of circadian rhythms. Although most c- tributions in this volume focus on mammalian circadian clocks, the historical int- duction and comparative clocks section illustrate the importance of various other organisms in deciphering the mechanisms and principles of circadian biology. Circadian rhythms have been studied for centuries, but only recently, a mole- lar understanding of this process has emerged. This has taken research on circadian clocks from mystic phenomenology to a mechanistic level; chains of molecular events can describe phenomena with remarkable accuracy. Nevertheless, current models of the functioning of circadian clocks are still rudimentary. This is not due to the faultiness of discovered mechanisms, but due to the lack of undiscovered processes involved in contributing to circadian rhythmicity. We know for example, that the general circadian mechanism is not regulated equally in all tissues of m- mals. Hence, a lot still needs to be discovered to get a full understanding of cir- dian rhythms at the systems level. In this respect, technology has advanced at high speed in the last years and provided us with data illustrating the sheer complexity of regulation of physiological processes in organisms. To handle this information, computer aided integration of the results is of utmost importance in order to d- cover novel concepts that ultimately need to be tested experimentally.

Circadian rhythmicity is a fundamental endogenous process of almost every organism living on Earth. For instance, the alternation of hunger and satiety is not continuous over 24 h, but is instead structured in time along the light/dark cycle. In mammals, the temporal organization of metabolism, physiology and behavior around 24 h is controlled by a network of multiple cellular clocks, synchronized via neuronal and hormonal signals by a master clock located in the suprachiasmatic nuclei of the hypothalamus. This central circadian conductor in the brain is mainly reset by ambient light perceived by the retina, while secondary circadian clocks in other brain areas and peripheral organs can be reset by meal timing. Chronic disruption of circadian rhythms, as seen in human shift-workers (up to 20% of the active population), has been associated with the development of a number of adverse mental and metabolic conditions. Understanding of the functional links between circadian desynchronization and overall health in animal models and humans, however, is still scarce. Interactions between circadian clocks and metabolism can occur at different levels: the molecular clockwork, internal synchronization via neuro-hormonal signals, or external synchronization via photic or feeding cues. Book excerpt: This dissertation lays the groundwork for Circadian Computing with a novel and broad vision of technologies that support and adapt to our innate biological rhythms. Similar to most terrestrial organisms, human physiology and behavior are shaped by a 24-hour periodicity known as circadian rhythm. Indeed, almost every neurobehavioral process including our sleep, metabolism, cognitive performance, and mood reflects circadian rhythms. These rhythms ensure synchronization across different processes and as such, are crucial for our health and well-being. Persistent circadian disruption increases risk for cancer, obesity, and cardiovascular diseases. It has been associated with occupational accidents and serious loss of productivity in the workplace as well. Recent findings have also started identifying links between circadian disruption and mental illnesses including bipolar disorder and schizophrenia. However, in our modern world, circadian disruption is becoming increasingly widespread. The invention of artificial light fundamentally changed our ancestral sleep and wakeup patterns. Since then, we have gradually moved towards a 24-hour society. The recent development in entertainment and communication technologies has also resulted in an "always-on" ethos. The resulting trend is worrisome. Sleep pathologies are reaching an epidemic level with 70\% of the population suffering from significant circadian disruptions. As a result, recently there has been an increased focus on monitoring and identifying disruptions in circadian rhythms. However, these methods and findings are often limited to controlled lab environments. As a result, they are not adequate for granular monitoring of circadian disruptions in the wild over a longitudinal period of time. As such, there is a need for novel pervasive technologies for tracking, monitoring, and modeling circadian disruptions and its impact in the real world. There is also an opportunity for developing intervention tools for maintaining circadian stability. This dissertation is a leading step towards the broad vision of circadian-aware technologies for sensing, adapting to, and stabilizing our innate biological rhythms. In my PhD work, I have shown the feasibility of bringing a circadian-aware perspective across different application domains. Specifically, I have developed and evaluated methods for unobtrusively assessing circadian disruptions. I have also showed that behavioral and contextual data can be used for modeling and predicting alertness — a circadian process integral to our cognitive performance. I have also developed, deployed, and evaluated a data driven tool focusing on identifying circadian anomalies in patients with bipolar disorder. With this groundwork in place, I believe that there is an exciting opportunity lying ahead for Circadian Computing. In particular, circadian-aware technologies can potentially reshape a number of application domains including education and learning, optimized scheduling, mental health care, and chronotherapy. I hope this dissertation motivates a circadian perspective in future technology development and contributes to the shared effort of improving our productivity, health, and well-being.
