Book excerpt: One of the most significant challenges facing mankind in the twenty-first century is the development of a sustainable global economy. Within the scientific community, this calls for the development of processes and technologies that will allow the sustainable production of materials from renewable natural resources. Plant material, in particular lignin, is one such resource. During the annual production of about 100 million metric tons of chemical wood pulps worldwide, approximately 45 and 2 million metric tons/year of kraft lignin and lignosulfonates, respectively, are also generated. Although lignosulfonates have found many applications outside the pulp and paper industry, the majority of kraft lignin is being used internally as a low-grade fuel for the kraft pulping operation. A surplus of kraft lignin will become available as kraft mills increase their pulp production without expanding the capacity of their recovery boilers that utilize lignin as a fuel. There is a tremendous opportunity and an enormous economic incentive to find better uses of kraft lignin, lignosulfonates and other industriallignins. The pulp and paper industry not only produces an enormous amount of lignins as by products of chemical wood pulps, but it also utilizes about 10 million metric tons of lignin per year as a component of mechanical wood pulps and papers. Mechanical wood pulps, produced in a yield of 90-98% with the retention of lignin, are mainly used to make low-quality, non-permanent papers such as newsprint and telephone directories because of the light-induced photooxidation of lignin and the yellowing of the papers.

In recent years, the widely used indocyanine green (ICG) has made the surgical maneuver of inner limiting membrane peeling tremendously safer and efficient. Also, numerous dyes have been applied in experimental settings with promising or devastating results. This volume highlights the major clinical and experimental results of currently used novel vital dyes. The first chapters describe the transparent structure of the vitreous body and summarize historical considerations to visualize its structure by optical coherence tomography, dye injections or autologous cells during surgery and for diagnostic purposes. The following contributions describe the advantages and disadvantages of ICG during vitreoretinal surgery and experimental applications. Alternative approaches by recently approved vital dyes such as trypan blue, patent blue and brilliant blue are evaluated in the subsequent chapters. Finally the last few chapters give an outlook on novel vital dyes, which are currently under evaluation, as well as alternative enzymatic approaches to remove the vitreous from the retinal surface. Available in PDF, EPUB and Kindle. Book excerpt: The human genome and other large-scale genome sequencing projects have inevitably led to a focus on the proteins encoded by genes. The field of proteomics has grown enormously as a result and a number of high-throughput technologies have now been developed allowing discovery-led investigations of protein populations rather than more traditional hypothesis-led studies on single proteins. These high-throughput techno- gies include gene and protein microarrays, the yeast two-hybrid system, and various mass spectrometry methodologies. However, despite developments and improvements in these technologies, two-dimensional electrophoresis (2DE) remains one of the most widely used approaches. This technique was revolutionised by the development of immobilised pH gradient strips which are now commercially available. This has made possible highly reproducible separations of matched samples. Developments in staining, mass spectr- etry, and bioinformatics supported these developments and have led to a measure of standardisation in design, execution, and analysis of proteomics experiments. This book began life as a proposed update of the excellent volume 2DE Protocols edited by Andrew Link of the University of Washington at Seattle. However, we re- ised that 2DE has undergone major development in aspects of its technology in recent years and we were anxious to reflect these in the present volume. We are also conscious that many researchers have now begun to apply proteomics methodologies to a growing range of biological material and we were anxious to include contributions to reflect the challenges posed in sample preparation in less widely used organisms.
